In a mouse study reported by the Leibniz Institute for Neurobiology on 14 October 2026, inhibitory synapses on dendrites were reorganized more often after learning, while synapses at the axon initial segment became comparatively stable. The total number of inhibitory synapses changed little, pointing to a change in arrangement and persistence rather than a broad gain or loss of inhibition.
What the study found
Inhibitory synapses reduce or constrain a neuron’s electrical activity. The new work suggests that their response to learning depends strongly on where they are located on the same cell. Dendritic synapses, positioned on the branches that receive and integrate incoming signals, showed more frequent reorganization after the animals learned. Inhibitory synapses at the axon initial segment (AIS), the short region where the neuron’s electrical output is initiated, were more stable.

That distinction matters because a neuron can alter how it processes incoming information without constantly changing the mechanisms that determine whether it sends an output signal. The researchers describe this as a location-based division of labor: input-side inhibition remains flexible, while output-side inhibition is stabilized.
Alessio Attardo of the Leibniz Institute for Neurobiology said, “It is surprising that inhibitory synapses within the same nerve cell do not respond uniformly to learning. Their exact position appears to help determine whether they are remodelled or stabilised. This gives us a new insight into how a neural network can remain both plastic and stable at the same time.”

How dendritic and axon-initial-segment synapses differed
| Synapse location | Observed outcome after learning | Functional interpretation |
|---|---|---|
| Dendrites | Inhibitory synapses were reorganized more frequently | Flexible control of incoming-signal integration |
| Axon initial segment | Inhibitory synapses became more stable | More consistent control of output initiation |
The circuit roles in the table are an interpretation of the location-specific pattern, not proof that one population is universally more important. Dendrites combine signals arriving from other neurons, so remodeling inhibition there could adjust which combinations of inputs have influence. The AIS sits at the transition between the cell body and axon; stabilizing inhibition there could help keep the threshold for producing an electrical signal within a reliable range while other parts of the cell adapt.
How researchers followed the synapses
The study used a longitudinal design so that the same neurons and synaptic contacts could be examined repeatedly rather than compared only between separate groups of animals.

A neuron’s dendrites are its main input branches. Signals arriving there are combined across space and time, and inhibitory contacts can change how strongly those inputs affect the cell. The AIS has a different job: it is the region where the combined electrical state of the neuron is converted into the start of an action potential, the signal that travels down the axon.

Because those locations sit at different stages of information processing, changing them in different ways offers a way to learn without making the entire cell unstable. The study’s result does not mean that AIS synapses never change or that dendritic synapses are always replaced. It reports different tendencies across the measured populations after this learning task.
How learning can be flexible without becoming chaotic
A common challenge for neural circuits is balancing plasticity with stability. If every inhibitory contact changed freely, the circuit’s output could become unpredictable. If none changed, new experiences could have little effect. The observed pattern offers a possible solution: remodel inhibition where incoming information is evaluated, while preserving a steadier control point for output generation.

The nearly unchanged total synapse count reinforces that interpretation. Learning did not simply add inhibition throughout the neuron or remove it everywhere. Instead, the important variables were which contacts persisted, which were reorganized and where those contacts were located.

- It provides evidence that learning-related changes in inhibitory synapses can be location-specific within a single hippocampal neuron.
- It shows more frequent reorganization on dendrites and greater stability at the AIS under the tested learning conditions.
- It does not demonstrate that learning increases or decreases inhibition everywhere in the brain.
- It does not establish a treatment for memory, a clinical mechanism or a direct explanation of human learning.
- Because the experiments were performed in mice, further research is needed to determine whether comparable location-dependent processes occur in humans.
Why the finding matters
Many descriptions of synaptic plasticity treat a neuron as if all of its connections respond similarly. This study argues for a more precise view in which the physical address of an inhibitory synapse helps determine its response to experience. That perspective could improve models of how hippocampal circuits encode memories while preserving reliable signaling, but the result is still a fundamental mechanism demonstrated in an animal model rather than a confirmed account of human memory.
