Start by preserving the raw observations and the exact conditions for every run. Then determine whether the problem is random scatter, a consistent bias, or a qualitatively different outcome such as no reaction or a lower yield. Check the measurement system and calculations before changing reaction conditions, and change one controllable factor at a time. Replicates can show how much results vary, but averaging cannot remove a systematic error.
This method works for titrations, synthesis, analytical measurements, and other chemistry experiments. It is general guidance rather than a diagnosis of one protocol, so the experiment’s written method, safety data, institutional procedures, and supervisor’s instructions always take priority.

1. Describe exactly what is inconsistent
“Different results” can describe several different patterns. Name the pattern before looking for a cause; each pattern points to a different test.

Wide scatter between apparently identical runs
Results move up and down without a clear direction. This is consistent with random variation, but it can also indicate an uncontrolled condition that changes from run to run, such as temperature, mixing, timing, or liquid handling.

A tight cluster that is always too high or too low
Close agreement among repeats demonstrates repeatability, not accuracy. A consistent offset from an accepted value, reference material, or independently established result suggests systematic error in the instrument, calibration, method, calculation, or sample preparation.
A qualitatively different outcome
“No reaction,” an unexpected color or precipitate, a phase change, or a substantially lower yield requires comparison of reaction conditions, reagent identity and quality, and the order and rate of additions—not just more arithmetic.
Make a run-by-run record before changing anything
Do not discard an inconvenient result or rewrite a notebook entry from memory. Preserve the raw readings and observations, including zeros, failed runs, unusual colors, temperatures, and times. For each run, record:

- Temperature, reaction time, waiting time before a reading, and the timing of each operation.
- Mixing method and duration, container, order of addition, and rate of addition.
- Observations such as cloudiness, gas evolution, color, precipitate, leaks, or signs of evaporation.
- Units, conversions, significant figures, equations, spreadsheet formulas, and any data-processing steps.
Place the runs side by side. The first useful finding is often a condition that changed, not a person who “made a mistake.”
2. Use a controlled troubleshooting sequence
A disciplined sequence prevents several changes from masking the cause. After recording the evidence, work from the measurement and calculation layers toward the chemistry.
- Classify the pattern. Mark the result as scattered, consistently shifted, or qualitatively different.
- Compare records. Highlight every changed quantity, setting, lot, temperature, time, mixing action, and addition step.
- Verify measurement suitability. Confirm that the instrument can measure the required magnitude and precision.
- Verify zeroing and calibration. Follow the applicable procedure; do not assume a recently used instrument is still correctly set.
- Recheck calculations and data handling. Recalculate from the raw readings with units visible.
- Separate random variation from bias. Use consistent replicates for spread and a suitable reference or independent method for accuracy.
- For reactions, compare physical and chemical conditions. Examine scale, reagents, solvent, temperature, time, mixing, and addition sequence.
- Change one factor under approved conditions. Record the change and its rationale, then repeat the appropriate control or run.
If a change improves a result, that is evidence about the changed condition, not proof that every other condition was correct. Keep the original and modified records distinct.

3. Check the measurement system before blaming the chemistry
Match the instrument to the task
Measurement instruments differ in sensitivity and capacity. The University of Iowa CHEM:1110 Lab Manual explains that those properties affect results. An instrument may be capable of displaying many digits yet be unsuitable for the sample size, range, or precision required. Check the method’s stated range and resolution against the actual quantity being measured.

