The most practical way to measure galactose oxidase (GalOx) is an initial-rate assay that couples the enzyme’s hydrogen-peroxide output to horseradish peroxidase (HRP) and ABTS. In a transparent 96-well plate, monitor the rise in absorbance at about 414–420 nm, convert the initial slope with a hydrogen-peroxide standard curve, and report activity using a stated unit definition.
GalOx methods are preparation-specific. Buffer pH, substrate concentration, temperature, copper activation, enzyme concentration, coupling-reagent amounts, wavelength, path length and the definition of one unit must all accompany the result. The protocol below gives a published microplate starting point, a cuvette variant, calculation formulas, controls, troubleshooting and direct-product alternatives.

What GalOx activity measures
Galactose oxidase oxidizes the C6 hydroxyl group of galactose or a galactoside to an aldehyde. Molecular oxygen is reduced at the same time, producing one equivalent of hydrogen peroxide for each substrate molecule oxidized. The overall analytical relationship is therefore:

For Fgr GalOx, the stoichiometric statement used in the 2022 RSC Advances method is that oxidation of 1 mole of substrate consumes 1 equivalent of O2 and generates 1 equivalent of H2O2. Most routine assays measure that peroxide indirectly rather than measuring the oxidized carbohydrate itself.
In the workhorse coupled assay, HRP uses GalOx-generated H2O2 to oxidize ABTS. The resulting ABTS radical cation is colored, so its formation is followed spectrophotometrically. During the validated initial-rate interval, the increase in absorbance is proportional to the rate of peroxide generation and, by stoichiometry, to GalOx activity.
Choose the readout before you prepare samples
Use an HRP/ABTS assay for fast activity screening and routine comparisons. Choose Amplex Red when fluorescence or a low-background peroxide readout is useful. Use HPAEC-PAD when you need to identify and quantify the oxidized carbohydrate product directly, or when peroxide coupling is not trustworthy.
| Method | What is detected | Typical instrument | Best use | Main limitations |
|---|---|---|---|---|
| HRP/ABTS | Colored ABTS oxidation caused by GalOx-generated H2O2 | Absorbance plate reader or spectrophotometer | High-throughput activity screens and routine kinetics | Indirect; blank oxidation, peroxide cross-reactivity and ABTS-radical instability can distort the signal |
| HRP/Amplex Red | Resorufin formed from H2O2 | Fluorescence-capable reader or spectrophotometer | Fluorescence-compatible peroxide measurements and small-volume assays | Still an indirect peroxide assay; reagent and sample compatibility must be validated |
| HPAEC-PAD | Oxidized galactosyl product separated and measured directly | High-performance anion-exchange chromatograph with pulsed amperometric detection | Mechanistic work, enzyme-variant comparisons and product-resolved kinetics | Lower throughput and requires carbohydrate-analysis expertise and chromatography equipment |
Do not treat values from these methods as interchangeable unless they have been calibrated against the same substrate, preparation and unit convention. A coupled method reports peroxide-equivalent formation; HPAEC-PAD reports the product concentration determined by chromatography.
Published 96-well ABTS protocol
The following is a practical starting condition from a 2022 RSC Advances assay for Fgr GalOx. It is not a universal recipe: confirm the linear range and adjust concentrations for your enzyme preparation.
Reagents and equipment
- Transparent 96-well plate suitable for absorbance measurements.
- 50 mM sodium phosphate buffer, pH 7.0.
- ABTS at 0.25 mg mL−1 final concentration.
- HRP at 0.1 mg mL−1 final concentration.
- Galactose or lactose at 0.1–7 mM final concentration, selecting a point or series appropriate to the question.
- Fgr GalOx at 5 nM final concentration unless a different amount is required to keep the trace within the initial-rate range.
- Hydrogen-peroxide standards spanning 1–10 μM.
- Reader configured for 414 nm and temperature control at 23 °C.
The cited protocol uses a 100 μL final volume per well and starts reactions by adding substrate to a premix containing buffer, ABTS, HRP and enzyme. Prepare all solutions in the same buffer system used for the assay, and keep enzyme dilution and reaction timing identical between wells.
Plate layout
Reserve enough wells to distinguish enzyme activity from reagent background and to build the calibration curve.

