Researchers at the Broad Institute and MIT used custom upconverting nanoparticle probes to track EGFR, HER2, and HER3 receptors on living cells. In a highlighted experiment, the system recorded receptor movement and pairing at 100-millisecond resolution for more than 16 minutes. The work, published online in Cell on April 28, 2026, offers a longer view of molecular behavior—not a new cancer test or treatment.
How the researchers tracked the receptors
EGFR, HER2, and HER3 are members of a receptor family involved in cell signaling. These proteins can pair with another copy of the same receptor, forming a homodimer, or with a different family member, forming a heterodimer. Pairing can initiate signaling inside a cell, so researchers want to know which partners receptors choose and how long those pairings last.

Conventional fluorescent dyes can lose their signal after seconds of laser illumination. The team instead engineered upconverting nanoparticles (UCNPs) containing rare-earth ions and customized them to label the three receptor types. A custom-built imaging setup then followed their movement and interactions in living human cells. Broad Institute’s account describes the receptors attaching to and separating from partners; GEN reports that the platform tracked all three types in three colours at 100-millisecond resolution for more than 16 minutes in the highlighted run. The researchers ended that run after capturing the long EGFR homodimers they wanted to study. These are reported results from this platform and experiment, not a performance guarantee for other samples or laboratories.

What the team observed about receptor pairing
EGFR dimers persisted longer
After stimulation, EGFR receptors remained paired for several minutes, according to the Broad Institute’s account. The group could not observe pairings lasting that long with traditional dyes. In the reported experiments, cancer-related EGFR mutations also made dimers more stable, including without an external stimulus.
Wild-type HER3 formed stable homodimers
The surprising result was that unmutated, or wild-type, HER3 formed unexpectedly stable homodimers. The researchers initially suspected an artifact, then repeated the experiment using different labels and probes; GEN reports that the result held.

The team’s proposed interpretation is that these stable HER3 homodimers may form a signaling-inactive pool, leaving less HER3 available for signaling-active pairings with other receptors. That is a working model of what the observations could mean, not an established clinical mechanism.
Mutation effects varied by receptor
The reported mutation effects were not uniform across the family. Studied HER3 mutations destabilized HER3 homodimers, potentially freeing HER3 to pair with other receptors. Studied HER2 mutations modestly increased homodimer stability. EGFR mutations stabilized EGFR dimers in the experiments. These molecular observations do not establish how a particular mutation affects a patient’s outcome or whether it predicts response to a therapy.

Why the probes use rare-earth elements
Rare-earth ions help the nanoparticles produce sustained luminescent signals under laser excitation. GEN identifies ytterbium, erbium, and thulium in the team’s probes. Broad says UCNP signals can remain stable for minutes, hours, and potentially years, and that probe colour can be engineered by changing the type and dose of ions. In this study, the materials are components of a research imaging probe—not a treatment given to patients.
The probe chemistry is only one part of the method. Peng described the integration of nanomaterial chemistry, molecular labeling, optics, and computation as the challenge that made the pipeline difficult. This is a specialized research platform, not an off-the-shelf consumer device.

What this means—and what it does not
The study provides a way to examine receptor pairing for longer than conventional dyes allowed in the reported experiments. That longer observation window revealed persistent EGFR pairings and an unexpected stable HER3 pairing pattern, giving researchers questions to investigate about how receptor availability and signaling relate.

Rare-earth supply figures need careful context
The scientific use of rare-earth ions also intersects with a concentrated supply chain, but the available figures refer to particular materials and a particular year—not directly to the custom probe salts or a current price quote. GEN attributes the following 2025 figures to Benchmark Mineral Intelligence:
- China accounted for 85% of total rare-earth oxide production.
- China accounted for an estimated 99% of dysprosium oxide and terbium oxide production.
- The average North American price for dysprosium oxide was 4.4 times the Chinese price.
These oxide production and price figures should not be treated as measures of the specific ytterbium, erbium, and thulium salts used in the probes. GEN reports that Peng’s lab buys rare-earth salts in vials from a U.S. distributor, but does not name the distributor or confirm the mine origin; the report says the material likely comes from China. It also reports that China added export controls covering ytterbium, erbium, holmium, thulium, and europium in October 2025, and that the expansion was suspended under a U.S.–China trade truce. That reported status is time-sensitive and should not be read as a current regulatory determination.
