1Repair Windows errors before they cause bigger problems2Scan for outdated or missing drivers - takes under a minute3Clear out junk files and repair common Windows errorsResearchers identify ancient drug formulations by combining two incomplete records: what historical texts say a preparation contained and how it was made, and what chemical traces survive in vessels, tools or preserved mixtures. Neither record is a complete label. Texts can omit tacit techniques or reflect later copying, while residues can be contaminated, degraded or left by a non-medical substance. The most defensible reconstruction aligns philology, archaeological context, analytical chemistry and, where possible, a carefully documented experimental recreation.
What counts as evidence for an ancient medicine?
“Ancient medicine” is not a single, neatly bounded category. A recipe may describe an ointment, powder, pill, drink, fumigation or plaster, but the same kinds of ingredients also appear in cosmetics, embalming materials, incense, craft products and ritual mixtures. Researchers therefore begin by asking what kind of evidence they have and what the object was used for.

There are two main evidence streams:
- Written evidence: recipe books, medical compendia, papyri, tablets and labels that name ingredients, symptoms, preparation and application.
- Material evidence: residues in containers, applicators, tools or burial objects, analyzed for surviving organic and inorganic compounds.
A strong interpretation uses both streams when they are available. A text can suggest what a residue might be, while chemistry can test whether a named ingredient or a plausible substitute survives. When only one stream exists, the conclusion should remain narrower: a chemical profile may show wax and resin without proving that the material was a medicinal ointment, and a recipe may describe an intended preparation without proving that anyone made it exactly as written.
How the reconstruction process works
Most projects move through several linked stages. Each stage answers a different question, and uncertainty at one stage cannot be silently converted into certainty at the next.

- 12-Month Warranty: Backed by a 12-month manufacturer warranty for peace of mind. Designed to meet strict quality control standards for reliable everyday performance.
- Establish the historical setting. Researchers date the text, vessel or object, identify its findspot and ownership history, and determine whether it belongs to a medical, domestic, funerary, cosmetic or industrial context.
- Interpret the language. Philologists examine the original terms, variant spellings, recipe formulae, disease categories and historical meanings of plant, mineral and animal-product names. A modern plant name is often an interpretation rather than a one-to-one translation.
- Characterize the object. Shape, capacity, closures, internal compartments, wear and associated finds can indicate whether a container was intended for storage, mixing, application or another purpose.
- Sample conservatively. Analysts document where a residue came from and distinguish material from the vessel itself, later conservation products and possible contamination.
- Profile the chemistry. Instruments separate and identify surviving organic molecules and characterize inorganic elements or minerals. Different techniques see different portions of a degraded mixture.
- Recreate and compare. When a recipe is sufficiently specified, researchers make a test preparation using historically plausible ingredients and processing, then compare its chemical profile with the archaeological sample.
- State the inference at the right level. The result might be “contains beeswax and conifer resin,” “consistent with an ointment,” or “supports a reading of this recipe.” It is rarely “this is definitely drug X at the original dose.”
Why recipe texts are not complete laboratory instructions
Ancient pharmacological knowledge travelled through both oral and written channels. L. M. V. Totelin’s study of Hippocratic recipes describes compilers drawing on diverse sources rather than recording a single, controlled authorial protocol (UCL Discovery). A written version may preserve one transmission of a recipe while other practitioners used variants.
Recipe formulae commonly contain a title, an indication, ingredients, preparation and application, but the preparation section can be remarkably short. The Oxford Academic chapter on medical recipes notes that techniques, apparatus and practical judgments are often left unstated because readers were expected to possess tacit knowledge (“Technologies of Knowledge: Pharmacology, Botany, and Medical Recipes”). “Boil,” “pound” or “mix” does not necessarily specify vessel material, temperature, duration, particle size, order of addition or filtration.
Translation adds another layer of interpretation. A historical word may cover several species, a substance may have changed name, and an illness category may not map cleanly onto a modern diagnosis. Researchers compare parallel passages, contemporary glossaries and material findings before assigning a modern identity. Even then, the result is usually a reasoned range of possibilities, not a guaranteed equivalence.
What a text can and cannot establish
| Textual detail | Useful inference | What remains unproven |
|---|---|---|
| Named ingredient | What a compiler said should be included | Whether the ingredient was available, authentic or present in every batch |
| Indication or symptom | The condition the recipe claims to address | A modern diagnosis or clinical effectiveness |
| Preparation verb | A broad operation such as grinding or heating | Exact apparatus, temperature, duration and sequence |
| Application instruction | Route or body site described by the text | Actual use by patients and safe dosage |
What the laboratory methods reveal
No single instrument acts as a direct label-maker. Researchers combine methods because ancient mixtures contain volatile compounds, fats, resins, sugars, pigments, salts and degradation products with very different analytical behavior.

