Participants reportedly distinguished several deuterated musk odorants from their ordinary forms, even though replacing hydrogen with deuterium should leave the molecules’ overall ground-state shape nearly unchanged. The result was compatible with the idea that receptors might respond partly to molecular vibrations. It did not demonstrate the theory’s proposed receptor mechanism—vibrationally assisted electron tunnelling—and later receptor experiments reported no difference for the isotope pairs they tested.
What the “boost” was—and what it was not
The headline refers to a study by Gane and colleagues published in PLOS ONE on 25 January 2013. The researchers tested whether people could tell an odorant containing ordinary hydrogen from an isotopomer in which some hydrogen atoms had been replaced by deuterium, a heavier isotope of hydrogen.

The central result was selective. In the tests reported, participants did not distinguish acetophenone from its deuterated counterpart. They did distinguish several deuterated musk odorants from the corresponding undeuterated compounds. The authors argued that this pattern was “consistent with a vibrational component in human olfaction.” That wording matters: it describes compatibility with a hypothesis, not direct detection of vibrations by a known receptor.

Chemistry World’s contemporary report described the finding as a boost for Turin’s theory. Turin’s proposal says that an odorant’s molecular vibrations could enable electron transfer through an olfactory receptor, helping the receptor identify molecules in addition to recognizing their shape. The experiment did not observe electron transfer, identify a receptor, or measure a vibrational signal in a living nose.

How the isotope experiment was supposed to test smell theories
Hydrogen versus deuterium
Deuterium has the same number of protons as hydrogen but a neutron in its nucleus, making it roughly twice as heavy. Substituting deuterium for hydrogen changes the frequencies at which chemical bonds vibrate. In the study’s framing, the substitution was useful because it could alter vibrational modes without substantially changing the molecule’s ground-state conformation—the three-dimensional arrangement that a shape-based receptor would be expected to recognize.
This creates a conceptual test between two explanations. If smell depended only on a molecule fitting a receptor’s binding site, an isotope swap that preserved the relevant shape might make little or no perceptible difference. If vibrational information also mattered, changing the bond vibrations could, in principle, change the odor even when the overall structure remained almost the same.
What people were asked to distinguish
The researchers compared normal and deuterated odorants in human psychophysical tests. Their reported outcomes were not a blanket demonstration that people can smell every isotope substitution. Acetophenone isotopomers were not distinguished in the reported tests, while several deuterated musk compounds were.

That selectivity is scientifically important. It means the result depends on the chemical pair, its concentration and preparation, and the details of the test. A positive result with some musks cannot be expanded into a claim that human smell generally reads molecular mass or that all deuterated compounds acquire a different odor.

The two competing explanations for odor recognition
| Feature | Receptor-ligand docking model | Vibration theory |
|---|---|---|
| Primary information | The molecule’s shape, chemical groups and interactions allow it to fit a receptor. | The molecule’s vibrational modes provide additional identifying information. |
| Proposed physical event | Binding changes receptor behavior, activating olfactory neurons. | Vibrationally assisted electron transfer or tunnelling is proposed at the receptor. |
| Prediction for a carefully matched isotope pair | If shape and interactions remain functionally equivalent, receptor responses may be similar. | A changed vibrational spectrum could alter receptor activation or perceived odor. |
| What the 2013 human result addressed | It supplied a behavioral observation that could be difficult to explain with shape alone for the tested musks. | It was interpreted as compatible with a vibrational contribution, but did not measure the proposed electron-transfer step. |
“Docking” in this context does not mean a single rigid lock-and-key interaction. Olfactory receptors can respond to overlapping sets of molecules, and odor identity is represented across populations of receptors and neurons. The docking model is therefore a broad molecular-recognition framework rather than a claim that one receptor corresponds to one smell.
The vibration theory is more specific about the extra information it proposes. In Turin’s account, the receptor would act somewhat like a molecular sensor that allows an electron to move only when the odorant supplies an appropriate vibrational energy. Demonstrating that idea requires evidence at the receptor or molecular level, not just a report that two samples smelled different.
Why a behavioral difference cannot prove the receptor mechanism
A psychophysical discrimination result answers a limited question: can people, under the test conditions, detect a difference between two samples? It does not identify the biological step that produced the difference. Human participants can be influenced by concentration, impurities, sample handling, expectation, adaptation and metabolism before an odorant reaches a receptor.

The way compounds behaved during preparation could have produced differences unrelated to the intended isotope substitution. Tiny impurities might have contributed an odor signal. Nasal enzymes might also metabolize the normal and deuterated compounds differently, creating distinct products that receptors detect. These possibilities were reported criticisms and hypotheses, not established explanations for the musk result.

Purification is especially consequential in an isotope experiment. If an undeuterated sample contains a trace contaminant, or if synthesis leaves a mixture of isotope patterns, a participant may be responding to a minor chemical component. A convincing design therefore needs rigorous chemical characterization, matched handling, blind presentation and controls that show the effect tracks isotope substitution rather than preparation history.
The study’s negative acetophenone result also limits interpretation. If vibrational sensing were a universal and dominant feature of olfaction, one might expect every suitable isotope substitution to produce a difference. The reported pattern instead suggests that any effect, if real, could depend on the odorant, receptor, isotope placement or test conditions.
What later receptor experiments found
The 2015 PNAS study
In 2015, Block and colleagues examined responses from tested human and mouse olfactory receptors to normal and isotopically labelled odorants. For the receptor–ligand pairs they studied, the receptors responded similarly to the ordinary and labelled compounds. The authors concluded that these results did not support vibration theory for those tested systems.
This is important counterevidence because it moves the question closer to the proposed mechanism. A receptor assay can compare activation while controlling exposure more directly than a whole-person smell test. However, the result is bounded: it does not prove that every olfactory receptor, every isotope placement or every odorant has been tested. “No difference” in the examined pairs is not the same as proving that a vibrational contribution is impossible in all biology.

