Two distributors may be selling material made at the same plant, from the same critical starting material, or over the same transport route. A defensible dependency map therefore follows each material from upstream inputs to the finished medicine, records the actual manufacturer and site behind every intermediary, and tests whether an alternative is independently qualified and usable for the product and market in scope.
This guide presents a practical mapping method grounded in FDA supply-chain and API-good-manufacturing guidance. It separates regulatory record requirements from additional risk-analysis techniques, so a supplier list does not get mistaken for proof of resilience.

Define the dependency before you count suppliers
A single-source dependency exists when a defined product or critical process has only one independently usable source at the relevant tier. State the scope explicitly: it might be one ingredient, one manufacturing site, one finished drug, or one market. “Independently usable” means more than a second name in a purchasing system; the source must be technically acceptable, qualified for the intended use, available within the required lead time, and permissible under the applicable regulatory change process.
Use actual manufacturers as the basic unit of count. A broker, trader, or distributor can provide access to material without making it. FDA’s Q7A API GMP guidance says that when a supplier of a critical material is not its manufacturer, the manufacturer’s name and address should be known. The same guidance calls for traceability through distributors and intermediaries, including transportation and distribution records.

Keep three concepts separate in your records:
- Commercial source: the company from which you purchase or receive the material.
- Actual manufacturer: the organization and site that physically makes the material or performs the relevant processing step.
- Independent usable source: an actual manufacturer and supply path that can be qualified and used for the product, process, and market under consideration.
A supplier count that includes several commercial sources but only one actual plant is a single-source dependency, even if invoices show multiple vendors.
Map every tier from starting material to finished dose
The map should be a linked graph of materials, organizations, sites, products, and logistics—not a flat vendor spreadsheet. The U.S. Government Publishing Office’s Mapping of the Supply Chain describes the progression from raw materials and intermediates to active pharmaceutical ingredient (API) and finished dosage form. Extend that chain far enough upstream and downstream to expose common dependencies.
Raw materials and critical starting materials
Begin with solvents, reagents, chemicals, and regulatory starting materials that enter API production. Record the material identity and specification, the supplying organization, and the manufacturing site when known. Mark which inputs are critical to the process, difficult to replace, subject to long lead times, or sourced from a concentrated region or facility.

Intermediates
List each critical intermediate separately from the API. An intermediate is material undergoing further molecular change or purification before becoming the API; it is not simply another name for the drug substance. Capture the producer, site, process step, batch or lot references, and upstream inputs. A second API supplier can still depend on the same intermediate producer, so this tier is essential for finding hidden concentration.
API or drug-substance manufacture
Connect each API to its actual manufacturing organization and site, then link the API to every finished product that uses it. Include process version, approved site information, batch history, capacity or allocation constraints where available, and the commercial distributors used to move the material. Keep the distributor relationship separate from the manufacturing relationship.
Finished dosage form
Map the site that combines the API with inactive ingredients and makes tablets, capsules, injectables, or other finished forms. Include critical excipients, sterilization or fill-finish steps, and any single-use or specialist process dependency. A finished-dose source may be constrained even when the API has two manufacturers.