| Method | What it is good at | Important limitation |
|---|---|---|
| Gas chromatography–mass spectrometry (GC-MS) | Separating and identifying many volatile or derivatized organic compounds, including lipid, resin and carbohydrate components | Very polar, non-volatile or heavily degraded material may require preparation or remain difficult to identify |
| X-ray fluorescence (XRF) | Detecting and comparing elemental signatures, useful for inorganic salts and pigments | Elemental presence does not by itself identify a compound, recipe or function |
| Raman spectroscopy | Characterizing molecular vibrations in inorganic and some organic materials, often with little sampling | Fluorescence, mixtures and weak signals can complicate interpretation |
| Nuclear magnetic resonance (NMR) | Profiling molecular structures and mixtures in recreated preparations or extracts | Requires a suitable quantity and condition of sample; a spectrum is not automatically a historical identification |
| Liquid chromatography–mass spectrometry (LC-MS) | Separating and detecting less-volatile, thermally fragile or polar compounds | Results depend on extraction, reference standards and the survival of diagnostic molecules |
Sampling location and controls matter as much as the instrument. A residue from a sealed interior compartment carries a different evidential weight from material on an exposed exterior. Analysts compare blanks, vessel materials and known conservation substances where possible, and report whether a compound was directly detected or inferred from a degradation product.
Case study: Roman bronze medicine containers
R. J. Stacey’s 2011 study examined residues from seven samples in cylindrical bronze containers held by the British Museum. The project paired GC-MS for lipid, resin and carbohydrate components with XRF and Raman spectroscopy for inorganic materials (PubMed record and abstract).
The reported materials included beeswax, animal fat, conifer resin, gum-derived sugars, elemental carbon, and lead and zinc salts. Taken together with the containers’ form, the findings were consistent with varied preparations such as ointments and powders or pills intended for medical use. That wording is important: chemistry supports a use interpretation; it does not assign every detected substance to a therapeutic role.
Residues differed between compartments of a multi-compartment container. In one compartment, beeswax was consistent with the “Punic wax” described by Pliny. The researchers made and analyzed an experimental version for comparison, using the modern profile to test whether the historical description could plausibly produce the archaeological signal. The comparison strengthens the argument for a wax preparation but does not recover the complete original formula, proportions or dosage.

This study illustrates the value of convergence. A vessel designed to hold preparations, a residue containing plausible pharmaceutical materials and a historical comparison can reinforce one another. Remove any one element and the claim becomes less specific.
Case study: rebuilding Egyptian recipes before testing them
A 2026 Journal of Ethnopharmacology study combined philological analysis with experimental recreations and modern chemical profiling (article record). The researchers used NMR, LC-MS and GC-MS to examine preparations made from recipe instructions and historically identified ingredients.
One recreation using Hordeum vulgare (barley) produced hordenine activity that corresponded to the sickness description associated with the recipe. Other components showed synergistic effects in the chemical analysis. Recreations involving Linum usitatissimum (flax) and Ceratonia siliqua (carob) showed limited or partial correspondence with categories associated with cough and fever.

