One free scan finds every outdated or missing driver and matches the right update for your exact hardware.Free scan · exact hardware matchChoose the material from the holder’s job outward, not from the filament label. A dry carrier that positions a slide or grid can prioritize stiffness, flatness and dimensional stability; a chamber in the optical path must also control autofluorescence and light scattering; a part touching culture medium or a specimen adds cleaning, extractables and sterilization questions; and a liquid chamber or glass-bonding surface demands leak-tight, repeatable surfaces.
For many FDM prototypes and external carriers, PLA is a practical starting point and PETG is a reasonable alternative when you need more durability. Neither is automatically optically inert or suitable for every biological workflow. If fluorescence matters, measure the exact printed color and formulation in your microscope. If a chamber must contain liquid reliably or bond to glass, a microscopy design study found FDM inadequate for its requirements and moved those chambers to MSLA resin while retaining an FDM carrier. Treat that as a design precedent, not a blanket approval for every resin.

External carrier or positioning fixture
An external carrier supports a slide, grid, dish or insert without placing printed plastic in the imaging field or liquid. Geometry, stiffness, flatness, objective clearance and repeatable interfaces dominate the decision. PLA is often the simplest FDM material for an initial design; PETG is another accessible candidate when the carrier will see more handling or needs greater toughness. If the carrier is outside the optical path and never contacts a sample, fluorescence and extractables are lower-priority risks, although they still matter if stray light or cleaning is involved.

Optical-path component
A window surround, spacer, aperture, grid frame or other part close to the specimen can contribute background light even when it does not touch liquid. Material color, additives, surface finish, distance from the specimen and the microscope’s excitation and emission bands all influence the result. A filament that looks opaque under room light can still emit under illumination. Select only after a blank-part check under the intended settings.
Liquid-containing chamber
A chamber has to hold the intended liquid for the full experiment without leaking, swelling, wicking through layer lines or losing its bond to glass. Its walls, ports, gasket lands and bonding surfaces require tighter process control than a dry carrier. Layer adhesion and surface defects become functional defects, not cosmetic imperfections.
Sample- or culture-contact surface
When printed plastic touches tissue, cells, culture medium or a specimen grid, the exact formulation and post-processing become part of the protocol. A generic “biocompatible” label does not establish suitability for your cell type, exposure time, cleaning agent or sterilization method. Validate the printed part, not just the material family.

What the common material choices can and cannot tell you
| Material/process | Useful starting role | Important unresolved checks | Evidence boundary |
|---|---|---|---|
| FDM PLA | Fast prototypes, dry carriers and low-complexity fixtures | Autofluorescence, layer-line leakage, heat exposure, cleaning and sample contact | Microscopy guidance identifies PLA as a simple starting material; suitability remains application-specific. |
| FDM PETG | External carriers or parts needing more durability than a basic prototype | Fluorescence under your wavelengths, dimensional behavior, chemical exposure, sterilization and bonding | Microscopy guidance lists PETG with PLA for ease of use; it is not a universal optical or biological certification. |
| MSLA resin | Fine-detail chambers and surfaces where watertightness or glass bonding is critical | Resin formulation, residual monomer, wash and post-cure, fluorescence, extractables and biological compatibility | A 2022 Mod3D study used MSLA resin for its chambers after FDM failed its leak, resolution and bonding requirements. That result applies to its design and workflow. |
| Conductive PLA | Specialized holders for applications such as charge dissipation in electron microscopy | Electrical performance, optical background, surface finish and chemical/biological compatibility | A 2017 SEM study reported fabrication in under 30 minutes and below $0.50 per piece at that time; those figures do not establish current cost or optical-microscopy performance. |
Printing process can matter as much as polymer. FDM builds anisotropic layers and leaves seams and ridges; MSLA can produce finer features and smoother walls but introduces washing, post-curing and resin-handling variables. Compare complete, finished parts rather than assuming that a resin or filament name predicts the outcome.
Control fluorescence before it contaminates an image
Autofluorescence is the most easily missed material failure in an optical holder. A biomedical-imaging study found PLA and PETG emissions that could be similar in intensity to tissue signals under its tested optical conditions. In one comparison, PETG background made skin and bone samples indistinguishable. Those observations do not apply to every grade, pigment, geometry, microscope or wavelength, but they are strong evidence that “black” or “opaque” is not a sufficient specification.

