How Electron-Spin Measurements Reveal Chemical Signals in Porous Crystals

Electron paramagnetic resonance (EPR) reveals changes around unpaired electrons—not a direct signal from every molecule trapped in a pore. In porous crystals, those electrons can belong to the framework, an adsorbed species, or a deliberately introduced spin probe. When adsorption changes a spin’s local environment, EPR can help researchers follow gas uptake, locate adsorption sites, and investigate host–guest interactions.

What EPR measures—and why porous materials need a spin

Electron paramagnetic resonance, also called electron spin resonance (ESR), detects paramagnetic species: atoms, ions, molecules, or other centers with unpaired electron spins. The unpaired electron is the source of the measurable signal. A porous crystal does not automatically produce a useful EPR signal just because it contains empty pores or adsorbed molecules; the experiment needs an EPR-active center to observe.

That center may be a transition-metal ion already present in the material’s framework, a paramagnetic species that has been adsorbed, or an organic radical introduced into the pore or framework. If a guest molecule enters a pore near an EPR-active center, it can change the spin’s local environment. EPR can then register a change associated with adsorption, even when the guest itself is not the signal source.

This distinction matters when interpreting a result. If an introduced radical reports a change, the immediate observation concerns the radical and its surroundings. That can provide useful evidence about the pore environment and host–guest interaction, but it is not automatically a direct measurement of every guest molecule or the total quantity of material inside the pores.

Why pores make local measurements useful

Porous crystals, including metal–organic frameworks (MOFs) and porous organic frameworks, contain internal spaces where molecules can adsorb. A measurement of the material as a whole may not show which internal environment a guest encounters. A spin associated with a framework site or placed in a pore can serve as a local reporter: if adsorption perturbs its surroundings, its EPR signal can provide information about what is happening near that center.

The word “local” is important. EPR evidence is tied to the center producing the signal. It can help characterize that center’s local structure or response to adsorbed molecules; it does not, by itself, guarantee a complete map of every pore, site, or guest in a crystal.

What chemical information an EPR signal can provide

Local framework structure

Paramagnetic framework centers can be used to characterize their local environment. The inference depends on which center is being observed and what structural question is being asked. A signal from one metal ion, for example, is evidence about that EPR-active site and its surroundings—not a universal readout of the full framework architecture.

Adsorption sites and host–guest interactions

A spin probe placed in a pore can report how its environment changes as molecules adsorb. A researcher can use these changes to investigate where adsorption occurs and how the guest interacts with the host material. The 2025 review by Muhammad Fernadi Lukman and Andreas Pöppl focuses on dihydrogen-isotopologue adsorption in porous materials and describes EPR approaches to monitoring adsorption and desorption as well as examining host–guest interactions at microscopic scale.

The method is especially useful when the question is not simply whether a material takes up a gas, but how adsorption affects a particular spin-bearing site. The measurement still needs careful interpretation: the probe’s response gives information through its local environment, so researchers must distinguish a change in the probe from a direct signal of the guest.

Gas uptake in a specific sensor demonstration

A 2023 study, “Embedded nano spin sensor for in situ probing of gas adsorption inside porous organic frameworks,” used an embedded Sc3C2@C80 molecular spin sensor in Py-COF. The study examined adsorption of nitrogen (N2), carbon monoxide (CO), methane (CH4), carbon dioxide (CO2), propylene (C3H6), and propane (C3H8) using EPR signal intensity and linewidth.

This example shows how an embedded spin sensor can be used to probe gas adsorption inside a porous organic framework. It should not be generalized into a claim that EPR can always identify any gas in any porous crystal. The sensor, framework, gases, and measured signal characteristics belong to that particular demonstration. Signal intensity and linewidth are observables researchers can examine; identifying a guest or comparing adsorption behavior requires interpretation in the context of the specific experiment.

Reactive species and chemistry

EPR can also be relevant when the question concerns paramagnetic reaction species rather than equilibrium gas uptake. A 2023 review of magnetic-resonance methods in MOFs cites EPR observation of NO2 dimerization in MFM-502 and work on paramagnetic atomic sites for catalysis. These examples illustrate a broader use: EPR can provide evidence about chemistry involving species with unpaired electrons in or around porous materials.

Choosing between continuous-wave and pulsed EPR

The choice of EPR approach follows the experimental question. The 2025 review discusses in situ continuous-wave EPR and pulsed EPR for porous-material adsorption studies. It describes continuous-wave measurements as suitable for monitoring adsorption and desorption in the examples surveyed, while pulsed techniques can provide higher-resolution insight into host–guest interactions.