These findings demonstrate why processing and combinations matter. Grinding, heating, soaking or mixing can change which compounds are extracted and how they interact. They do not constitute clinical proof that an ancient patient was cured, nor do they establish that a modern person could safely use the recreation. The experiment tests chemical plausibility under stated reconstruction choices, not historical efficacy in a controlled trial.

Why labels and residues can still mislead
Apothecary jars marked “hiera picra”
A 2022 Rijksmuseum Bulletin study analyzed residues from two apothecary jars inscribed with names associated with hiera picra, a preparation with a history extending over two millennia (“A Bitter Pill”). The objects were much later than antiquity: one was made in Italy in the early sixteenth century and the other in Delft more than two centuries later. Chemical analysis did not establish that the sampled residues were hiera picra.
The case separates three claims that are often conflated: a label can indicate what a container was supposed to hold; an object’s date and provenance can show when it was made; and chemistry can identify what remains. Only by assessing all three can investigators decide whether the residue supports the label. A later jar may have been reused, incompletely cleaned or filled with a related preparation.
An Egyptian black coating that was not a drug
A British Museum and UCL project analyzed more than 100 samples from 20 Egyptian funerary objects of the 22nd Dynasty, approximately 900–750 BCE (project findings). The black coating contained plant oil, animal fat, conifer resin, pistacia resin, beeswax and bitumen. Its chemical richness might resemble a complex pharmaceutical mixture, but the archaeological context associated it with a funerary process, not medicine.
This is a necessary counterexample. Multiple oils, fats and resins are not diagnostic of a drug. Researchers must keep ritual, funerary, cosmetic and technical materials visible as competing explanations rather than treating every unusual residue as pharmacological.

How researchers rank confidence
When comparing a proposed formulation with another, five questions expose where the argument is strong or weak:
| Axis | Higher-confidence situation | Lower-confidence situation |
|---|---|---|
| Evidence type | Text and residue independently point toward the same preparation | Only a generic residue or only a copied recipe survives |
| Context quality | Dated, characterized vessel with documented provenance and secure interior sample | Uncertain provenance, later inscription or exposed material |
| Analytical coverage | Organic and inorganic methods address the expected ingredients | One technique detects a narrow fraction of a complex mixture |
| Interpretive step | Compound directly detected and matched to reference material | Intended use, ingredient identity or therapeutic effect inferred indirectly |
| Historical scope | Evidence belongs to the period and culture under discussion | Early modern or modern material used as if it were ancient |
Confidence is therefore layered. “Beeswax is present” can be a direct analytical result. “The wax was Punic wax” may be a historical comparison. “The preparation was a wound ointment” adds an inference from vessel form and texts. “It healed wounds” is a further therapeutic claim requiring evidence the residue cannot supply on its own.
Keeping ancient and early modern pharmacy separate
Later historical medicines can demonstrate how scientists analyze preserved formulations, but they should not be folded into an ancient narrative. A 2021 study of remedies from the Roman apothecary Santa Maria della Scala, founded at the end of the seventeenth century, used GC-MS to investigate natural resins and balsams (study record). Those materials belong to early modern pharmacy. They can inform analytical practice and the history of formulation, yet they do not prove what an Egyptian, Greek or Roman-era medicine contained.

- Is the object securely dated, and does its context indicate medical use rather than burial, cosmetics or craft?
- Was the sample taken from an original interior residue, or could it be later contamination or conservation material?
- Which compounds or elements were directly detected, and which ingredient names were inferred?
- Does a contemporary or near-contemporary text describe a matching substance, preparation or container?
- Are translation choices and uncertain plant or mineral identifications made explicit?
- Were missing preparation steps reconstructed, and could different processing choices change the chemical result?
- Is the conclusion about composition, intended use, or therapeutic effectiveness—and does the evidence support that level?
Readers who want a broader map of the field can follow the Yale Ancient Pharmacology Program, which brings together historical, archaeological and scientific approaches. The central discipline remains the same: treat texts as historically transmitted instructions, treat residues as chemically altered traces, and make the strongest claim that both kinds of evidence can actually sustain.
