Researchers have captured two detailed structural snapshots of the trypanosomatid trans-spliceosome, a molecular machine parasites use to process messenger RNA. The 2026 study shows how this complex prepares and then joins a short RNA leader to parasite pre-mRNA. The finding explains an essential part of parasite RNA biology; it does not report a drug or treatment.
What is the parasite’s hidden molecular machine?
The machine is the trans-spliceosome. Trypanosomatid parasites produce many protein-coding genes in long precursor RNA transcripts. To make mature messenger RNAs, they attach a short, capped sequence called the spliced leader (SL) exon to the 5′ end of each pre-mRNA.

This process, called SL RNA trans-splicing, shares core chemical steps with conventional spliceosomal intron removal, but it joins RNA molecules that began as separate transcripts. Conventional cis-splicing is rare in these organisms; SL trans-splicing is central to their messenger RNA maturation.
How does SL RNA trans-splicing work?
In practical terms, the parasite’s trans-spliceosome joins the same short RNA leader to the front of many messenger RNAs. The 2026 study focuses on the second reaction step: ligating the SL exon to the pre-mRNA. Its structures show the complex immediately before that join and after it has occurred.

What did the study capture?
In a study published in Nature Communications on 23 September 2026, researchers isolated endogenous complexes from Leishmania tarentolae using affinity-tagged CDC5L. They analyzed purified material by mass spectrometry and used single-particle cryogenic electron microscopy (cryo-EM) to determine the structures. AlphaFold2-multimer interaction predictions helped the team assign structural densities and describe molecular contacts.
| Structure | Reaction state | Overall resolution |
|---|---|---|
| trans-C* | Before ligation; the SL exon is positioned to join the pre-mRNA. | 2.7 Å |
| trans-P | After the SL exon has been ligated to the pre-mRNA. | 2.8 Å |
The study’s structural models assign four small nuclear RNAs (snRNAs), one pre-mRNA strand and 68 proteins to the complex, with an estimated molecular mass of approximately 3.2 MDa. These are measurements and assignments from the authors’ models, not counts of molecules in a parasite cell.

What makes the complex distinctive?
The structures reveal a conserved spliceosomal core alongside features adapted to SL trans-splicing and trypanosomatids. These include the SL small nuclear ribonucleoprotein (SL snRNP), lineage-specific proteins and expansions of conserved proteins. In the pre-ligation state, the SL exon is positioned for joining; in the post-catalytic state, it is already attached to the pre-mRNA. Together, the snapshots show conserved catalytic chemistry operating through remodeled RNA and protein interactions.
Which parasites does this finding apply to?
The structures were determined from Leishmania tarentolae. The paper discusses the broader trypanosomatid group, which includes Trypanosoma brucei, associated with sleeping sickness; Trypanosoma cruzi, associated with Chagas disease; and Leishmania species, which cause leishmaniasis. The study did not determine structures from every species in that group, so its specific molecular observations should not be treated as direct evidence about every parasite.

Could this discovery lead to a treatment?
The structures offer a starting point for asking whether parasite-specific parts of the trans-spliceosome could be disrupted selectively. Because SL trans-splicing is essential to parasite RNA processing and differs in important ways from human RNA processing, those differences may guide future drug-discovery research.
That possibility remains a research direction, not a demonstrated therapy. The study does not identify a drug candidate or show that a compound can disrupt the complex, kill parasites, spare human cells or treat patients. Its contribution is structural and mechanistic: a clearer view of how the machinery is arranged and works.
