How to Choose Batch or Flow for Each Automated Chemistry Step

Some synthetic steps are most practical in a flask, while others benefit from a continuously moving stream. A 2016 Cambridge platform was built around that fact: it let one multistep procedure assign batch or flow operation to each step, and it incorporated workup tasks rather than treating them as a separate manual phase. The demonstration converted 2-hexanone into 5-methyl-4-propylthiophene-2-carboxylic acid.

This was a specific proof-of-concept report, not evidence that every reaction should move to flow, that the apparatus is a current commercial product, or that it outperforms batch-only and flow-only systems in every setting.

What “batch and flow” means in this context

Batch chemistry performs a reaction in a vessel: reagents are charged to a flask, allowed to react under controlled conditions, and then processed before the vessel is emptied. Flow chemistry sends reactants through a channel or coil at a controlled rate. Material enters, reacts as it travels, and exits for the next operation.

The Cambridge concept does not treat those approaches as competing identities. It treats them as selectable modes within one synthesis. A chemist can keep a step in a round-bottom flask when that is the sensible operation, switch to a flow segment for another step, and then carry out separation or solvent changes in the same connected sequence. The point is orchestration, not forcing every reaction into a single reactor type.

That distinction matters because a multistep route is more than its named reactions. Transfers between vessels, quenching, solvent replacement and other workup operations can consume time and create opportunities for handling errors. The platform described by Steve Ley and Daniel Fitzpatrick at the University of Cambridge was intended to coordinate those operations with reaction steps. The developers described the aim as “a more holistic understanding of synthesis practice,” so that moving from discovery toward larger-scale processing would involve fewer boundary changes. Chemistry World’s 2016 report attributes that statement to Ley.

How the reported platform was assembled

The apparatus combined three recognizable pieces of laboratory equipment with automation around them. It was based on a commercially available modular flow-chemistry reactor, but the report did not identify a manufacturer or model. That omission is important: the publication demonstrates a configuration and control idea, not a named product that can be ordered from the information supplied.

Element Function in the hybrid sequence What the report establishes
Round-bottom flask Provides a batch vessel for a step that the chemist chooses to run as a conventional charge-and-react operation. The system description explicitly includes one; the report does not specify its size or a universal recipe for when to use it.
Flow coil Provides a continuously moving reaction zone for a selected step. The system includes a flow coil; the report does not give a single residence-time, diameter or throughput value that applies to all sequences.
Distillation column Handles a separation or solvent-management operation within the connected process. A distillation column is part of the described assembly; the publication does not present a general performance specification for it.
Control and switching hardware Coordinates transfers and the order of batch, flow and workup operations. The sequence was described as automatable and capable of running as one continuous phase after setup.

In practical terms, the combination gives the operator more than a choice between two isolated machines. It provides a route-level control system in which the next operation can receive material from either a flask or a flow outlet, with separation equipment available in the same chain.

The demonstration: from 2-hexanone to a thiophene acid

The team used the platform to prepare 5-methyl-4-propylthiophene-2-carboxylic acid from 2-hexanone. The reported route contained several chemical steps and workup tasks, including changes of solvent. Those details make the example significant: it tested integration across a sequence rather than showing a single reaction in a flow tube.

  1. Start with the feedstock. 2-Hexanone was the starting material for the multistep route.
  2. Assign each operation to a suitable mode. The platform allowed a selected reaction to take place in the round-bottom flask or in the flow coil, rather than prescribing one mode for the entire route.
  3. Carry intermediate material forward. Connections and controls moved material to the next operation, reducing the need for a researcher to perform every transfer manually.
  4. Include workup in the sequence. Solvent changes and other processing tasks were part of the demonstrated procedure, with the distillation column included in the equipment arrangement.
  5. Collect the final product. After setup, the sequence was reported to run as one continuous phase under the management of one researcher.

The source does not provide a general recipe that can be copied for unrelated substrates, nor does it report a yield table, throughput figure or quantified labor saving for this demonstration. The defensible conclusion is narrower: a hybrid, automated arrangement was shown to carry a named multistep synthesis through reaction and workup operations.

Operating envelope and what “automated” means here

The reported temperature range was −70 °C to +150 °C, as reported by Chemistry World in 2016. The article also says that the system can operate at high pressure, but it gives no numerical pressure rating. “High pressure” should therefore not be converted into a bar or psi value.

Automation in this report refers to controlling the combined sequence after the setup has been prepared. One researcher could manage the continuous run, which the article presents as a way to reduce time-consuming manual laboratory work. It does not quantify minutes saved, staffing reductions, operating cost or unattended runtime. Those measures would require separate experiments and are not established by the report.

The temperature span and pressure statement describe the reported platform’s stated operating envelope, not a guarantee for every reaction. A particular reagent, solvent, seal, pump or workup may impose a tighter limit. Likewise, the existence of a flow coil does not make every chemistry suitable for flow; the report explicitly frames reaction amenability as a selection criterion.

Choosing batch or flow for each step

The useful decision is made step by step. A route can contain a batch operation, a flow operation and a separation operation without becoming conceptually inconsistent.

