How Paleontologists Estimate How Long Collagen Survives After Burial

There is no universal expiration date for collagen in buried bone. Paleontologists estimate its expected survival by reconstructing the specimen’s temperature history, converting that history into a temperature-normalized “thermal age,” and then testing the bone for collagen-derived molecules. The estimate is conditional: burial chemistry, water movement, minerals, humidity, microbes and handling can make actual preservation better or worse than a temperature-only model predicts.

Most thermal-age calculations use 10 °C as a reference temperature and an assumed activation energy of 173 kJ/mol for collagen loss from bone. The result is an estimate of cumulative degradation, not a stopwatch that proves collagen is present or assigns an exact age to a peptide.

What “how long collagen lasts” actually means

Two different questions are often mixed together:

  • How much degradation should a burial history cause? This is what thermal age estimates.
  • Can collagen-derived material still be recovered and authenticated? This requires laboratory measurements on the bone.

A specimen can have a favorable modeled thermal age yet yield little usable collagen because of acidic groundwater, oxidation, microbial activity or repeated wetting and drying. Conversely, a site can preserve collagen beyond what a simple temperature model would suggest. “Survival” also needs a defined threshold. Detectable traces, enough peptide for species identification, and enough uncontaminated sequence for molecular interpretation are not the same standard.

Calendar age alone therefore gives a poor prediction. Two bones buried for the same number of years can experience very different temperatures and chemical environments. A cold, stable burial can produce less modeled degradation than a much younger bone that spent part of its history warm and wet.

How thermal age converts temperature history into an estimate

1. Reconstruct the effective burial temperature

Investigators begin with the temperature the bone probably experienced, rather than simply using today’s surface temperature. Depth, local climate, seasonal variation and major climatic changes such as glacial and interglacial intervals can all matter. The reconstruction may need to represent different periods of burial instead of one constant value.

Site information is equally important. Sediment, groundwater, mineral contacts, drainage and humidity influence the chemical reactions that damage collagen. A temperature history that cannot be defended from the geological and archaeological context creates a correspondingly weak thermal-age estimate.

2. Apply temperature-dependent degradation kinetics

Chemical reactions generally proceed faster at higher temperatures. Thermal-age models use that relationship to translate a changing temperature record into the equivalent amount of degradation that would occur at a fixed reference temperature, commonly 10 °C. The calculation is based on an Arrhenius-style kinetic model and an activation energy. In the collagen treatment described by Collins and colleagues (2009), the assumed activation energy for collagen loss from bone is 173 kJ/mol.

A warm interval can therefore contribute disproportionately to the cumulative estimate. A bone that spent a relatively short period at elevated temperature may acquire a greater modeled thermal age than one that remained cold for a much longer calendar period. The output is usually expressed as an equivalent duration at the reference temperature.

3. Define what counts as “survival”

The model must specify a loss threshold. A projection to 1% of the original collagen concentration is a different claim from a prediction of when no peptide will be detectable or when sequencing will fail. Without the threshold, a number described as a “maximum age” is ambiguous.

Thermal age is consequently best used as a screening and comparison tool: it ranks specimens or sites by expected degradation potential and helps decide which samples merit destructive or non-destructive analysis. It does not establish that collagen was recovered.

What the published projections show

The 10 °C, 1% projection

Buckley and Collins (2011) extrapolated from experimental decomposition rates that, under an optimal burial environment held at a constant 10 °C, collagen would take approximately 0.2–0.7 million years to decline to 1% of its original concentration. The range reflects the model and its assumptions; it is not a universal lifespan or a guarantee that a bone of that age will yield collagen.

The estimate is also an extrapolation from higher-temperature experiments to much longer timescales. Real deposits do not maintain one temperature indefinitely, and the “optimal” chemical setting used in the projection is not representative of every cave, floodplain, soil or seabed.

Reported peptides from Weybourne Crag

The same 2011 paper reported collagen peptides in bones from Weybourne Crag dated to about 1.5 million years. That is a site-specific observation about material examined in that study. It shows that collagen-derived peptides can be recovered from at least some Lower Pleistocene bone under particular conditions; it does not establish intact collagen survival for millions of years in all environments.

The authors also noted that, even after accounting for glacial intervals in British Quaternary temperature calculations, their more conservative estimate extended the potential range for collagen sequencing into the Lower Pleistocene. The observation and the model are related but answer different questions: one predicts degradation potential, while the other reports laboratory recovery.

Why actual sites can outperform or underperform the model

Evidence from subtropical Australia

Peters and colleagues (2023) reported collagen preservation in bones more than 50,000 years old from subtropical Australian contexts. Their study did not rely on temperature alone. It combined thermal-age estimates with Fourier-transform infrared spectroscopy (FTIR), collagen extraction and peptide mass fingerprinting using ZooMS, plus protein-deamidation calculations.

Results varied substantially among sites. The authors concluded that Australian collagen preservation does not follow one pattern, but depends on site-specific burial environments and environmental conditions in addition to age, temperature and humidity. In practical terms, two deposits with similar modeled thermal ages can produce different ZooMS success rates or molecular alteration profiles.

