Researchers report that colorectal cancer cells can switch on a YAP1-linked regenerative program associated with metastasis. In mouse models, ceramide production connected a high-fat diet to YAP1 activation, and genetically targeting YAP1 or ceramide-production genes reduced liver metastasis. Human analyses found associations between YAP1-program activity, BMI and survival—but they do not show that diet causes metastasis in people. The findings are not a treatment or a basis for dietary advice.
What pathway did the researchers identify?
The study, titled “Ceramide synthesis mediates colorectal cancer metastasis through a YAP-driven regenerative program”, focuses on YAP1, a protein that can activate genes involved in cell growth and regeneration. The researchers report that metastatic colorectal cancer cells shared activity in a YAP1-linked program associated with development, stem-cell maintenance and tissue repair.

In healthy gut tissue, a related regenerative response helps rebuild the intestinal lining after damage. The study proposes that cancer cells can hijack this repair-like state to support tumor growth and migration during metastatic progression.

How does the proposed ceramide–YAP1 sequence work?
- High-fat diet in mouse studies: The researchers report that this activated enzymes involved in producing ceramides, a class of lipid molecules.
- YAP1 activation: Ceramides were linked to release of a molecular brake that normally keeps YAP1 inactive. YAP1 could then enter the cell nucleus and switch on target genes.
- A regenerative gene program: The activated genes were associated with cell proliferation and migration, processes that can support metastatic progression.
This is the mechanism proposed by the study, not proof that a high-fat diet causes colorectal cancer to spread in people.

What evidence came from mice, and what came from people?

MIT’s September 24, 2026 summary describes the study’s use of tumor organoids from mouse models and patients with colorectal cancer. It does not provide sample counts, effect sizes or confidence intervals. The linked Science article is the primary paper, but its page was inaccessible to the source review, so those numerical details cannot be reported here.
Does this mean obesity or a high-fat diet causes colorectal cancer metastasis?
No. The diet-related finding described by MIT is from mouse studies. The human findings connect BMI and YAP1-activated gene expression through observational RNA-sequencing analyses; they do not establish that obesity or diet caused metastasis in those patients. The researchers also say the YAP1 program is not specific to obesity, though it may be accentuated in obesity.

Could this lead to a treatment?
The researchers plan to investigate drugs that inhibit DEGS1 and DEGS2, enzymes involved in ceramide production. That is a future drug-development direction, not a treatment patients can use now. The study summary does not establish an approved or available therapy targeting this pathway.

There is also a safety challenge: ceramides are essential in healthy tissues. A drug that suppresses their synthesis would need to act selectively enough to avoid disrupting normal biological functions.

What should patients do with this finding?
Do not change a cancer diet, take ceramide-related supplements or seek an unapproved pathway-targeting drug on the basis of this report. MIT says the findings do not yet translate into dietary advice for people diagnosed with cancer. Patients considering diet changes or treatment decisions should discuss them with their oncology team.
