In a 2004 study, butyrylcholinesterase retained all of its measured activity after entrapment, and 90% of the soluble enzyme was immobilized. Those results apply to that enzyme and formulation, not to every enzyme trapped in silica.
What “naturally trapped” means
The phrase is a headline description, not a general biological category. In the method reported by Heather R. Luckarift, Jim C. Spain, Rajesh R. Naik and Morley O. Stone, enzymes were immobilized in biomimetic silica: the enzyme was present as silica nanospheres precipitated around it. The approach imitates aspects of biological silica formation rather than implying that enzymes are naturally trapped inside organisms. Luckarift et al., Nature Biotechnology (2004)

How enzymes are trapped in silica
The researchers used silaffin polypeptides derived from diatoms to catalyze silica formation in vitro. The authors describe the process as working at neutral pH and ambient temperature and pressure. As silica nanospheres precipitate, enzyme molecules become entrapped in the resulting silica support. They presented this as a comparatively benign alternative to some conventional chemical synthesis routes that use harsher conditions. PubMed record for the study
Do trapped enzymes remain active?
They can, but activity after immobilization depends on the particular enzyme and support. Luckarift and colleagues reported that butyrylcholinesterase entrapped during silica-nanosphere precipitation “retained all of its activity” as measured in their study. They also reported that 90% of the soluble enzyme was immobilized and that the immobilized enzyme was substantially more stable than the free enzyme. These are findings for the study’s butyrylcholinesterase and formulation, not a general guarantee.

Other enzyme-and-matrix combinations have produced different activity results. A separate 2004 sol-gel study reported relative activity, compared with the soluble enzymes, of 20% for alpha-amylase and up to 57% for catalase. The figures come from a different study and system, so they should not be read as a direct comparison with the biomimetic-silica result. Vera-Avila, Morales-Zamudio and Garcia-Camacho (2004)
Can immobilized enzymes be reused?
Immobilizing an enzyme can make it easier to separate from reaction material and recover, but reuse depends on how well its activity holds up and whether the support remains usable. The 2004 butyrylcholinesterase study reported use of the silica nanospheres in a flow-through reactor, demonstrating that the material could be used in that setup. It does not, on its own, establish a universal number of reuse cycles or performance for other enzymes. Luckarift et al. (2004)

Immobilization also involves trade-offs. A 2024 review on enzymatic disruption of lipid-rich microalgae cell walls notes that physical entrapment, typically in a silica matrix, can separate enzyme from reaction material and help recovery. It also cautions that the support can hinder hydrolytic efficiency through steric effects and may need replenishment. These are process considerations, not results established for the 2004 butyrylcholinesterase experiment. 2024 review

What to compare when evaluating an immobilized enzyme
- Activity retained: Check results for the exact enzyme and silica formulation; do not assume that one enzyme’s outcome transfers to another.
- Stability: Look for evidence over time or use cycles, rather than treating a single activity measurement as proof of long-term performance.
- Immobilization yield: Determine what proportion of the soluble enzyme was captured in the support.
- Substrate access: Consider whether the silica matrix restricts access to the enzyme or slows mass transfer.
- Recovery and flow: Check whether the support can be separated or used in the intended reactor configuration.