Run a blank-part measurement
- Print a representative coupon or the actual holder using the final color, layer height, infill, orientation, supports and post-processing.
- Place it at the same distance and angle it will have relative to the specimen and objective.
- Use the exact excitation source, filter set, detector gain, exposure and acquisition mode planned for the experiment.
- Record intensity with no specimen present, then compare it with the weakest biological signal you need to resolve.
- Repeat after the cleaning or sterilization cycle if the part will be reused; residues and surface changes can alter background.
If the blank contributes a structured pattern, bright edges or a broad glow in the field of view, move the part out of the optical path, redesign the geometry, change the material or use a non-printed optical interface. Do not subtract a background blindly when it overlaps the spectrum or spatial detail of the signal you are trying to measure.
Match stiffness, flatness and temperature to the assembly
A holder can fail through drift or misalignment even when it never leaks. Check the load path from the stage to the specimen, the unsupported spans, screw or clip forces, layer orientation and the clearance needed by the objective. FDM parts are direction-dependent: a feature loaded across layer bonds may behave differently from one loaded along the filament paths. Record orientation in the build specification so a replacement part is mechanically comparable.
What the 37 °C result means
The Mod3D optical system used a 4 mm-thick FDM holder with 20% grid infill and reported dimensional stability up to 37 °C. That is a result for that holder design, printer setup and material combination—not a universal temperature rating for PLA or PETG. If your experiment warms the stage, incubates a culture or cycles temperature, verify flatness and critical dimensions at the actual temperature and after the full dwell time.
Design checks that prevent fit failures
- Measure the printed datum surfaces with the same tool and reference points you will use for incoming inspection.
- Provide clearance for the objective nose, condenser, stage clips, tubing and coverslip edges; verify it with the largest accessory in the workflow.
- Keep locating features tied to one controlled datum rather than stacking several printed tolerances.
- Inspect overhangs, elephant-foot distortion and support scars where a grid, slide or glass window seats.
- Test an unloaded holder and a loaded holder for tilt, rocking and movement during stage travel.
Choose a different process when liquid sealing or glass bonding is critical
FDM is attractive because printers and PLA or PETG are accessible, but its layer lines and surface imperfections can defeat a chamber seal. The Mod3D authors wrote that they found FDM “unreliable, with the resolution not up to the standards required, a poor ability of FDM prints to be watertight, and a glass bonding surface with too many potential imperfections to allow reliable bonding.” Their solution was to keep an FDM part for the stiff carrier and fabricate the liquid chambers with MSLA resin.
Use that split as a design pattern: put forgiving, dry structural functions in FDM and reserve a higher-resolution process for the wetted geometry when your validation shows FDM cannot meet it. The study does not prove that every MSLA resin is leak-proof, optically clean or safe for cells. Resin identity, wash chemistry, cure schedule, wall thickness, support placement and bonding method all need their own acceptance tests.

Leak and bond acceptance tests
- Inspect the chamber under magnification for pinholes, uncured residue, support scars and cracks around ports or corners.
- Fill it with the intended liquid, or a compatible non-biological surrogate if your protocol requires one, and hold it for longer than the planned experiment.
- Place absorbent material beneath seams and ports so small leaks become visible; check again after temperature equilibration.
- Apply the same glass-cleaning and bonding procedure used in the experiment, then repeat the fill test after the bond has fully set.
- Cycle the chamber through the planned cleaning and reuse schedule and repeat the test before returning it to biological work.
A seal that passes a short room-temperature check but fails after warming, washing or stage movement is not reliable enough for a live experiment.
Evaluate sample contact and cleaning as one workflow
Published biological-use results are protocol-specific. A cryo-EM grid-holder study reported reuse up to 15 times without observed adverse effects on sample preparation, using its particular designs and procedures. That finding supports the value of a reuse test; it does not certify another filament, resin, geometry or cell system.
Define the exposure
- Identify whether the printed surface touches a specimen directly, contacts only a sealed coverslip, or is exposed to medium through a leak path.
- Record contact time, temperature, agitation and whether the part is single-use or reused.
- List every cleaning, rinse, disinfectant, UV or sterilization step and its duration.
- Inspect for swelling, softening, color change, cracking, tackiness or residue after each cycle.
Use controls for biological work
Run the same cell, tissue or sample preparation in a validated reference vessel alongside the printed holder. Include an empty-holder control and, when practical, a material-only extract or soak control. Compare viability, morphology, attachment, imaging background and any assay endpoint that could be affected by leachables or surface texture. If a result changes, you need to distinguish the holder from handling, geometry and optical differences before adopting it.
Alcohol and UV treatment reported for a particular cryo-EM grid holder should be read as that study’s procedure, not a universal cleaning prescription. A different resin may require a different wash or cure, and some polymers can be damaged by chemicals or heat that another part tolerates.
A step-by-step material-selection workflow
- Map every interface. Draw the specimen, liquid, glass, objective, stage and operator contact surfaces. Mark which printed faces are dry, wetted, in the field of view or outside it.
- Set the failure priority. Decide whether the unacceptable outcome is fluorescence, drift, leakage, a failed glass bond, contamination, deformation or slow fabrication. Rank these before comparing materials.
- Choose a process candidate. Start with FDM PLA for a dry prototype or carrier. Consider PETG when durability or handling favors it. Consider MSLA resin for fine-detail, liquid-containing or glass-bonding geometry only with a defined post-processing and validation plan.
- Print a test matrix. Keep geometry constant while varying material, color, orientation and the settings that affect the critical interface. Include the actual production configuration, not only a convenient coupon.
- Measure fit and mechanics. Check flatness, critical dimensions, objective clearance, stage travel and thermal behavior at the operating temperature. Record the build orientation and infill so the result is reproducible.
- Measure optical background. Acquire a blank under the real excitation, emission filters and detector settings. Reject or relocate any part whose background masks the weakest signal.
- Test liquid and glass interfaces. Perform the extended fill, movement and temperature tests, then repeat after bonding and cleaning. A visual inspection alone is not a leak test.
- Run contact and cleaning controls. Expose the finished part to the exact sample and cleaning workflow, with a reference vessel and suitable controls.
- Document acceptance limits. Write down maximum background, allowable dimensional change, leak duration, bond failures, reuse count and visual defects. Approve the material only for the tested design and protocol.
Evidence from published holder examples
| Application | Reported material/process result | How to use the result |
|---|---|---|
| Live-cell optical system (Mod3D, PLOS ONE, 2022) | FDM chambers did not meet the project’s watertightness, resolution and glass-bonding needs; MSLA resin was used for chambers while FDM remained the carrier process. | Separate structural and wetted functions; validate the chosen resin and bond for your own microscope. |
| Same system’s carrier | A 4 mm FDM holder with 20% grid infill was dimensionally stable up to 37 °C. | Use as a design-specific reference for warm-stage testing, not as a polymer-wide rating. |
| Biomedical-imaging fluorescence study | PLA and PETG emissions could approach tissue-signal intensity in tested conditions; PETG background obscured skin and bone in one comparison. | Measure the exact color and part in your excitation and detection bands. |
| SEM holder study (Meloni and Bertotti, PLOS ONE, 2017) | Conductive PLA holders were fabricated in under 30 minutes at an average cost below $0.50 per piece. | These are 2017 study figures for that fabrication context, not current prices or optical performance. |
| Cryo-EM grid-holder study (Faessler et al., Journal of Structural Biology, 2020) | Reported reuse up to 15 times without observed adverse effects on sample preparation under its procedures. | Plan a reuse-and-cleaning validation; do not generalize the count to another design. |
| NIH 3D TH GridPlate resource | Recommends a chemical-resistant material such as PETG, ABS or polypropylene and a water-resistant sealant over tray wells for that TEM grid-staining context. | Apply the advice only to the stated tray use, and verify that sealants do not interfere with your optical or biological workflow. |
Common failure modes and corrective actions
The image has a bright haze or colored background
Suspect autofluorescence from the holder, supports, pigment or residue. Reacquire a blank with the production part, then move the printed region out of the field or change the material and color. Do not assume a darker filament will solve it without measuring.

