You can focus light through a strongly scattering, visually opaque material by measuring or optimizing how the material scrambles light, then adjusting the incoming wavefront so the transmitted light interferes constructively at a chosen target. This does not make the material transparent: it redirects some light that gets through. The method needs detectable light along the target path and a measurement or feedback signal to find a useful correction.
Why a focus can form behind an opaque-looking material
A scattering layer redirects coherent light into a complicated pattern of bright and dark speckles. Although the original beam is scrambled, some light can still emerge. If the phases of its contributions are controlled, they can arrive in step at one selected point and add together there, forming a focus.
Here, “opaque” means strongly scattering or visually opaque—not a barrier that absorbs or blocks every photon. Wavefront shaping cannot recover light that never reaches the target or cannot be detected. It also does not create an arbitrary image with unlimited contrast: the result depends on the number of controlled optical modes, the target field of view, measurement quality, and how stable the sample remains. Three ways to find a wavefront that focuses

These approaches share the goal of learning a correction, but differ in what they measure: a scalar target signal, a returned complex field, or many input-output field pairs. A phase-conjugate solution can be understood as a matched-field approach; a matrix pseudoinverse can additionally allocate degrees of freedom to reduce selected background channels.

What a guide star does
A guide star is a signal source at, or associated with, the location where a focus is wanted. Its light or another target-linked signal gives the system information about which input correction will deliver light to that location. Without a guide star, target feedback, or a measured transmission matrix, simply putting a spatial light modulator (SLM) in the beam does not reveal the needed correction.
One proposed route uses ultrasound to encode light from an acoustic focus, providing a virtual guide star for optimization. In a 2013 preprint, Jian Wei Tay, Puxiang Lai, Yuta Suzuki, and Lihong V. Wang reported that light intensity at the acoustic focus increased by an order of magnitude in their experiment. That is a result from their particular setup, not a general enhancement guarantee: Digital ultrasonically encoded (DUE) optical focusing into random media (2013 preprint).

Transmission-matrix focusing: a representative lab workflow
A transmission-matrix experiment measures how controlled input patterns map to output optical fields. The resulting matrix can then be used to calculate a pattern for a chosen target. A typical sequence is:
- Define the optical conditions. Choose coherent illumination, the sample, wavelength, controlled input region, and target output region.
- Set up wavefront control. Relay the input beam onto the sample using a device such as an SLM.
- Measure calibration fields. Display a series of known input patterns and measure the output complex field for each, typically using a camera with a phase reference and an interferometric or holographic arrangement.
- Assemble the matrix. Store the measured relation between each input pattern and the output field as the transmission matrix.
- Calculate the target input. Use a conjugate-field solution or a matrix pseudoinverse, depending on whether the goal is a focus alone or a focus with selected background suppression.
- Apply and evaluate the correction. Display the calculated pattern, send it through the sample, and measure the focus and background.
- Recalibrate when needed. If the sample or optical setup changes enough to decorrelate the measured matrix, the old correction may no longer produce the intended result.
For scale, one 2017 ground-glass demonstration used 532 nm illumination from a 150 mW diode-pumped laser, a phase-only HOLOEYE PLUTO SLM, four-step phase-shifting holography, and 128 Hadamard input patterns to record its matrix. These are details of that study’s apparatus, not a universal build specification or a complete laser-safety guide: Focusing light through scattering media by transmission matrix inversion (2017).

What laboratory studies have demonstrated
Reported focus quality and sample thickness vary by experiment. The figures below are individual study outcomes, not pooled estimates or normal performance expectations.

These demonstrations establish that focusing through particular scattering samples is possible under experimental conditions. They do not establish a universal focus gain, a general tissue penetration depth, or clinical efficacy.

What equipment and calibration a setup needs
The exact apparatus depends on whether the experiment uses feedback optimization, phase conjugation, or a transmission matrix. In most laboratory implementations, the core needs are:
- Coherent illumination at a wavelength appropriate to the sample and optical components.
- Wavefront control, often an SLM whose wavelength range, polarization response, aperture, and update behavior suit the experiment.
- Detection and measurement matched to the chosen method—for example, a camera and phase-reference arrangement for complex-field measurements, or a detector for target-linked feedback.
- Optical relays and references where required to illuminate the sample and record or reconstruct the relevant field.
- A calibration method that measures the current sample and optical system, not a correction borrowed from a different configuration.
Laser power, optical alignment, detector sensitivity, measurement time, and modulation constraints must be selected for the particular experiment. The example apparatus above should not be treated as a complete specification or safety instruction.

How to choose an approach—and what can make it fail
The central design trade-off is how much information the system measures versus how quickly it can act on it. A moving or changing sample can decorrelate a calibration before the corrected wavefront is used, so latency and stability matter as much as the calculation.
- Consider feedback optimization when a usable target-linked signal is available and iterative adjustment is practical.
- Consider OPC/DOPC when a guide-star field can be recorded and the optical system can return its conjugate field with adequate alignment and speed.
- Consider a transmission matrix when input-output fields can be measured and the desired output can be specified computationally; use a pseudoinverse only with awareness that background suppression consumes available degrees of freedom.
- Check calibration quality. In the 2017 TM inversion study, the authors emphasized that accurately measuring the transmission matrix is very important. They also identify air turbulence, photon shot noise, and mechanical drift as sources that reduce performance. Study
- Check whether the correction is still valid. Sample motion or changes in the optical path can invalidate a previously useful wavefront or matrix.
- Set realistic output goals. The controllable modes, field of view, measurement noise, and phase-only hardware limit achievable focus contrast and background suppression.
Biological-tissue demonstrations are optical research results, not proof that a procedure is routinely safe, noninvasive, or clinically effective. The implementation determines what signal must be introduced or detected and what safety constraints apply.
