Escherichia coli can sense chemical traces released by neighboring gut bacteria and alter its own metabolism in response. A study published in Scientific Reports on 25 August 2026 identifies glucose-6-phosphate (G6P), or a closely related hexose phosphate, as the main signal released by tested Bifidobacterium species. E. coli detects that signal with the UhpC-UhpB-UhpA two-component system, which turns on the uhpT transporter gene.

What “metabolic eavesdropping” means

Metabolic eavesdropping is chemical sensing without direct contact. One bacterium releases metabolites while growing; another detects those molecules in the surrounding medium and changes gene expression or nutrient uptake. In the new work, E. coli is not responding to a deliberately emitted message. It is reading a by-product of Bifidobacterium’s metabolism.

Professor Kaneyoshi Yamamoto of Hosei University described the finding as “a new mechanism through which one bacterium detects the metabolic activity of the other.” The experiments were performed with bacterial cultures and conditioned media, so they demonstrate a molecular interaction rather than a proven human-gut health effect.

How E. coli detects the signal

G6P is the leading candidate

The strongest evidence points to extracellular G6P or a structurally related hexose phosphate. Researchers screened 54 E. coli transport systems representing 61 carbon sources. The screen highlighted the uhp pathway, which normally helps E. coli import hexose phosphates.

In this pathway, UhpC, UhpB and UhpA form a two-component sensory system. Detection outside the cell activates UhpA-dependent transcription of uhpT, the gene for a hexose-6-phosphate transporter. The system therefore acts as both a sensor and a switch for nutrient acquisition: E. coli detects evidence that a usable carbon source is nearby, then increases its capacity to bring related molecules inside.

The kinetic-trap test

To test whether G6P-like chemistry was responsible, the team treated Bifidobacterium-conditioned medium with glucose-6-phosphate dehydrogenase (Zwf), an enzyme that consumes G6P. A fast, wild-type Zwf removed the reporter-inducing activity far more quickly than a slow D177N Zwf mutant.

After 30 minutes, wild-type enzyme treatment reduced uhpT promoter activity by approximately tenfold. The slow mutant did not significantly reduce the signal. This kinetic result supports G6P—or a closely related hexose phosphate that Zwf can process—as the primary inducing molecule, while not proving that every active molecule in the medium is identical G6P.

Why the response is selective

The sensing response was not a generic reaction to any gut bacterium. Every tested Bifidobacterium species activated the uhpT reporter. The tested E. coli strains and five tested Bacteroidota species did not produce the same activation under the study conditions.

That selectivity matters because it suggests that taxonomic neighbors can leave chemically distinctive traces. A metabolite signal may be common enough within one genus to support cross-species sensing, yet absent or too dilute in secretions from other bacteria.

More than one signal is present

The conditioned medium contained independent information channels. Activation of another transporter gene, idnT, continued even when uhpT was deleted. If all activity traveled through one Uhp-to-Idn chain, removing uhpT would be expected to eliminate the downstream response. Instead, the result indicates that Bifidobacterium supernatant contains multiple signals sensed by separate pathways.

In practical terms, E. coli appears to be reading a chemical profile rather than a single all-purpose message: one component is detected through UhpC-UhpB-UhpA, while another independently activates the idn system.

What this suggests about gut ecology

A “non-dominant network”

The authors propose that precise molecular links can let low-abundance species coordinate metabolism alongside numerically dominant gut taxa. This is a “non-dominant network”: ecological influence is not determined solely by which organisms are most numerous, but also by whether a species supplies a signal that another organism can detect and use.

Short-range communication, with an important qualification

A research summary associated with the study describes the G6P signal as short-lived and suggests an approximately 40-micrometre spatial hub in which the interaction could operate. That distance is an ecological interpretation of signal lifetime, not a measurement made in a human intestine. It should be understood as a proposed scale for close bacterial neighborhoods, not a demonstrated gut-wide range.

What the experiments do—and do not—show

  • Established: E. coli reporter activity changes in response to metabolites in Bifidobacterium-conditioned medium.
  • Strongly supported: G6P or a closely related hexose phosphate is the main inducer of the Uhp response, based on the Zwf kinetic-trap experiment.
  • Established in the tested panel: Bifidobacterium supernatants activated the reporter, whereas the tested E. coli and five Bacteroidota species did not.
  • Established: At least one additional signal acts independently of uhpT, because idnT activation persisted after uhpT deletion.
  • Not established: that the same concentrations, distances or interactions occur in humans, or that changing this pathway improves health.
  • Not established: a commercial probiotic, approved therapy or clinical benefit.

Could this lead to a new probiotic?

Potentially, but only as a research direction. The authors mention next-generation probiotics and targeted gut therapies as possible applications. A future strain might be selected or engineered to release a useful metabolite, respond to a neighbor’s signal, or stabilize a local metabolic exchange. Turning that concept into a treatment would require work that this study did not perform: measuring the pathway in animal and human communities, determining safe and effective dosing, confirming persistence, and showing a reproducible clinical outcome.

The immediate value of the finding is mechanistic. It gives researchers a defined sensor pathway, a candidate signal and a way to test whether specific bacterial pairings exchange metabolic information.

Why the finding matters

Gut bacteria do not need a nervous system or a classic pheromone-like communication circuit to coordinate behavior. Nutrients and metabolic by-products can function as local information. In this case, E. coli appears able to infer that Bifidobacterium activity is nearby and adjust transport machinery accordingly.

The study expands the picture of microbiome regulation from competition for food to chemical interpretation of a shared environment. It also shows why abundance alone is an incomplete guide to ecological importance: a relatively uncommon organism may influence its neighbors if it produces a signal those neighbors are equipped to read.
