Researchers traced newly made sugar chains in mice by feeding cells a modified sugar precursor, then attaching a detectable tag to it with copper-free click chemistry. The key qualification: the sugar was labeled inside the living animal, but the reported Flag assay was performed after cells were extracted. The mouse study did not demonstrate direct live imaging.
How did the method reveal newly made glycans?
Glycans are complex sugar chains made by cells and found inside them and on their surfaces. Their patterns can change with cell type, developmental stage, and disease, but conventional detection methods such as antibodies and lectins have limitations for use in living animals.

In the 2010 mouse report, Pamela Chang and colleagues at the University of California, Berkeley, used a synthetic precursor of sialic acid that carried an azide chemical group. Cells incorporated the modified precursor into newly synthesized glycans. Researchers then added a strained cyclooctyne linked to a Flag peptide. The cyclooctyne reacted selectively with the azide in a copper-free click reaction, leaving the Flag tag attached to glycans made through the labeling pathway. A standard biological assay could detect that tag. Chemistry World’s report describes the 2010 work and cites the original PNAS paper.

Not with the readout reported in this study. The modified precursor was introduced into the living animal, so labeling began through biological synthesis in vivo. But the Flag assay required cells to be extracted before the tag could be detected. Chang discussed monitoring glycan-expression patterns across cell types and tissues as a potential use; she described attaching an imaging agent to the cyclooctyne as a future goal, not a result already achieved in the mouse experiment.

That distinction matters: metabolic labeling in a living animal is not the same as directly watching the labeled molecules in that animal. The report does not establish a clinical diagnostic application, nor does it show that every kind of glycan can be tagged.
How does the mouse work compare with earlier zebrafish imaging?
A separate 2008 study summarized in Lab Animal used azide-modified sugars and strained fluorescent alkynes to image sugar patterns in living zebrafish embryos. That report described bursts of sugar synthesis in the jaw, pectoral fins, and olfactory organs at 60 hours after fertilization. It is useful context for copper-free click chemistry in biological systems, but it is a distinct study with a different model and readout.

What does later azido-sugar research add?
A 2024 ChemSusChem paper reported enzymatic, gram-scale production of UDP-6-azido-GalNAc, a nucleotide sugar relevant to glycan engineering and click-chemistry applications. Frohnmeyer and colleagues reported production up to 2.1 g, yields up to 97% across five consecutive batch cycles, and 99.96% HPLC purity for purified product. These are results from that production study; they do not identify the precursor used in the 2010 mouse experiment.
As Chang put it, “In this way we can monitor the expression of patterns of glycans in different cell types and tissues.” In the mouse report, that describes the method’s potential for pattern monitoring, not direct imaging in a living mouse. Hermen Overkleeft, a professor of bio-organic synthesis at Leiden University, described the broader challenge: “Making two molecules react both efficiently and selectively in living animals is one of the most challenging yet ultimately most rewarding research objectives in chemical biology.”