- Substrate-free blank: buffer, ABTS, HRP and GalOx without galactose or lactose. This checks oxidation caused by the enzyme preparation or contaminants.
- Hydrogen-peroxide standards: 1–10 μM H2O2 in the same reporter mixture, but without GalOx substrate oxidation. Use the same plate, volume and wavelength as the samples.
Use replicate wells when sample is available. Randomizing wells or distributing standards across the plate can help expose edge or timing effects, but the essential controls are the complete reaction, blanks and a standard curve.
Step-by-step procedure
- Equilibrate the plate reader to 23 °C and select 414 nm. If the instrument cannot hold temperature, record the actual plate temperature because GalOx rates are temperature-dependent.
- Prepare a premix containing phosphate buffer, ABTS, HRP and GalOx. Keep the enzyme on the handling schedule appropriate for its preparation and avoid unnecessary delays between dilution and dispensing.
- Dispense the premix into the assigned sample and blank wells so that each will finish at 100 μL.
- Prepare the H2O2 standards in the same final reporter environment. Do not use a standard prepared in a substantially different buffer or volume and assume the response will be identical.
- Start each sample reaction by adding the substrate. Mix consistently, using the same order and mixing time for every well. Record the time at which the first absorbance reading begins.
- Read absorbance at 414 nm at intervals short enough to resolve the early linear portion of each trace. Continue long enough to see whether the signal later bends, but do not use the curved portion for the activity calculation.
- Subtract the corresponding blank slope from each sample slope. Convert the corrected slope with the H2O2 calibration curve, then calculate activity using the enzyme amount present in the well or the amount loaded into the assay.
The first part of every trace must be linear before a slope is used. If the reaction is already curved in the first readings, reduce the enzyme concentration, shorten the read window or lower the substrate/reporter load only after confirming that the change does not alter the assay chemistry.
Cuvette or tube implementation
A 2014 characterization of GalOx from Fusarium oxysporum provides a tube-style alternative. The enzyme is activated with 1 mM CuSO4 for 30 minutes at 25 °C, then assayed with 1 μmol ABTS, 2 U HRP and 100 μmol galactose in 20 mM potassium phosphate, pH 7.0. The absorbance change is recorded at 420 nm at 30 °C for 180 seconds.
That paper defines one unit as the amount of enzyme that oxidizes 2 μmol of ABTS per minute, which it equates to consuming 1 μmol of O2 per minute. This definition is specific to that assay and must not be silently substituted for the RSC definition of one unit as 1 μmol of H2O2 produced per minute.
A second Fusarium protocol uses the same ABTS/HRP principle at 30 °C and examines substrates including d-galactose and lactose. Use the tube format when you need a larger optical path, easier sampling, or a temperature-controlled cuvette rather than plate throughput. Because the optical path and mixing geometry differ from a microplate, establish a new blank, standardization and linearity check.
Copper activation: when and how to test it
Several recombinant GalOx protocols include a copper-activation step. The published F. oxysporum example uses 1 mM CuSO4 for 30 minutes at 25 °C; the related Fusarium sambucinum work also uses copper in its preparation or assay workflow. Copper treatment should match the history of your enzyme rather than being added automatically to every sample.
If the preparation may have lost its copper cofactor, measure an aliquot before activation and a matched aliquot after the documented CuSO4 treatment. Keep the activation time, temperature, copper concentration and subsequent dilution identical between comparisons. Excess copper carried into the reporter reaction can itself affect proteins or redox chemistry, so include the same copper carryover in the appropriate blanks and remove or dilute it only if that change has been validated.

Convert absorbance to GalOx units
1. Determine the initial slope
Fit a straight line to the early linear data for each well and record the slope as ΔA/min. Do not use the largest possible time window simply because it improves the visual fit; once substrate, oxygen, ABTS, HRP or peroxide becomes limiting, the slope no longer represents the initial enzyme rate.
2. Use the peroxide standard curve
Plot absorbance against the known H2O2 concentration for the standards measured under the same plate conditions. If the calibration is linear, use its slope to convert the blank-corrected sample absorbance rate into a peroxide-production rate. In symbolic form:
rate(H2O2, μM/min) = corrected sample slope (ΔA/min) ÷ calibration slope (ΔA per μM H2O2)
Multiply by the assay volume to obtain μmol H2O2 per minute. For a 100 μL assay, convert micromolar per minute to micromoles per minute using the volume in liters:
μmol/min = (μM/min) × 0.000100 L
Apply any dilution factor between the enzyme sample and the well. If you use an extinction coefficient instead of a standard curve, state the coefficient, wavelength, path length and solvent conditions; a plate reader’s effective path length is not automatically 1 cm.

3. Normalize and name the unit
- Specific activity: divide μmol H2O2 per minute by the enzyme mass in milligrams to report U mg−1, provided one unit is explicitly defined as 1 μmol H2O2 min−1.
- Volumetric activity: divide by the enzyme volume loaded to report U mL−1 under the same unit definition.
- Amount-based activity: report μmol H2O2 min−1 per assay when enzyme mass or concentration is unavailable or deliberately not normalized.
Always write the unit convention beside the result. The RSC method uses 1 U = 1 μmol H2O2 produced per minute. The 2014 F. oxysporum paper instead defines 1 U through oxidation of 2 μmol ABTS per minute, equivalent in that paper to 1 μmol O2 consumed per minute. Those labels look similar but are not comparable without reconciling the definitions and assay conditions.
Controls that make the rate believable
Blank correction
A rising enzyme-free blank means the reporter system is producing signal without GalOx. Possible causes include non-enzymatic ABTS oxidation, peroxide contamination, oxidizing impurities in the substrate or buffer, and reagent deterioration. Subtract the blank slope only when the blank is stable and behaves linearly; a large or erratic blank is a reason to remake reagents and investigate rather than conceal the problem mathematically.