Decision question Why it matters Possible choice in the hybrid approach
Is the reaction amenable to continuous flow? Some reactions or mixtures are not suitable for passage through a coil or associated equipment. Keep that step in the batch flask and reserve flow for compatible steps.
How many transfers and workups are required? Frequent manual moves add handling time and can interrupt a sequence. Connect the operation to the automated chain, using the distillation column or other configured workup hardware where appropriate.
Does the step benefit from controlled continuous operation? A continuously managed stream may fit a step that needs stable operating conditions and ordered handoff to the next stage. Use the flow coil for that stage, while retaining batch elsewhere.
What setup and monitoring are available? Hybrid operation requires configuration, compatible connections and a control strategy before the run starts. Choose the simplest mode that meets the chemistry and the available automation.

This framework avoids two common mistakes. The first is assuming that flow is automatically superior because it is newer or more automated. The second is assuming that a batch vessel should remain the default for every stage because the route began there. The Cambridge platform’s rationale is precisely that different steps can call for different modes.

What the 2016 report proves—and what it does not

The evidence supports a defined set of claims:

  • Batch and flow can be combined within one automated multistep synthesis.
  • The demonstrated assembly included a round-bottom flask, a flow coil and a distillation column.
  • The route converted 2-hexanone to 5-methyl-4-propylthiophene-2-carboxylic acid and included solvent changes and other workup tasks.
  • The stated temperature range was −70 °C to +150 °C, and high-pressure operation was mentioned without a numerical rating.
  • After setup, one researcher was reported to be able to manage the sequence as one continuous phase.

It does not establish a universal process recipe, a preferred mode for every reaction, a pressure specification, a yield advantage, a throughput benchmark, a cost reduction or a statistically measured time saving. Nor does it establish broad adoption, current product availability or superiority over a carefully run batch-only or flow-only process. The underlying paper is identified as D. E. Fitzpatrick and S. How a laboratory could apply the integration idea

The report is a demonstration, not a universal installation manual. A laboratory evaluating the same strategy would need to make its own chemistry and safety decisions. A sensible planning sequence is:

  1. Map the complete route. List reactions, quench steps, solvent replacements, separations and collection points, not only the named transformations.
  2. Classify each operation. Mark which steps are compatible with continuous flow, which need a conventional vessel and which require a dedicated workup.
  3. Define interfaces. Specify how material leaves the flask, enters the coil, passes through separation equipment and is collected. Compatibility of solvents, pressure, temperature and materials must be checked for the actual setup.
  4. Set the control sequence. Establish valve states, pump or transfer actions, timing and monitoring before introducing valuable substrate. The Cambridge report demonstrates automation, but it does not supply a universal control program.
  5. Test transitions with appropriate safeguards. Verify that an intermediate can be transferred and that the next module reaches its intended condition before committing to a full multistep run.
  6. Record performance for the specific route. Measure yield, selectivity, material balance, operator interventions, cycle time and waste if those metrics are needed for a process decision. None of those figures can be inferred from the 2016 demonstration.

This route-first approach keeps automation in service of chemistry. It also makes clear where a hybrid system adds complexity: every connection, control action and workup module becomes part of the process that must be checked.

Why experts viewed the idea as a change in mindset

The platform challenged the assumption that batch and flow represent mutually exclusive synthetic philosophies. Andreas Kirschning of Leibniz University of Hannover, identified in the Chemistry World account as a microreactor technology expert, said: “What is usually regarded to represent two distinct synthetic philosophies has been turned upside down here.” The quoted comment appears in the 2016 report.

Thomas Wirth of Cardiff University offered a forward-looking assessment: “The control of combined multistep flow/batch sequences by an automated platform will surely be taken up by other researchers enabling novel, advanced target synthesis.” That is an expert prediction, not a measurement of subsequent adoption.

Ley’s own closing qualification is equally important: “As with all new things, the system will have to evolve to increase versatility and autonomy, but with the availability of new microprocessor control chips and the inventive spirit of the chemist, this will happen.” The statement describes a development direction—greater versatility and autonomy—rather than claiming that the 2016 apparatus had already reached those endpoints.

Is this a product you can buy now?

The available account says only that the apparatus was based on a commercially available modular flow-chemistry reactor. It does not name the supplier, model, price, current software, support arrangement or present-day availability. Consequently, the report cannot support a buying recommendation or a claim that an identical Cambridge system is on the market today.

It is also a mistake to read “commercially available module” as “complete turnkey platform.” The demonstrated arrangement added a flask, coil, distillation column and control scheme around a modular reactor. Reproducing that capability would require matching equipment, integration work and a route-specific validation plan.

The practical takeaway

The lasting lesson of the Cambridge report is architectural: choose the reaction mode that fits each step, then connect those choices into one controlled sequence. Batch remains useful where a vessel and manual-style operation are appropriate; flow remains useful where continuous handling fits; workup belongs in the process design rather than being ignored. The 2016 demonstration shows that these choices can coexist in an automated route, while leaving performance claims, broad applicability and current commercial status open for separate verification.