The chamber slowly loses liquid
Look for layer-line paths, pinholes, port cracks and a defective glass bond. Repeat the extended fill test after warming and stage movement. If the FDM wall cannot meet the requirement consistently, redesign the chamber for a higher-resolution process rather than relying on an unvalidated coating.
The glass window bonds inconsistently
Inspect the bonding land for ridges, warping and support marks. Control the land geometry and preparation, then test multiple production parts. The Mod3D experience shows that surface imperfections can make FDM bonding unreliable for demanding chambers.
The specimen shifts or the objective clips the holder
Check flatness, datum stacking, stage travel and clearance with the complete optical train installed. Reorient layers or stiffen the carrier only after identifying the deformation path; adding material elsewhere may not correct a warped locating surface.
Cells or samples behave differently after reuse
Compare with the reference vessel and inspect the part after each cleaning cycle. Residue, incomplete resin cure, surface damage or a changed wetting behavior may be responsible. Reduce reuse or change the process only after the control experiment identifies a repeatable cause.
Practical recommendations by use case
For an early dry prototype
Print the carrier in PLA, document orientation and infill, and verify fit before investing in a final process. Keep the prototype out of the optical field and away from liquid whenever possible.

For a durable external carrier
Evaluate PETG alongside PLA for handling, clips and repeated loading. Choose from measured flatness, stiffness, thermal behavior and cleaning exposure rather than assuming PETG is automatically better.
For fluorescence imaging
Treat the blank fluorescence measurement as a release test. If the holder is near the specimen, test every color and formulation you may use, because optical background is not determined by polymer name alone.
For a sealed chamber or glass interface
Prototype the overall assembly in FDM if useful, but move the wetted chamber to a validated MSLA design when leak or bond tests show FDM is inadequate. Confirm resin washing, post-cure, optical background, extractables and cleaning compatibility before sample contact.
For repeated biological use
Approve a specific part, process and cleaning schedule together. A published reuse count or a material supplier’s generic claim cannot replace controls using your sample system.
Bottom-line selection rule
Use PLA first for simple, dry FDM carriers; consider PETG when the same external role needs more durability; and reserve resin printing for chambers or interfaces whose resolution, sealing or glass bonding justify the added validation. Regardless of the label, release a microscopy holder only after it passes the tests that match its location: fit and thermal stability for carriers, blank fluorescence for optical parts, leak and bond tests for chambers, and sample-contact and cleaning controls for biological workflows.
