Short answer: mushrooms and monosodium glutamate (MSG) can both make food taste more savory, but they do it as different kinds of ingredients. MSG is a defined sodium salt of glutamic acid, so it delivers glutamate directly. A mushroom is a food matrix containing glutamate and other amino acids, 5′-nucleotides such as guanylate, organic acids, and many peptides. Those compounds work together with the mushroom’s aroma and texture. Mushroom peptides are therefore one contributor to mushroom umami, not “natural MSG” and not the whole explanation.
The strongest interaction is between glutamate and certain 5′-ribonucleotides. Guanylate, which is found abundantly in mushrooms according to a 2023 perspective, can intensify glutamate’s umami effect. A 2025 review also reported that 55.66% of 155 identified edible-mushroom umami peptides had taste thresholds below MSG; that is a comparison of selected, identified peptides in review data, not proof that whole mushrooms universally taste stronger than MSG.

What are mushroom peptides and MSG?
“Mushroom peptides” describes short chains of amino acids found in mushroom tissue or produced when mushroom proteins are broken down during growth, drying, cooking, fermentation, or other processing. Their sequences and concentrations vary by species and by handling. Some can taste umami, some may alter the perception of other tastes, and many have no demonstrated sensory role in an ordinary serving.
MSG is a single, defined compound: the sodium salt of the amino acid glutamic acid. In a moist food it separates into sodium and glutamate ions. The glutamate is the part associated with umami taste; the sodium contributes to the ingredient’s salt content. MSG is not a protein fragment and is not chemically equivalent to a mushroom peptide.

A whole mushroom supplies a mixture rather than one purified signal. Reviews of mushroom umami describe proteins and peptides alongside free amino acids, organic acids, and 5′-nucleotides. This distinction matters when someone asks whether mushrooms “contain MSG.” Mushrooms can contain free glutamate, the same amino acid that MSG supplies after dissolving, but they are not made of MSG and do not have a single standardized MSG concentration.
How MSG produces an umami signal
Glutamate activates umami receptors
The 2023 perspective by Takashi Yamamoto and Chizuko Inui-Yamamoto identifies glutamate and MSG as umami substances. The best-characterized receptor is the T1R1/T1R3 heterodimer; metabotropic glutamate receptors have also been reported to participate in umami sensing. When glutamate occupies these receptors, sensory cells send a signal that the brain interprets as savory, broth-like taste.
A receptor pharmacology review published in 2021 likewise describes T1R1/T1R3 activation by recognized umami substances. The receptor biology explains why a small, chemically defined addition can change a dish even when it contributes little aroma or texture of its own: it targets a taste pathway rather than supplying a complete food flavor.
MSG is targeted, not a complete flavor system
Because MSG supplies glutamate in a predictable form, it is useful when a cook wants more umami without adding mushroom aroma, color, solids, or the other tastes of a food ingredient. That precision is also its limitation. MSG does not reproduce the roasted, earthy, sulfurous, or woody aromas that distinguish different mushrooms, and it does not provide mushroom texture.

