Scientists discover a plastic-eating “Pac-Man” enzyme with a surprising secret

Researchers at the University of Konstanz have identified a bacterial family-VIII esterase that can break down several biodegradable polyester plastics. The enzyme earned the nickname “Pac-Man” because its unusually open active site may let it attack polymer chains directly. Its more unexpected feature is that it also hydrolyzes beta-lactam antibiotics such as penicillin.

The finding, published in The ISME Journal in 2026 (DOI 10.1093/ismejo/wrag203), is evidence for a specialized plastic-degrading mechanism—not a discovery that bacteria can make every kind of plastic disappear.

What the Pac-Man enzyme actually does

The enzyme targets polyester chemistry containing hydrolysable ester bonds. In the Konstanz experiments, the relevant materials were LCAP, PHBV and PCL bioplastics. These polymers are chemically different from conventional polyethylene products such as HDPE, whose carbon-carbon backbone is far harder for environmental enzymes to attack.

“Pac-Man” describes the enzyme’s structure: modelling showed an unusually wide-open active site. That shape is consistent with accommodating portions of a polymer chain rather than only small dissolved molecules. The enzyme is a bacterial family-VIII esterase, a group of proteins that normally cleave ester bonds.

Material Reported result Conditions and qualification
LCAP bioplastic Completely degraded in roughly 250–330 days University of Konstanz forest-humus burial and laboratory microcosm work in 2026
PHBV bioplastic Completely degraded in roughly 250–330 days Same reported soil and laboratory conditions
PCL bioplastic Completely degraded in roughly 250–330 days Same reported soil and laboratory conditions
Cellulose Broke down after about 80 days University of Konstanz comparison in the reported experiment
HDPE Virtually no degradation Laboratory experiment; this result does not establish that HDPE disappears in nature

The times are environmental-experiment measurements, not an industrial processing rate. They also describe complete disappearance of the tested materials under the reported conditions, not necessarily recovery of purified, reusable monomers.

How scientists found it in forest soil

Harry Lerner, David Schleheck and colleagues combined field-like burial, laboratory chemistry and DNA sequencing.

  1. Buried films: Small LCAP bioplastic films were placed about 10 centimetres deep in forest humus at the university botanical garden.
  2. Built microcosms: Powdered plastics were mixed into soil in laboratory containers, where the team monitored carbon-dioxide production for a year as a sign of microbial activity.
  3. Recovered the films: Electron microscopy examined what remained after exposure to the soil community.
  4. Sequenced the community: Metagenomic analysis compared DNA from soils containing LCAP with the surrounding microbial community and identified an esterase gene enriched in the plastic-containing soil.
  5. Modelled the protein: Structural analysis revealed the open active site, a secretion signal and a membrane-bound lipid anchor.

The cell-sized holes revealed a different way to digest plastic

Electron microscopy found tiny holes in the recovered films. Their size and shape matched individual bacterial cells. That pattern led the researchers to propose that plastic-depolymerising enzymes are anchored on bacterial surfaces rather than released only as free-floating proteins.

With a membrane anchor, a bacterium could hold the esterase against the film, cut into the polymer and remain embedded in the material. The resulting holes would be the physical trace of cells digesting their way inward. This challenges the common assumption that microbes must secrete plastic-degrading enzymes into the surrounding soil before they can attack a solid surface.

The gene’s secretion signal and lipid anchor make that explanation biologically plausible, but the study is still a mechanistic demonstration. It does not show that every plastic-degrading microbe uses cell-surface enzymes or that the holes alone prove the complete pathway from polymer to reusable chemicals.

Why penicillin resistance is part of the story

In laboratory tests, the same enzyme also cleaved penicillin. Its structure resembles both an esterase and a beta-lactamase, the enzyme family that can deactivate beta-lactam antibiotics by opening their characteristic ring.

That dual activity could provide two advantages to a bacterium living on plastic. Surface-bound esterase activity could release small plastic-derived molecules close to the cell for rapid uptake, while beta-lactamase activity could inactivate an antibiotic outside the cell. The authors also caution that the penicillin-cleaving function may simply be an evolutionary remnant rather than an adaptation selected by plastic.

This is relevant to antibiotic-resistance ecology, but it is not proof that the discovery has created a new resistant pathogen. The experiments show enzyme activity against penicillin in the laboratory; they do not establish transmission of a resistance gene, a clinical risk, or resistance levels in a particular environment.

Can it eat ordinary plastic?

No. The reported results apply to biodegradable polyesters with ester bonds. LCAP, PHBV and PCL degraded in the Konstanz tests, whereas HDPE showed virtually no degradation in the laboratory experiment. A plastic bag, bottle cap or food container made from conventional polyethylene should not be assumed to respond to this enzyme.

The distinction is chemical: polyester chains contain bonds that water and esterases can hydrolyse, while HDPE is built from a much less reactive hydrocarbon backbone. The nickname therefore should not be read as a claim of universal plastic-eating ability.

Could the enzyme be used for recycling?

Possibly, but the study does not demonstrate an industrial recycling process. No reactor design, operating cost, yield of purified monomers or commercial deployment was reported.

For enzymatic recycling, researchers would need to show that the enzyme can work at useful concentrations, temperatures and material thicknesses; that it converts polymer into defined monomers rather than a difficult mixture; and that those products can be separated and reused. A cell-surface mechanism could be useful in a controlled biological reactor, but it could also complicate production because the active protein is tied to a living cell or membrane system.

The Konstanz work instead points to a design principle: plastics intended for biological or enzymatic end-of-life treatment should contain predictable biochemical breaking points. The authors argue that polymers with hydrolysable ester bonds are more realistic targets than chemically inert polyethylene.

What this discovery changes—and what it does not

  • It provides a plausible cell-surface model for how soil bacteria penetrate polyester films.
  • It links polyester depolymerisation with beta-lactamase-like antibiotic activity in one enzyme.
  • It shows that the tested polyester bioplastics can be completely degraded in roughly 250–330 days under the reported soil and laboratory conditions.
  • It does not show that conventional polyethylene is biodegradable, that plastic pollution can be solved by releasing bacteria, or that an industrial recycling method is ready.

David Schleheck describes the “plastisphere” as a relatively new habitat: humans have introduced large quantities of plastic for only about 50 to 75 years, giving microbial communities a recent additional source of carbon and energy. The practical lesson is narrower than the headline but important: microbial evolution may uncover faster ways to process certain designed polymers, while the chemistry of the material still determines what is possible.

How it differs from other plastic-degrading research

Other studies have reported PET-degrading enzymes in wastewater microbes, including work on Comamonas testosteroni. Those systems concern a different polymer, enzyme and environment. The Pac-Man enzyme study is specifically about LCAP, PHBV and PCL in forest soil and laboratory microcosms, with an apparent cell-surface anchor and measurable penicillin-cleaving activity. Those distinctions matter when judging claims about “plastic-eating” bacteria.