Packaging and container closure
Include critical containers, closures, coatings, and other packaging components when they protect product quality or are required for release. Packaging is part of the practical supply chain: a medicine cannot ship if a qualified vial, stopper, blister component, or specialized closure is unavailable.
Distribution and transport
Record each distributor, intermediary, warehouse, route, handoff, and relevant transport condition. FDA Q7A calls for records that identify API distributors and intermediaries, the original manufacturer, and transportation or distribution information. Treat a route, port, specialized carrier, or temperature-controlled lane as a dependency when an interruption would block delivery.
Use a record structure that preserves traceability
Every node and relationship should be identifiable and time-stamped. The following compact schema works as a minimum dependency register; add product-specific fields required by your quality system.
| Field | What to capture | Why it matters |
|---|---|---|
| Material and tier | Material name, internal code, specification, and tier (starting material, intermediate, API, excipient, packaging, or finished dose) | Prevents an intermediate, API, and finished product from being counted as the same source |
| Commercial supplier | Supplier name, control or identification number, and contract role | Shows who sells or ships the material without assuming that party manufactures it |
| Actual manufacturer | Legal manufacturing organization and address | Provides the independent-source unit required for concentration analysis |
| Manufacturing site | Specific plant, building or campus, country or region, and relevant process | Reveals supposedly different suppliers sharing one site |
| Upstream dependencies | Critical starting materials, intermediates, utilities, specialist processes, and shared parent or production network where known | Exposes common-cause failures above the named supplier |
| Downstream use | API, finished drug, market, dosage form, and packaging configuration that consumes the material | Defines the scope and patient or operational consequence of a disruption |
| Batch and receipt evidence | Batch or lot, quantity, receipt number and date, test or release results, and material-use trace | Supports reconciliation from a shipment to the product batches that used it |
| Qualification status | Approved, conditionally qualified, under evaluation, or not qualified; include required studies and approval path | Distinguishes a usable alternative from a merely possible source |
| Evidence date and owner | Date confirmed, evidence type, next review date, and mitigation owner | Keeps the map current and assigns action when a gap is found |
| Logistics path | Distributor, warehouse, route, carrier, lead time, and temperature or handling requirements | Identifies transport and handoff dependencies that a manufacturer-only map misses |
FDA Q7A specifically identifies manufacturer, identity and quantity for each shipment or batch, supplier name and control number when known, receipt number and date, test results, and records tracing how material was used. Retain those records in the quality and batch systems that can be reconciled, not only in a procurement database.
Build the map step by step
- Set the scope. Name the finished medicine or process, dosage form, market, manufacturing site, and time period. Decide whether the question is continuity of API, finished dose, packaging, or the complete product.
- Inventory consumed materials. Extract approved bills of materials, master production records, specifications, and critical-process lists. Include APIs, intermediates, starting materials, excipients, solvents, reagents, and critical container-closure components.
- Join purchasing to quality records. For every material, reconcile purchase-order suppliers with approved-supplier files, receiving records, certificates, test results, batch records, and supplier-audit reports. Do not accept a distributor name as the manufacturer without confirmation.
- Verify the actual site. Obtain the manufacturer’s legal name, address, site identifier, process performed, and the date on which the information was confirmed. Normalize aliases, former legal entities, spelling variations, and site names so one plant cannot appear as several nodes.
- Trace upstream. Ask each actual manufacturer for the critical starting materials and intermediates that could constrain supply. Record shared sources, specialist steps, utilities, and production networks where the information is available and contractually shareable.
- Trace downstream and laterally. Link each API or component to every finished product and market that consumes it. Add distributors, warehouses, transport lanes, temperature controls, and alternate routes.
- Attach evidence and status. Store batch or lot references, receipt dates, test or release evidence, supplier confirmations, audit findings, qualification reports, and regulatory status. Mark each alternative as qualified, conditionally qualified, under evaluation, or hypothetical.
- Run an independence test. Count actual manufacturers, then compare their sites, parent or production networks, upstream materials, specialist processes, utilities, and logistics. A source is independent only to the degree relevant to the failure scenario being assessed.
- Assign an owner and review date. Every unresolved dependency needs a quality, operations, procurement, regulatory, or supply-chain owner and a dated action. A graph without ownership becomes an obsolete directory.
Find hidden single-source dependencies
Compare actual manufacturers, not vendor names
Group all commercial suppliers by verified actual manufacturer and site. If three distributors point to one plant, the product has one manufacturing source. If two legal entities share a campus or production line, treat them as potentially common-cause sources until the relevant process and failure modes have been separated.

Test geography and route concentration
Record site and route geography, but do not use country of origin as a proxy for resilience. Two plants in different countries can share one upstream chemical, port, specialist carrier, or temperature-controlled lane. Conversely, two sites in one region may have genuinely separate inputs and routes. Document the specific dependency and the evidence supporting it.
Separate possible from usable alternatives
A catalog listing, historical supplier, or unapproved plant is not an operational alternative. Record qualification state, product-specific fit, lead time, capacity, minimum order, testing needs, and regulatory constraints. Only count a source as independent for continuity planning when it can supply the defined product and market within the required window.
Rank the risk after the graph is built
Use a risk register alongside the map. The dimensions below combine FDA-identified disruption causes with practical continuity questions; they are decision axes, not a regulatory scoring formula.

| Risk dimension | Questions to answer | Evidence to retain |
|---|---|---|
| Independent qualified sources | How many actual manufacturers can supply this exact material and product? | Approved-source list, site confirmation, qualification decision, and capacity statement |
| Substitutability | Can the material or process be replaced, and how long do studies, validation, and approval take? | Technical assessment, validation plan, change-control record, and regulatory determination |
| Site and upstream concentration | Do alternatives share a plant, critical input, utility, specialist process, or parent network? | Site map, supplier questionnaire, audit findings, and upstream declarations |