This applies to balances, volumetric devices, thermometers, pH meters, spectrometers, burets, pipettes, and other equipment. First use the laboratory’s existing instrument that meets the method’s requirements; inconsistent results do not automatically justify buying new equipment.
Check zero, calibration, and operating state
Confirm zeroing, calibration, warm-up or equilibration requirements, correct units, and the selected range. Follow the instrument’s approved procedure and record the check. A calibration performed for one range, mode, or probe condition may not establish performance in another. Inspect for dirty or damaged contact surfaces, bubbles where the method excludes them, unstable readings, leaks, or a warning message, but do not improvise a repair.
Test the system with a known value
Cornell’s Virtual Lab guidance recommends using known reference values to evaluate a procedure. A suitable reference can reveal a consistent offset that repeated sample readings would hide. If feasible, measure the reference with an independent instrument or technique as well. Independence matters: two devices that share the same calibration or preparation fault can reproduce the same bias.
4. Recalculate from raw data
Precise-looking numbers can still be wrong because of a unit conversion, transcription, formula, or data-entry error. Rework the calculation from the original observations, preferably independently of the spreadsheet or program that produced the first answer.
- Write units beside every measured value and confirm that they cancel correctly through each conversion.
- Check whether a mass, volume, concentration, dilution factor, or stoichiometric coefficient was entered in the intended units.
- Inspect decimal placement, copied cells, row and column references, hidden filters, and formulas that were overwritten by values.
- Apply significant-figure rules consistently; do not create extra certainty by reporting digits the measurement cannot support.
- Check that the selected equation matches the procedure and that the computer program behaves as expected for a simple test case.
Cornell specifically advises checking equations and computer programs against expected behavior. Keep the unrounded intermediate values for calculation, but report a final value whose rounding matches the uncertainty or spread you can justify.

5. Distinguish random error from systematic error
Random variation
Random error produces variation among measurements made under apparently identical conditions. Replicates help estimate its size. A larger spread means that a single result is a weak description of the procedure; it does not identify which operation caused the variation.
Systematic bias
Systematic error shifts results in a consistent direction. A method can therefore be highly repeatable and still inaccurate. Cornell notes that “Systematic error can be difficult to identify and correct.” Averaging more results does not remove that offset; it can make the biased mean look more convincing.
Use the right comparison
| Question | Useful evidence | What it can and cannot establish |
|---|---|---|
| How tightly do repeats agree? | Spread of replicate results under consistent conditions | Repeatability or precision; not accuracy |
| Are results close to an accepted value? | Certified or otherwise suitable reference, or a validated comparison value | Evidence about accuracy; the reference must be appropriate |
| Are repeats independent? | Separate preparations and runs, rather than repeated readings of one preparation | Whether the result includes preparation-to-preparation variation |
| Could both methods share a bias? | Comparison with an independent instrument, technique, or preparation | Stronger evidence than two methods sharing the same source of error |
Use a reference or an independent method when feasible, and test the whole procedure rather than only the final instrument reading. If the reference is also prepared incorrectly, it cannot expose that preparation error.
6. Design replicates that answer the question
Know what is being repeated
Repeated readings of one sample characterize reading noise or short-term instrument variation. Independent experimental runs include separate weighing, dilution, reagent preparation, or reaction setup and therefore test more of the procedure. State which type you performed; the distinction changes the meaning of the spread.
Keep conditions consistent
To estimate ordinary variability, keep the method, operator instructions, instrument settings, temperature, timing, and sample handling consistent. If you deliberately vary one factor, label the runs as a comparison experiment rather than pooling them as ordinary replicates.
Report mean, spread, and number
Report the number of observations and a suitable measure of spread with the mean. The University of Iowa manual gives the illustrative titration report 12.04 ± 0.02 mL (n=5), with the mean rounded to match the reported standard deviation. This is a worked example from a 2020 lab manual, not a universal precision target for titrations or other chemistry experiments.

- Please Note: To reach 10Gbps speed, keep the cable under 3.3 ft. For USB A Male to USB C adapters, try flipping the USB C connector. USB C Male to USB A adapters support bidirectional 10Gbps transfer within 3.3 ft
Also identify the source of the replicates—for example, one sample stock, separate wells or containers, separate preparations, separate instrument runs, or separate experimental runs. NIH Assay Guidance Manual reporting advice emphasizes this distinction and notes that temperature, instrument limits, liquid handling, and assay fluctuations can contribute to random variation. That guidance was written for non-clinical assay reporting and should be adapted to the chemistry experiment rather than copied as a universal rule.

7. Compare reaction conditions systematically
When a reaction works once but not the next time, compare the complete condition set with the original notebook or procedure. The University of Rochester Department of Chemistry identifies the following reproducibility factors.
Scale
Compare the absolute masses and volumes, not only the nominal ratios. Small-scale work can make transfer losses, evaporation, or measurement resolution proportionally more important. A change in scale can also alter heat removal, mixing, and the time needed for additions.
Reagent and solvent identity or quality
Verify names, concentrations, grades where specified, storage, preparation date, appearance, and lot information. Do not silently substitute a similar-looking reagent or solvent. If the procedure depends on a prepared solution, check its concentration and preparation record.
Temperature and time
Compare actual temperature, not merely the set point, and record when the reaction reached the relevant temperature. Check reaction time, preheating or cooling time, hold time, and the delay before sampling or measurement. A run can differ if the timer starts at a different procedural point.