How mushrooms create savory flavor
Several chemical families contribute at once
The 2025 review by Wang and colleagues surveys mushroom umami across proteins, peptides, amino acids, organic acids, and 5′-nucleotides. The relative contribution of each family depends on species, maturity, storage, drying, extraction, and cooking. A broth made from dried mushrooms is therefore not chemically identical to a broth made from fresh mushrooms, even when both are described as “mushroom flavor.”
- Free amino acids: glutamate and other amino acids provide taste signals directly or modify balance with salt, acidity, and bitterness.
- Peptides: some short sequences have intrinsic umami activity or can influence taste intensity and persistence. Their activity is sequence-specific, so “peptide” is not a synonym for “umami.”
- 5′-nucleotides: compounds such as guanylate (5′-GMP) can strongly reinforce glutamate’s umami response.
- Organic acids and other metabolites: these affect sourness, aroma, mouthfeel, and the way savory notes are perceived in a complex food.
- Volatile aroma compounds: aroma is not umami taste, but it contributes heavily to the recognizable identity of a mushroom dish.
That combination is why mushroom flavor cannot be reduced to a single peptide number. Two species can contain different mixtures, and the same species can change after drying or extraction. Current reviews do not establish a universal umami-peptide concentration or a potency ranking for all edible mushroom varieties sold to consumers.
Guanylate helps explain the “brothy” effect
Guanylate is a 5′-ribonucleotide identified as abundant in mushrooms in the 2023 perspective. On its own, it is one component of taste; in the presence of glutamate, it can produce a much stronger umami impression than either substance would produce alone. The amount in a particular mushroom product is not established by the general literature, so it is not valid to assign every mushroom powder or dried mushroom the same guanylate level.
Why glutamate and nucleotides taste stronger together
The interaction between glutamate and 5′-ribonucleotides is often called umami synergy. The 2023 perspective describes a marked rise in perceived umami when glutamate is mixed with nucleotides such as inosinate or guanylate. In practical terms, a mushroom ingredient that supplies guanylate may taste especially savory when paired with a food that supplies glutamate, and an MSG-seasoned dish may taste fuller when it also contains a nucleotide source.

Synergy is not a fixed recipe ratio and does not mean every mushroom product will boost every dish by the same amount. The perceived result depends on concentration, salt level, acidity, temperature, aroma, and the other ingredients in the meal. The evidence supports the interaction as a general sensory principle, not a guaranteed intensity for a particular soup, sauce, or commercial seasoning.
What mushroom peptides add beyond glutamate
Peptides are sequences, not one ingredient
A peptide is a chain of two or more amino acids joined by peptide bonds. Its taste depends on the sequence, length, charge, hydrophobicity, and concentration. Some mushroom-derived peptides identified in laboratory work taste umami at low concentrations; others may be tasteless, bitter, or active only in combination with other compounds. Calling all mushroom peptides “natural MSG” erases these chemical differences.
The 55.66% threshold figure, correctly interpreted
A 2025 PubMed-indexed review reported that 55.66% of 155 identified edible-mushroom-derived umami peptides had taste thresholds below MSG. A threshold is the concentration at which a trained sensory panel can detect a taste under the review’s test conditions. The statistic applies to the selected peptides that had been identified and evaluated, not to every peptide in every mushroom.

It also does not compare a whole mushroom, a serving of mushroom powder, and a measured amount of MSG in the same food. The concentration of each peptide in an intact mushroom, its stability during cooking, its release into broth, and interactions with salt and aroma all matter. The result is evidence that some isolated mushroom peptides can be highly taste-active, not evidence that mushrooms as a class are universally more potent than MSG.

Umami and kokumi are related but not identical
Umami is a taste quality associated primarily with glutamate and certain nucleotides. Kokumi is used for a different sensory dimension often described with words such as thickness, mouthfulness, continuity, and persistence. Kokumi compounds may have little taste on their own but can make existing flavors seem more substantial.
Yamamoto and Inui-Yamamoto propose that glutamate can interact with kokumi substances in complex foods. Their perspective states: “The requisite for sufficient palatability and kokumi flavor is a good balance of umami and kokumi substances.” They also say that the direct interactions and the mechanism by which MSG makes food more palatable remain poorly understood. This is a proposed framework, not a settled explanation of every mushroom flavor effect.
Do not assume that every mushroom peptide is a kokumi compound. A peptide can be umami-active without producing kokumi sensations, and a kokumi-active substance need not taste strongly umami by itself. More work is needed to connect individual mushroom molecules with the sensory experience of a complete meal.

Choosing between dried mushrooms, mushroom powder, and MSG
Choose a mushroom ingredient when you want a recognizable mushroom profile
Dried shiitake and other dried mushrooms bring aroma, color, solids, and a species-specific flavor as well as umami-related compounds. Rehydrating them for broth or grinding them into a powder preserves more of that food identity than adding a purified glutamate source. The trade-off is variability: the savory strength and aroma depend on the mushroom and its processing, and the solids may not suit a clear sauce or delicate dish.

