Chiral covalent organic frameworks (CCOFs) are porous, ordered networks whose mirror-image pores can favor one enantiomer over the other. Researchers now make that selectivity by building chirality into the linkers, adding it after framework formation, or inducing it during assembly—and are using the resulting spaces for drug separations, chiral polymer “memory,” circularly polarized-light detection and spin-selective electronics.
What makes a covalent organic framework chiral?
A covalent organic framework becomes chiral when its structure lacks a mirror plane and creates a handed three-dimensional interaction environment. Chirality may come from stereocenters in molecular linkers, helical or otherwise twisted stacking, or the way otherwise achiral building blocks are organized into asymmetric pores. The key is not simply that a framework shows a circular-dichroism signal; its handed pores must also produce a measurable difference in how they bind, transform or transport mirror-image molecules.

A 2022 Chemical Science review identifies three established construction routes. They differ in when chirality enters the material and in how much the framework’s crystallization is coupled to chiral chemistry.
| Strategy | How chirality is introduced | Main design advantage | Main engineering issue |
|---|---|---|---|
| Direct synthesis | Optically pure diamines, aldehydes or other chiral monomers are used during reticulation. | Chirality is incorporated throughout the framework as it forms. | Chiral monomers can complicate crystallization and make control of order and handedness difficult. |
| Post-synthetic modification | An achiral parent COF is formed first, then chiral molecules or groups are covalently attached or loaded into its pores. | Framework formation and chirality installation can be optimized separately. | The modification must preserve pore access, connectivity and long-range order. |
| Chiral induction | A chiral auxiliary or environment biases assembly of otherwise achiral components. | Chiral function can emerge without making every building block enantiopure. | The induced handedness must be reproduced and distinguished from effects caused by morphology or aggregation. |
In direct synthesis, the 2024 Journal of the American Chemical Society study by Xing Han and colleagues condensed chiral 1,2-diaminocyclohexane with tri- or tetra-salicylaldehyde derivatives. The resulting Zn(salen) CCOFs-9–12 combine a metal-salen coordination environment with a permanently porous, chiral network.

Why handed pores separate enantiomers
Enantiomers have identical bulk formulas and many identical achiral properties, but they present different three-dimensional arrangements to a chiral surface. Inside a CCOF pore, hydrogen bonds, π-stacking, electrostatic contacts, steric fit and confinement can therefore give the two forms different adsorption free energies. In catalysis, the same contacts can stabilize one transition-state geometry more than its mirror image.
That interaction field supports several functions:
- Adsorption and separations: one enantiomer can bind more strongly or diffuse differently through the pore.
- Asymmetric catalysis: a chiral cavity can favor one reaction pathway and increase product enantiomeric excess.
- Molecular recognition and sensing: a guest’s handedness can alter binding and the resulting optical or electrical signal.
- Template chemistry: the pore can orient reactive monomers so that chirality is transferred to a new material.
The strongest quantitative example in the cited studies is material-specific: the Zn(salen) CCOF-9 series adsorbed racemic drugs with enantiomeric excess (ee) of up to 97% in the 2024 JACS report. That value is a maximum for the tested framework and drug conditions, not a universal performance level for all CCOFs.

How chirality can be encoded and retained
The most literal form of chirality encoding uses a CCOF as a nanoreactor. In a 2024 Angewandte Chemie International Edition study, achiral monomers polymerized inside two-dimensional or three-dimensional homochiral COF pores. The chiral, π-rich nanospaces directed enantiospecific propagation and self-propagation, a process the authors compared with a nonenzymatic polymerase-chain-reaction-like system.

The isolated material separated racemic amino acids with up to 85% ee, and the reported chiral memory survived heating to 200 °C. In this design, “programmable” has two separate meanings: framework pore geometry and linker stereochemistry determine the handed template, while the confined monomer chemistry determines which polymer receives the imprint.

Several measurements must be kept distinct. Circular dichroism can establish a chiroptical response from the framework or guest, but it does not by itself prove a particular separation efficiency. Enantiomeric excess measures product or adsorbate composition, while a liberated polymer’s optical response tests whether information was transferred and retained. A convincing claim identifies the material and reports the relevant metric at each level.
Optical and electronic functions beyond separation
Detecting circularly polarized light
CityU-7 and CityU-8 use porphyrin-based amines with chiral 2-methylbutoxy terephthalaldehyde linkers. Devices made from these frameworks directly detected left- and right-handed circularly polarized light at 405 nm. The CCOF is the active photodetection layer, rather than a passive chiral additive, showing how framework handedness can be coupled directly to a device readout.

Generating spin-selective current
The Zn(salen) work also explores chirality-induced spin selectivity (CISS). CISS is the proposed ability of a chiral material to transmit one electron-spin orientation more readily than the other. The 2024 JACS authors describe tunable Zn(salen) CCOFs as a route toward spin-polarized current without a permanent magnetic layer. This remains a design direction rather than a general specification shared by every chiral COF: spin selectivity must be measured for the particular framework, contact geometry and operating conditions.
How to evaluate a CCOF design
Comparing materials requires more than asking whether a sample is called “chiral.” Use the following checklist to connect structure to function:

- Identify the chirality source. Record whether it is a linker stereocenter, a helical or stacked architecture, pore confinement, post-synthetic group or induced assembly.
- Describe the framework. Note whether it is two-dimensional or three-dimensional, the linkage chemistry, pore dimensions and degree of crystallinity or order.
- Verify handedness with an appropriate control. Compare enantiomeric frameworks, achiral analogues and morphology controls where possible so that aggregation is not mistaken for intrinsic chirality.
- Match the readout to the claim. Use circular dichroism for chiroptical response, ee for separation or catalysis, wavelength-specific responsivity for circularly polarized-light detection, and a defined spin measurement for CISS.
- Test retention under the intended process. Check whether chirality survives guest exchange, chemical processing, framework removal and heating; the polymer-imprinting study reported persistence to 200 °C, but that temperature should not be generalized to other systems.
- Report the application conditions. Adsorption solvent, concentration, temperature, film thickness, electrode arrangement and cycling history can all change the measured result.
What remains difficult
CCOF construction is still a developing area. Reproducibly controlling handedness from batch to batch and at useful scale is difficult, particularly when chirality is induced rather than supplied by an enantiopure monomer. Researchers must also separate intrinsic framework chirality from effects of particle shape, film texture and aggregation.
There is no established universal conversion between one circular-dichroism metric and enantioselective performance. A large optical signal cannot be treated as a guaranteed 97% or 85% ee result. Device studies face a related challenge: a framework can respond optically to circular polarization without yet delivering the stability, signal-to-noise ratio or integration required for a practical sensor or spintronic component.
The field’s direction is nevertheless clear. Directly synthesized CCOFs provide highly defined chiral environments; post-synthetic and induction routes offer additional process flexibility; and templated polymerization shows that a framework can transfer handedness into a second material that remains chiral after the template is gone.
