A pharmaceutical AIoT pipeline is a governed connection between production equipment, control and data-acquisition systems, edge infrastructure, and higher-level applications such as historians, MES, quality systems, and analytics. Design it as interoperating layers—not as one “AI platform”—and define each data stream’s purpose, authority, security controls, and validation needs before connecting systems. The architecture can support manufacturing visibility and analysis, but it does not by itself establish regulatory compliance or make an AI output suitable for a GxP decision.

What belongs in a pharmaceutical AIoT pipeline?

Pharma 4.0 applies Industry 4.0 concepts to pharmaceutical product lifecycle and manufacturing while accounting for regulatory practice and the complexity of pharmaceutical products and processes. It is an organizational and quality-system transformation as well as a technology effort. ISPE’s Pharma 4.0 mission is “to provide practical guidance, embedding regulatory best practices, to accelerate Pharma 4.0™ transformations.”

A useful design pattern is a flow from process instruments to control or acquisition systems, then through a managed edge connection to operational and analytical systems. That pattern is grounded in an ISPE proof of concept, not a universally prescribed topology. A plant may already have historians, laboratory or quality applications, and enterprise systems that shape where data should go.

Layer Typical role Design concern
Equipment and instruments Measure process variables or equipment condition. Identify each signal’s source, units, context, timestamp, and quality status.
Control and acquisition A DCS or other system performs process control and local operational visibility. Keep control behavior and safety functions distinct from downstream analytics unless a separately assessed design explicitly connects them.
Edge connectivity Connect devices and systems, select or route data, and provide a managed exchange point between OT and IT or cloud environments. Define boundary controls, local behavior during disconnection, and the device’s configuration and security lifecycle.
Operations and analytics Historians, analytics, MES, laboratory and quality systems, and enterprise applications consume or act on information according to their roles. Establish which system is authoritative for each record and how data context and lineage are preserved.

MES is one possible destination or integration point in this flow, not necessarily a direct endpoint for every sensor. Determine whether a signal is needed for execution, review, traceability, maintenance, or analysis, then route it to the systems responsible for that use. Accessing data at its source may avoid some replication and associated data-integrity or configuration and validation overhead, but it does not automatically eliminate those obligations.

How do sensors connect to MES and other systems?

Start with the instrument and its existing control or acquisition path. A DCS may remain responsible for control while an edge device exposes selected data to other systems. From there, information may pass to a historian, MES, quality or laboratory applications, or an analytics environment. The integration should preserve enough context for the receiving system to interpret the value correctly; a number without source identity, timestamp, units, process context, and lineage may not be useful evidence.

An ISPE proof of concept illustrates one possible arrangement: a smart sensor connected to a DCS using OPC UA and also to an edge device using Bluetooth Low Energy and OPC UA. Two secure channels to a cloud environment were implemented: an edge-device path using MQTT and a connectivity-server path using WebSocket Secure. The cloud platform handled collection, analytics, and presentation, and communications paths were implemented redundantly in that demonstrator. This is an example of a tested setup, not a requirement to send all plant data to a cloud platform or to reproduce its topology.

Where do OPC UA, MQTT, ISA-95, PackML, and IEC 61499 fit?

These technologies and standards address different parts of interoperability. Selecting a protocol does not, on its own, define the meaning, quality status, or regulatory significance of every data item.

Technology or standard Role described in ISPE material What it does not establish by itself
OPC UA Cross-platform data exchange; used in the proof of concept for sensor connections to the DCS and edge device. That every connected system interprets the same data identically or that a particular implementation is qualified.
MQTT Used for the proof-of-concept edge-device path to the cloud environment. That a connection is secure or suitable for a particular use without appropriate configuration and controls.
Bluetooth Low Energy Used in the proof of concept as part of a smart-sensor connection to an edge device. Universal compatibility with installed equipment or suitability for every plant environment.
ISA-95 A way to describe information flow between manufacturing operations management and other systems. Compliance with pharmaceutical requirements merely by adopting the standard.
PackML A way to improve consistency in machine data. A complete data model for every pharmaceutical process or system.
IEC 61499 Discussed in the ISPE material in relation to distributed automation. Regulatory approval of a distributed control architecture.