What the 2018 reviews said
A 2018 review by Block, drawing on olfactory-receptor deorphaning and olfactory-receptor-neuron activation studies, said that a receptor-ligand docking model was favored over a bond-vibration model by the cumulative evidence it assessed. That is a review-level judgment tied to its evidence base, not a timeless declaration that the debate has ended.
A separate 2018 review in Frontiers in Physics surveyed arguments and evidence surrounding vibration theory. Its existence shows that the proposal remained an active subject of discussion, but a review published in 2018 cannot by itself establish the state of research in 2026. The sources available for this article do not support a claim about a definitive post-2018 consensus.
How to weigh the evidence
| Evidence level | What it can show | What it cannot show by itself |
|---|---|---|
| Human psychophysics (2013) | Whether participants reported or demonstrated discrimination between specified odorant samples. | Which receptor, metabolic product or physical mechanism caused the discrimination. |
| Receptor assays (2015) | Whether selected human or mouse receptors changed their responses differently for selected isotope pairs. | That all receptors and odorants behave the same way, or that an untested pair cannot reveal an isotope effect. |
| Evidence reviews (2018) | How an author evaluates results across receptor identification and neuron-activation studies. | A guarantee that later experiments will reach the same conclusion or that every competing model has been eliminated. |
The levels are complementary rather than interchangeable. A person’s ability to discriminate is the starting observation. Receptor experiments test whether a proposed molecular sensor responds differently. Reviews then weigh patterns across many experiments. Treating the first level as if it were the third is the central mistake in reading the “boost” headline.
What the 2013 result changed in the debate
The study made isotope substitution look like a potentially informative probe of olfaction. Because deuteration can change vibrational frequencies while preserving much of a molecule’s gross structure, a reproducible odor difference would challenge a simple version of shape-only recognition. It gave vibration theory a concrete, testable prediction rather than leaving it as a purely theoretical alternative.

It also forced researchers to sharpen controls. Future work has to establish the isotopic composition and purity of each sample, verify that concentrations are matched, rule out decomposition and metabolic conversion, and test whether isolated receptors reproduce the human effect. Testing both positive and negative examples—such as the musk compounds and acetophenone isotopomers reported in 2013—is necessary to determine whether any effect is general or chemically specific.
Turin called the implication potentially very broad, saying it “raises the awe-inspiring possibility that half of physiology and pharmacology may be vibration-based.” That is a statement about the theory’s reach, not evidence that half of physiology and pharmacology has been shown to operate that way. Other scientists were more cautious. Theoretical biophysicist Ilia Solov’yov described the protocols as carefully done but said he was not convinced the results proved unequivocally that humans distinguish deuterated odorants. Olfaction researcher Leslie B. Vosshall argued that attempts to prove the theory should concentrate on mechanisms at the receptor rather than indirect psychophysical testing.
What readers should conclude today
- The 2013 paper reported discrimination of several deuterated musk odorants, while its acetophenone tests did not show discrimination.
- The authors’ phrase “consistent with a vibrational component” is an interpretation of a behavioral pattern, not a demonstration of vibrationally assisted electron tunnelling.
- Preparation artifacts, trace impurities and different nasal metabolism were identified as possible alternative explanations, but none was established as the cause.
- 2015 receptor experiments found similar responses for the tested normal and labelled odorants, providing bounded counterevidence to the proposed mechanism.
- A 2018 evidence review favored receptor-ligand docking on the studies it assessed; the available sources do not justify claiming a final 2026 verdict.
The fairest reading is therefore conditional. The deuterated-musk finding was interesting because it supplied a human result that a vibration-based account could accommodate and that invited more precise tests. It did not establish that people sense molecular vibrations, and subsequent receptor-level evidence made the mechanism more difficult to sustain. The scientific question remains one of matching a perceptual effect to a demonstrated molecular event.
FAQ
Did people smell deuterium itself?
No. The experiment compared odorant molecules that differed by replacing selected hydrogen atoms with deuterium. Any perceived difference would be a property of the whole isotopomer and its interactions, not a smell produced by free deuterium as a separate substance.
Does the study show that all isotopically labelled smells are different?
No. The reported tests found no discrimination for acetophenone isotopomers and discrimination for several musk isotopomers. The result was compound-specific in the experiments described.
Did the 2015 work disprove vibration theory?
It challenged the theory for the human and mouse receptors and isotope pairs that were tested by reporting similar receptor responses. It did not examine every possible receptor, odorant or isotope placement, so it is bounded counterevidence rather than universal disproof.
Which model is currently better supported by the cited evidence?
Block’s 2018 review said the cumulative receptor and olfactory-neuron evidence it assessed favored receptor-ligand docking over a bond-vibration model. That conclusion should be attributed to that review and not extended into an unsupported claim about all research after 2018.