Choose MSG when you want precise glutamate seasoning
MSG seasoning is useful when the dish already has the desired aroma and texture but tastes thin or lacks savory lift. It does not replace the flavor of mushrooms, meat, seaweed, fermented foods, or browned ingredients; it supplies glutamate more directly. Use the amount specified by the recipe or product instructions and judge the finished dish in context, because salt, acidity, sweetness, and aroma change the perceived result.
Use both when the dish benefits from mushroom identity and controlled umami
A broth can obtain mushroom aroma and nucleotide-rich complexity from dried mushrooms while using a small, measured glutamate addition for consistency. Conversely, a dish seasoned with MSG can gain a more distinctive character from mushroom powder. The combination is chemically plausible because glutamate and guanylate can work synergistically, but the literature does not provide a universal amount that guarantees a particular sensory intensity.
A practical way to build savory flavor
- Decide which outcome is missing. If the dish lacks mushroom aroma or texture, start with a mushroom ingredient. If those are already present but the taste seems flat, a targeted glutamate source may address the gap.
- Account for the base ingredients. Foods such as meat, tomatoes, cheese, seaweed, fermented sauces, and mushrooms may already contribute glutamate or nucleotides. The final balance matters more than the identity of one ingredient.
- Add one variable at a time. Adjust the mushroom component and the MSG component separately so you can identify whether aroma, taste intensity, saltiness, or mouthfeel changed.
- Taste after the dish reaches its serving condition. Dilution, heat, acidity, and cooking time alter perception. A seasoning that seems strong in a concentrated sample may be less noticeable in a finished broth.
- Stop when the dish is balanced. More umami is not always better; excessive seasoning can make a dish seem harsh, salty, or out of balance with its aroma and acidity.
What current evidence can and cannot establish
- It can establish that mushrooms contain multiple umami-related chemical families and that MSG is a defined glutamate salt.
- It can support the general synergy between glutamate and 5′-ribonucleotides such as guanylate.
- It can report taste-threshold results for selected, isolated mushroom peptides.
- It cannot provide a universal peptide content for all edible mushrooms or all retail mushroom products.
- It cannot show from threshold data alone that whole mushrooms taste stronger than MSG in ordinary meals.
- It cannot assign a specific kokumi effect to every mushroom peptide.
- It cannot serve as a safety, sodium, or nutrition comparison; those questions require dedicated regulatory and nutrition evidence.
Common misconceptions about mushroom umami
“Mushrooms are naturally full of MSG.”
Mushrooms can contain free glutamate, but MSG is the sodium salt of that amino acid. A mushroom is a complex food containing many other molecules. Describing the entire mushroom as MSG is chemically inaccurate.
“All mushroom peptides are stronger than MSG.”
The 55.66% figure concerns 155 identified peptides reviewed in 2025. It does not include every peptide, does not describe every species, and does not measure whole mushrooms or prepared meals head-to-head against MSG.
“Umami and kokumi mean the same thing.”
Umami is a taste quality linked to glutamate and nucleotides. Kokumi describes proposed changes in thickness, mouthfulness, and continuity. They can interact, but they are not interchangeable terms.
“Adding a mushroom ingredient guarantees nucleotide synergy.”
Synergy requires the relevant compounds to be present at effective concentrations. Species, processing, storage, dilution, and the rest of the recipe determine whether the interaction is noticeable. General mushroom descriptions do not establish a guanylate level for a particular product.
The bottom line for cooks and food formulators
MSG is the predictable route to more glutamate-driven umami. Mushrooms are the broader route: they combine glutamate, other amino acids, peptides, guanylate and other nucleotides, organic acids, and aroma compounds into a variable food matrix. Mushroom peptides can be potent in laboratory threshold tests, but they are only one part of that matrix. For a dish with a distinctive mushroom character, use a mushroom ingredient; for controlled savory adjustment, use MSG; for layered flavor, combine them while treating the glutamate–nucleotide interaction as a useful principle rather than a guaranteed formula.