Order and rate of addition
Compare which material was added first, whether additions were continuous or portionwise, and how quickly they occurred. Addition rate can change local concentration, heat release, precipitation, or gas evolution. Match the documented sequence instead of treating “added” as a sufficient description.
The Rochester guidance also warns that procedures may omit details and that a published or recorded result may not reproduce exactly as written. Treat an undocumented condition as unknown. Do not change several unknowns at once; otherwise an apparent improvement cannot identify the cause.
8. A practical decision tree for the next run
If results are scattered
- Check timing, temperature, mixing, addition rate, sample handling, and instrument stability for run-to-run changes.
- Use independent preparations if preparation variability is part of the question.
- Repeat enough consistent runs to estimate spread, while recording every deviation.
If results cluster but miss a reference
- Check calibration, zero, units, calculations, and the reference’s suitability.
- Measure a known quantity and, where feasible, compare with an independent instrument or technique.
- Do not expect a larger sample of the same biased measurements to correct the offset.
If the reaction outcome changes qualitatively
- Compare reagent identity and quality, solvent, scale, temperature, time, mixing, and addition sequence.
- Inspect records for contamination, evaporation, incomplete dissolution, or an unrecorded delay.
- Repeat only within the method’s safety controls and with appropriate supervision.
9. Change one factor and preserve the comparison
After identifying a plausible cause, write a testable prediction: for example, “If the temperature during addition was lower than specified, restoring that temperature should return the reference response while the other conditions remain unchanged.” Define the measurement that will support or contradict the prediction.

Keep a control or reference run when feasible, and label the modified run with the exact change. If several changes are unavoidable for safety or protocol reasons, record them as a package and acknowledge that the individual causes cannot be separated from that comparison.
10. Safety and escalation
Troubleshooting is not permission to override a method, substitute an unapproved chemical, bypass a guard, increase scale, or repeat a hazardous reaction. Follow the experiment-specific safety instructions, safety data, institutional procedures, waste requirements, and supervisor directions. Stop and seek supervision for an unexpected exotherm, pressure, gas evolution, exposure, damaged equipment, unknown labeling, or any condition outside the approved procedure.
Ask a supervisor or experienced colleague to review the run records when the cause remains unclear, when a reference test fails, or when the result affects a consequential decision. Provide the raw data, condition comparison, instrument checks, calculations, and the exact change proposed; a complete record is more useful than a conclusion that the result was simply “human error.”
11. How to report the final finding
A useful report states what happened, what was tested, and what remains uncertain.
Do these 3 things before closing this tab:
1Repair Windows errors before they cause bigger problems2Scan for outdated or missing drivers - takes under a minute3Clear out junk files and repair common Windows errors- Describe the inconsistency and show the raw observations or a traceable summary.
- State the number and type of replicates, the mean, and the spread statistic.
- Identify instrument checks, calibration or reference results, and calculation verification.
- List the reaction conditions that differed, including scale, materials, temperature, time, mixing, and addition sequence.
- Separate evidence of repeatability from evidence of accuracy.
- State which change was tested, what happened afterward, and which alternatives remain possible.
Measurements are rarely exact; the University of Iowa manual puts it plainly: “Measurements are rarely exact; associated with each measurement is an uncertainty or error.” Reporting that uncertainty makes a result more credible, not less useful.
Quick Recap
12. A compact checklist before you repeat
- Raw readings and observations are preserved.
- Every run’s sample, reagent, lot, quantity, scale, instrument, setting, temperature, time, mixing, and addition sequence are recorded.
- The instrument is suitable, zeroed, and checked or calibrated under the applicable procedure.
- Units, conversions, significant figures, equations, formulas, and data entry have been independently reviewed.
- You have identified whether the issue is scatter, bias, or a qualitative reaction change.
- Replicates are labeled as repeated readings or independent runs.
- A suitable reference or independent method has been considered for accuracy.
- Only an approved, safety-compliant change will be made, with other conditions held as constant as possible.