In its proof of concept, ISPE reported OPC UA and MQTT were easy to implement and robust in information transfer. That observation belongs to the demonstrator; it is not a controlled comparative benchmark or a guarantee for another plant, network, or implementation.

What should edge computing do?

Use the edge as a managed connectivity and data-handling layer, not as an assumed replacement for control systems or quality systems. Depending on the design, it can connect devices using plant-relevant interfaces, select or route signals, and mediate exchange across an OT/IT boundary. Specify which functions run locally, which data are forwarded, and what happens when a sensor, network, or receiving service is unavailable.

The ISPE proof of concept demonstrates that smart sensors and edge connectivity can be added to existing plant equipment. It supports brownfield feasibility in that setup; it does not establish low cost, easy integration, universal compatibility, or a particular implementation’s suitability. For any retrofit, check the installed interfaces, data semantics, environmental constraints, access controls, support lifecycle, and change-control impact.

How should AI be governed in the pipeline?

Treat AI as a governed analytics capability whose role is explicitly bounded. Before using a model output, state whether it is informational, advisory, or part of a decision process; identify the intended users and the consequence of an incorrect or unavailable result. Do not assume that collecting data through a connected architecture makes an AI model validated, accepted, or appropriate for a specific GxP decision.

FDA’s CDER FRAME page lists public feedback on regulatory considerations for AI in drug manufacturing published in May 2025 and a related FDA/PQRI workshop held September 26–27, 2023. This indicates active regulatory discussion; it does not establish a blanket approval rule or mean that FDA has approved a particular architecture or model.

What governance questions should the design answer?

ISPE identifies an established pharmaceutical quality system and controlled processes and products as prerequisites for applying Industry 4.0 concepts in pharmaceutical settings. It also emphasizes that the transformation involves organizational, cultural, process, resource, and technology considerations, and that increased connectedness raises security needs.

  • Purpose and authority: What is the intended use of each data stream and AI output? Is it process-control data, quality evidence, maintenance information, or advisory analysis? Which system is authoritative for the record?
  • Meaning and integrity: Can the receiver establish the source, timestamp, identity, units, context, and lineage of a value? How are missing, delayed, duplicated, or suspect readings represented?
  • Security boundaries: How are OT and IT environments separated and protected? Which users and services can access each connection, and how is trusted access controlled?
  • Degraded operation: What does the system do during a network outage, loss of a data source, or bad sensor reading? Which functions continue locally, and how are gaps or recovery events recorded?
  • Lifecycle control: How are configurations, interfaces, access permissions, software, and AI models changed and documented? What audit trail is needed?
  • Assurance: What qualification or validation evidence is appropriate for each component and use, based on intended use and risk? How will a change affect that evidence?

The architecture is only one input to these decisions. The required assurance depends on what a system is intended to do and the risks of its use; a protocol, edge device, or AI component does not confer compliance simply by being connected.

How should teams choose an implementation approach?

There is no universally best topology in the source material. Compare candidate designs against the plant’s actual systems and intended uses rather than selecting a platform first.

  • Interoperability: Can the design work with installed sensors, DCS, MES, historians, and enterprise applications?
  • Protocols and semantics: Are the interfaces supported, and will systems preserve the meaning and context of the data?
  • Data placement: Is source access practical, or is replication needed? What are the implications for integrity, availability, and controlled records?
  • Security: Are OT/IT boundaries, identities, permissions, and trusted access explicitly designed?
  • Traceability: Can users follow a value from source through transformation to its use in a record or analysis?
  • Validation and change: What lifecycle controls follow from the system’s intended use and risk, and how burdensome are changes across connected components?
  • Resilience: What happens during a disconnection or degraded sensor quality, and how does the design distinguish stale or invalid data?
  • Scale: Can the approach be maintained across additional lines and sites without losing consistent governance?

ISPE materials describe integration challenges as systems scale and discuss secure edge-driven transfer and regulatory data integrity as architecture considerations. Those are reasons to assess scale and governance early, not proof that a particular architecture will solve them automatically.