Approach Role described in the 2025 review Question it can help address
Continuous-wave (CW) EPR In situ monitoring of adsorption and desorption How does the spin-bearing center respond as uptake or release proceeds?
Pulsed EPR Higher-resolution insight into host–guest interactions What can the spin’s response reveal about interaction details?

These are complementary roles, not a claim that one method is always better. The appropriate choice depends on the paramagnetic center, the guest and process, and whether the goal is to monitor uptake or resolve details of an interaction. A method that tracks a changing signal during adsorption may address a different question from one chosen to investigate local host–guest relationships.

How to plan and interpret an EPR adsorption experiment

  1. Identify the signal source. Determine whether the material already contains EPR-active framework centers, whether the adsorbed species itself is paramagnetic, or whether an organic radical or other spin probe must be introduced. Without a spin-active center or probe, EPR is not a direct detector for an arbitrary guest.
  2. Define the target claim. Decide whether the experiment is meant to characterize a local framework center, monitor adsorption or desorption, investigate adsorption sites and host–guest interactions, or observe a paramagnetic reaction species. These aims require different interpretations of the signal.
  3. Choose the measurement approach for that aim. For in situ monitoring of adsorption and desorption, CW EPR is among the approaches described in the 2025 review. For higher-resolution information about host–guest interactions, that review discusses pulsed EPR.
  4. Specify what signal feature is being interpreted. In the Py-COF sensor study, researchers used signal intensity and linewidth to probe adsorption. A change in a measured feature is an observation; the chemical conclusion depends on how that feature relates to the relevant center and experiment.
  5. Keep the claim proportional to the center. If the signal comes from an introduced probe, describe the result as evidence about the probe’s local environment and response. Do not turn that result into a claim of direct, universal chemical identification or total pore loading unless the specific experiment supports it.
  6. Use complementary characterization where needed. EPR is one part of a wider toolkit for MOF and porous-material characterization. Broader conclusions about structure or material function should be considered alongside other relevant measurements, rather than treating one spin signal as a complete description of a complex solid.

Limits that shape what the result means

  • Only paramagnetic species provide the relevant spin signal. Diamagnetic framework components and guests do not automatically produce an EPR signal. An experiment may need a naturally present paramagnetic center, a paramagnetic adsorbate, or an introduced spin probe.
  • A probe reports its own local experience. Introducing a spin probe can make an otherwise inaccessible pore environment observable, but the immediate evidence concerns the probe and its surroundings. Whether that observation generalizes to other sites or guests is a separate question.
  • A signal change is not automatically a chemical identification. The cited sensor demonstration used intensity and linewidth to study a defined set of gases in a particular porous organic framework. It does not establish that EPR alone can distinguish any arbitrary adsorbate in every porous crystal.
  • EPR is not, on its own, a total-loading measurement. A local spin response can reveal adsorption-related changes, but the evidence presented here does not establish a universal relationship between an EPR signal and the total amount of guest in all pores.
  • No universal sensitivity or detection limit follows from these examples. The cited sources establish uses and particular demonstrations, not a single sensitivity figure that applies across centers, materials, guests, and experimental conditions.

How EPR fits into the wider porous-materials toolkit

Magnetic resonance methods can contribute information that complements other ways of studying MOFs. In their 2023 review, Lena Marie Funke and co-authors write: “Magnetic resonance spectroscopy adds value to MOF research in various ways, including monitoring crystal growth to ascertain mechanisms for synthesis, measuring adsorption isotherms with spectroscopically resolved site resolution, and aiding in the discovery of a new material class, MOF glasses.” That broader perspective helps explain why EPR should be seen as one route to site-sensitive information, rather than a replacement for every structural or adsorption measurement.

The same review reports a filtered set of 124 NMR/EPR studies of MOFs published from 2017 through 2022, including 26 EPR studies. The authors describe a Scopus and Web of Science search with stated filters, so those figures are counts from that review’s defined search—not a census of all MOF or EPR publications. The review also reports 450 MOF solid-state NMR studies from January 2017 through July 2022; that is a different count with a different search scope and should not be combined with the 124-study figure.

A practical way to read an EPR claim

When evaluating a report about molecules inside a porous crystal, start with the source of the unpaired electron. Then ask what changed in the measurement, what the authors infer from that change, and how narrowly their conclusion applies. A framework ion, adsorbed paramagnetic species, and introduced radical do not support identical claims, even if all produce EPR signals.

Next, separate three levels of conclusion: the observed spin response; the local structural or interaction interpretation; and any broader statement about gas identity, adsorption across the material, or total loading. The further a conclusion moves from the specific paramagnetic center, the more it depends on supporting evidence from the experiment and complementary characterization. This is the key to understanding what EPR can reveal: it is a selective window into spin-bearing sites and their environments, not a chemical microphone for every molecule in a pore.