| Lead time and logistics | How much inventory coverage exists, and which routes, carriers, ports, or handling conditions can fail? | Lead-time history, inventory days, route map, carrier qualification, and contingency lanes |
| Demand and patient impact | How critical is the medicine, how volatile is demand, and what happens if supply stops? | Demand scenarios, service-level target, clinical criticality assessment, and allocation plan |
| Regulatory recovery | What notification, approval, supplement, or change-control work is needed to add or switch a source? | Regulatory assessment, submission plan, approvals, and implementation timeline |
FDA lists manufacturing and quality problems, economic conditions, increased demand, single or limited suppliers, foreign dependence, weather and environmental events, public-health responses, transportation, and geopolitical situations among possible disruption causes. The agency says shortage response depends on the cause, patient-care impact, and public-health risk, so a risk score should connect dependency exposure to consequence rather than treating every single source as equally urgent.
Qualify an alternate source and control the change
Adding a name to the supplier master does not make the source usable. The alternate must pass the technical, quality, manufacturing, and regulatory checks applicable to the material and product. Evaluate identity and specifications, impurity profile, process capability, analytical methods, stability or compatibility, packaging, transport conditions, minimum quantities, and lead time. The exact studies depend on the material and dosage form; document the rationale instead of assuming that a prior approval transfers automatically.
FDA notes that new sources of raw materials, regulatory starting materials, or active pharmaceutical materials may need FDA approval to increase supply. The change pathway depends on the product, filing, market, and proposed modification. Confirm the requirement with the responsible regulatory organization before promising that an alternate can be activated.

FDA’s Q7A guidance states: Changing the source of supply of critical raw materials should be treated according to Section 13, Change Control.
Use that principle to open a formal change record, define affected products and markets, assess validation and stability work, obtain quality approval, and complete required regulatory actions before routine use.
Use public shortage data as a signal, not as your supplier map
The FDA’s openFDA Drug Shortages data and the agency’s shortage pages can reveal that a product is constrained, but they do not disclose every confidential supplier relationship, upstream source, approved site, current capacity, or product-specific qualification status. FDA says manufacturers provide most shortage information and that its database reflects situations known to the agency. Reconcile public signals with purchasing, batch, receiving, supplier-audit, quality, and direct supplier-confirmation records before declaring a dependency.
FDA also states in Supply Chain: FDA’s Role that it cannot ensure continuity or resilience of entire industrial supply chains and cannot require companies to produce products or report information that the law does not require. Public absence of a shortage notice therefore does not prove that a source is independent or that capacity is available.
Interpret shortage statistics in their original context
The FDA Drug Shortage Task Force’s Drug Shortages: Root Causes and Potential Solutions report identifies three broad root causes: weak incentives to produce less-profitable medicines, insufficient market reward for mature quality systems, and logistical or regulatory challenges that make recovery difficult.
In a February 2020 recalculation published on the FDA report page, 42 percent of drugs that had been in shortage between 2013 and 2017 experienced a significant production increase. FDA defined that increase as new suppliers entering and/or existing suppliers increasing output enough to restore at least half of the unavailable quantity. The same recalculation found that 30 percent had their quantity fully restored to pre-shortage levels. These are retrospective figures about those shortage drugs, not the success rate of a supplier-mapping program or a promise that a second source will restore supply.

Connect systems
Link enterprise-resource-planning purchase and receipt data to the approved-supplier and quality systems, then connect those records to master production and batch records. Use stable identifiers for material, site, product, and batch so a manufacturer change can be traced to affected inventory and finished doses.
Escalate unresolved concentration
When the graph shows one independent source, name the mitigation owner and choose an action: qualify a second manufacturer, secure additional inventory, validate another route, negotiate capacity, redesign the process, or accept the exposure with documented rationale. The action should match the failure mode; buying from another broker does not mitigate a shared manufacturing plant.
Keep evidence proportional but auditable
Some upstream details may be confidential or unavailable. Mark them as unknown, state the date and source of the last confirmation, and identify the consequence of that uncertainty. Do not convert an unverified assumption into an “independent” node. The map is decision-grade when a reviewer can follow a material from receipt through use and see why each source was counted.
Quick wins for a faster PC:
Repair Windows errors before they cause bigger problemsFix Now →Scan for outdated or missing drivers - takes under a minuteDriver Scan →A compact example of the independence test
Suppose a finished tablet lists two commercial API suppliers. The map shows Supplier A buys API from Plant North and Supplier B buys from Plant South. That appears to be two sources until upstream records show that both plants purchase the same critical intermediate from one manufacturer. The API tier has two plants, but the intermediate tier has one independent source. If the intermediate is essential and cannot be replaced within the product’s regulatory and lead-time constraints, the tablet retains a single-source dependency at that tier.
A different case may show two API plants with separate intermediates but one temperature-controlled shipping lane. The manufacturing sources are independent for a plant-failure scenario, yet logistics remain concentrated. Record both conclusions rather than forcing the whole product into a single “safe” or “unsafe” label.
What a defensible conclusion looks like
State the result in scope-specific language: “For Product X in Market Y, there is one independently qualified manufacturer of Intermediate Z as of the evidence date; two commercial distributors do not add independence. The principal exposure is the shared plant, and the alternate-source change would require technical qualification and regulatory review.” This wording identifies the material, tier, market, evidence date, and action without claiming more certainty than the records support.
The central discipline is simple: follow the material, verify the maker and site, test common upstream and logistics dependencies, and count only sources that are qualified and usable. That approach exposes concentration that a vendor list hides while keeping regulatory obligations, operational analysis, and evidence limits distinct.
