For many exercising adults, a useful starting point is 20–40 g of high-quality protein per meal, or about 0.25 g per kilogram of body weight, with a balanced supply of essential amino acids (EAAs). The International Society of Sports Nutrition (ISSN) says an acute protein dose should contain 700–3,000 mg of leucine and/or a higher relative leucine content—but treats that as broad guidance, not a guaranteed on/off switch for muscle building.
Why leucine matters—but is not the whole meal
Leucine is an essential amino acid and one of the branched-chain amino acids. It helps signal muscle protein synthesis (MPS), the process of building muscle proteins. But the body needs all nine essential amino acids to build those proteins; leucine alone cannot supply the full set of building blocks.

The NIH Office of Dietary Supplements says most complete proteins are about 40% EAAs. That means a meal or snack with 25 g of complete protein provides about 10 g of EAAs. The practical goal is therefore a sufficient serving of complete protein, not merely a high leucine number on its own. NIH Office of Dietary Supplements: Dietary Supplements for Exercise and Athletic Performance.
What amount is a useful target per meal?
For healthy exercising people, the ISSN’s 2017 position stand gives a broad acute-dose guideline of 20–40 g of high-quality protein, or about 0.25 g/kg, alongside 700–3,000 mg of leucine and/or a higher relative leucine content and a balanced array of EAAs. These figures describe a range for a protein dose, not a precise threshold that guarantees an MPS response for every person or mixed meal. ISSN Position Stand: Protein and Exercise.

How much protein and leucine a meal supplies depends on the food or protein source and serving size. A complete-protein meal can contribute leucine as part of its amino-acid profile; you do not have to add isolated leucine simply to make the meal count.
Does leucine act as an MPS “trigger”?
The trigger idea is useful as a model, but the evidence does not establish a universal leucine switch. A 2021 systematic review assessed 29 eligible studies; 16 supported the hypothesis that leucine helps explain differences in post-meal MPS responses. The review found the model applied most clearly in older adults and when researchers used isolated protein sources rather than protein-rich whole foods. Leucine concentration alone did not explain every response. Zaromskyte et al., systematic review of the leucine-trigger hypothesis.

That distinction matters when applying supplement or isolated-protein findings to ordinary meals: a whole meal contains a mixture of nutrients and amino acids, and its effects cannot be reduced to one leucine number.

Aging may increase the leucine proportion needed to stimulate MPS, a phenomenon discussed as anabolic resistance. The ISSN’s 2023 position stand on essential amino acids notes that EAA-induced MPS occurs across doses and appears to plateau at around 15–18 g of EAAs, while aging may call for a greater proportion of leucine. This is an EAA finding, not a settled leucine-only target. ISSN Position Stand: Effects of Essential Amino Acid Supplementation on Exercise and Performance.

A 2021 review of nutritional interventions in older adults reports a proposed dose of 10–15 g of amino acids containing at least 3 g of leucine. The review also describes substantial variation in dose, timing, intervention length, and results, so that proposal should not be treated as a universal prescription. Nutrition Reviews: Factors influencing the efficacy of nutritional interventions on muscle mass in older adults.
How leucine fits with training and daily protein
Resistance exercise and protein intake each stimulate MPS, and the ISSN describes them as synergistic when protein is consumed before or after resistance exercise. A leucine-rich meal does not replace training or make total daily protein irrelevant.

The NIH ODS summarizes athlete guidance as about 0.3 g/kg of high-quality protein after exercise and then every 3–5 hours. That is general athlete guidance, not a requirement that applies identically to every person. Consider your overall diet and training pattern rather than judging progress by whether one meal reaches a particular leucine figure. NIH Office of Dietary Supplements: Dietary Supplements for Exercise and Athletic Performance.
Can food or protein powder provide enough leucine?
Complete protein foods can provide leucine alongside the other EAAs needed for protein synthesis. If a protein powder is a convenient way to meet your protein needs, compare its total protein per serving and complete amino-acid profile rather than assuming every serving contains the same amount of leucine.

NIH ODS notes that whey has more leucine than casein or soy, but describes the evidence as inconsistent on whether BCAA supplements stimulate MPS beyond adequate amounts of high-quality protein. A whey powder can be an optional convenience; the evidence does not establish that isolated leucine is necessary for people already getting enough complete protein. NIH Office of Dietary Supplements: Dietary Supplements for Exercise and Athletic Performance.
Quick Recap
How to use the numbers in practice
- Start with the meal: consider whether it provides a useful serving of complete, high-quality protein; the ISSN’s broad reference is 20–40 g or about 0.25 g/kg per acute dose for exercising people.
- Treat leucine as part of the amino-acid profile: the ISSN’s 700–3,000 mg range is guidance for acute protein doses, not a universal cutoff.
- Look beyond a supplement label: compare total protein, serving size, and complete amino-acid content; an isolated leucine claim does not show that the product provides all the EAAs.
- For individualized needs: older adults and people with clinical nutrition concerns may benefit from advice from a registered dietitian or qualified sports dietitian rather than applying a single target on their own.