Tripot Cave and the limits of explanation

The Australian study discusses possible mineral-related explanations for unusual preservation at Tripot Cave. However, detailed geochemical information for the cave was unavailable, so the mechanism could not be identified. This is an important scientific boundary: an anomalous preservation result can be documented without claiming to know exactly which mineral or reaction caused it.

Because such exceptions are real, Peters and colleagues called for critical re-examination of thermal age as a prediction method. They also discussed non-destructive near-infrared approaches as a possible way to assess collagen preservation before committing to more invasive sampling.

How paleontologists test a bone rather than trusting the model

A robust assessment uses several independent lines of evidence. Each addresses a different part of the preservation question.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
Approach What it measures What it can and cannot establish
Thermal age Modeled cumulative degradation from temperature history and kinetic assumptions Predicts degradation potential; does not prove collagen was recovered
Collagen extraction and peptide mass fingerprinting (ZooMS) Whether collagen-derived peptides can be recovered and matched to taxonomic markers Provides direct molecular recovery evidence and can support identification; success depends on preservation and contamination control
FTIR Spectroscopic characteristics of the mineral and organic components of bone Provides a preservation assessment or proxy; it is strongest when interpreted with molecular tests
Protein deamidation Chemical alteration of amino-acid residues in surviving proteins Offers another preservation indicator; it is not a standalone age clock
Authentication analysis Sample context, contamination, sequence quality and molecular provenance Tests whether an apparent signal is endogenous and supports the claimed interpretation

A practical evidence sequence

  1. Document context. Record stratigraphic position, associated dates, sediment, depth, hydrology, minerals, storage and any conservation treatment. A later handling step can affect molecular results.
  2. Build the temperature model. State the reference temperature, activation energy, kinetic model and how temperatures changed through time. Report uncertainty instead of presenting one temperature as a measured fact when it is reconstructed.
  3. Set the preservation threshold. Specify whether the goal is 1% of original collagen, detectable peptide, ZooMS identification or sequence-level interpretation.
  4. Screen the bone. Use methods such as FTIR or a proposed near-infrared assessment where appropriate, preferably before taking a large sample.
  5. Recover and identify peptides. Collagen extraction and mass spectrometric peptide fingerprinting can show whether collagen-derived molecules remain and whether they carry taxonomic information.
  6. Compare molecular alteration. Deamidation and related measurements help characterize the condition of the recovered proteins, but should be interpreted alongside extraction success and spectral data.
  7. Audit authenticity. Evaluate blanks, modern handling, conservation materials, environmental contamination, sequence plausibility and whether the claimed molecules are endogenous to the fossil.

How to compare thermal-age studies fairly

A number from one specimen cannot be compared meaningfully with another unless the underlying definitions match. For each study or sample, record the following:

Comparison point Question to ask
Reference temperature At what temperature is equivalent time reported—10 °C or another value?
Kinetic parameters What activation energy and degradation model were used?
Threshold Does “survival” mean 1% concentration, detectable peptide, identification or sequencing?
Temperature reconstruction Were depth, seasonal changes and glacial or interglacial intervals included?
Burial setting What are the sediment, groundwater, mineral, humidity and microbial conditions?
Direct evidence Were collagen peptides extracted, and were FTIR, ZooMS or deamidation results reported?

Keeping these axes together prevents a model projection from being mistaken for a measured survival time. It also explains why an older bone from a favorable deposit may outperform a younger bone from a chemically destructive one.

Why microscopic preservation is not proof of ancient collagen

Preserved structures visible under a microscope can be valuable contextual evidence, but they do not by themselves establish that authentic endogenous protein remains. Molecular claims require evidence about the sample’s age and environment, contamination and handling history, sequence data and whether the interpretation fits the observed spectra and peptides.

Schweitzer and colleagues (2009), in their analysis of mass-spectrometric evidence associated with Tyrannosaurus rex, emphasized the need to weigh those issues rather than equating a visually preserved structure with biomolecular preservation. Saitta and colleagues (2019) further showed why recent material must be considered: Cretaceous dinosaur bone can contain recent organic matter and provide an environment conducive to microbial communities. A peptide signal therefore has to be tested for modern or microbial origin before it is treated as surviving dinosaur collagen.

Can collagen survive for millions of years?

It can be recovered from exceptionally favorable, well-documented contexts at ages far beyond the everyday archaeological range, but there is no settled universal maximum. The 0.2–0.7-million-year figure is a model projection to a stated 1% threshold at constant 10 °C under optimal conditions. The approximately 1.5-million-year Weybourne Crag result is a site-specific report of collagen peptides. The more-than-50,000-year Australian examples demonstrate that subtropical preservation can exceed simple chemical expectations in some locations.

Those statements should not be collapsed into “collagen normally lasts 1.5 million years.” They describe different thresholds, environments and types of evidence. The scientifically defensible conclusion is that thermal age estimates likelihood and guides sampling, while direct molecular tests determine what a particular bone actually preserved.