Substrate dependence
Run more than one substrate concentration when you need kinetic interpretation. The RSC example spans 0.1–7 mM galactose or lactose, but those values are a starting range, not a guarantee that the enzyme is saturated or that every preparation remains linear across it. Record the exact substrate identity, concentration and purity.
Linearity and enzyme loading
At least two enzyme dilutions are useful for checking that the measured initial rate scales with enzyme amount. A curved trace at every dilution usually indicates a reagent, oxygen or mixing problem; a linear trace only at the lowest concentration indicates that the higher load is outside the assay range.
Do these 3 things before closing this tab:
1Repair Windows errors before they cause bigger problems2Scan for outdated or missing drivers - takes under a minute3Clear out junk files and repair common Windows errorsTemperature and timing
Report whether the assay ran at 23 °C, 25 °C or 30 °C and whether the plate was pre-equilibrated. Start timing at substrate addition, not when a batch of wells has been read. Staggered addition without a matching time correction can create an apparent activity difference between wells.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone.Amplex Red/HRP as a peroxide readout
Amplex Red provides another indirect route: GalOx-generated H2O2 is converted by HRP into resorufin. The Thermo Fisher A22179 manual lists approximate resorufin maxima of 571 nm for absorbance and 585 nm for fluorescence. Choose the detection mode supported by your reader and build a calibration curve in the same GalOx reaction matrix.
Use the same logic as for ABTS: include enzyme-free and substrate-free controls, establish the linear initial-rate interval, and test whether the sample or buffer quenches fluorescence or contributes background. Amplex Red can be useful when the plate reader has better fluorescence performance than absorbance sensitivity, but it does not turn the assay into a direct measurement of the oxidized carbohydrate.
Direct product measurement with HPAEC-PAD
High-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD) separates and quantifies the oxidized galactosyl product itself. The 2022 RSC method uses this approach for direct product analysis, making it valuable when the peroxide reporter is unstable, when sample components interfere with HRP or ABTS, or when different enzyme variants may alter peroxide coupling behavior.
Free tools Windows power users keep installed
One-click scans. No signup required.
For a direct-product experiment, collect time-point samples under conditions that stop or sufficiently slow the enzyme reaction, then resolve the carbohydrate products chromatographically against appropriate standards. The instrument, column chemistry, eluent program, electrode condition and product standard determine the quantitative result, so an HPAEC-PAD value should not be presented as though it were an ABTS-derived peroxide unit.
Best Value

- 4 per pack Features Clog Resistant Marker Tip Writes through Dusty, Wet and Oily Surfaces Durable Marker Tip for Writing on Concrete, OSB and Rough Surfaces
- Clog resistant tip writes on dusty, wet and oily surfaces and is optimized for rough surfaces such as OSB, cinderblock and concrete
- Hard hat clip- attaches for easy access

The trade-off is practical: HPAEC-PAD requires specialized hardware, method development and carbohydrate-analysis experience, and it generally supports fewer samples per hour than a microplate reader. Its strength is specificity rather than convenience.
Troubleshooting decision guide
The blank rises
Inspect ABTS, HRP, peroxide standards, substrate and buffer for contamination or oxidation. Prepare fresh reporter reagents, verify that the substrate-free and enzyme-free controls are included, and subtract a blank only after confirming that its slope is reproducible.
The trace curves immediately
Use a lower GalOx concentration, shorten the read interval and verify mixing. A high initial rate can consume a limiting reagent or leave the calibrated range before the reader collects enough points.
The trace is flat
Check that substrate was actually added, that the wavelength is correct, that HRP and ABTS were not omitted, and that the enzyme dilution is active. If copper activation is part of the preparation history, compare matched activated and non-activated aliquots rather than assuming inactivity is intrinsic to the protein.
Replicates disagree
Look first for timing differences, incomplete mixing, edge evaporation, bubbles and inaccurate low-volume pipetting. Use a consistent addition order and discard wells with visible bubbles or optical artifacts according to a predeclared rule.
ABTS and direct product results disagree
Check peroxide stability, HRP competence, ABTS background and matrix inhibition. The two methods measure different points in the reaction chain: ABTS is a peroxide-coupled proxy, whereas HPAEC-PAD quantifies the oxidized carbohydrate. A disagreement is a reason to inspect coupling chemistry, not to force one result to match the other.
What to report so another lab can reproduce the assay
- GalOx source, construct or preparation identifier, concentration and whether it was copper-activated.
- Substrate identity and concentration; for mixed substrates, state the exact composition.
- Buffer identity, concentration and pH.
- ABTS, HRP and any Amplex Red concentrations, including supplier-independent chemical names where possible.
- Final reaction volume, plate or cuvette type, optical path treatment and instrument model class.
- Temperature, wavelength or fluorescence settings, read interval and total monitoring time.
- Blank composition, standard concentrations and the calibration method.
- The time interval used for the initial-rate fit and the rule used for excluding curved traces.
- The unit definition, normalization basis and any dilution or copper carryover correction.
These details prevent a numerical activity value from being mistaken for a universal GalOx constant. They also allow a reader to distinguish a genuine enzyme difference from a change in reporter chemistry, temperature, substrate loading or unit convention.
