Cells can squeeze through gaps narrower than the nucleus that carries their genome. They do this by deforming the nucleus and coordinating forces from the cytoskeleton with a reinforced nuclear envelope, nuclear lamina and chromatin. Those structures reduce mechanical stress, but they are not an impenetrable shield: severe confinement can rupture the envelope or damage DNA without any visible rupture.

Protection therefore works as a sequence rather than a single barrier. The nucleus yields enough to pass, senses the mechanical strain, limits the duration of an envelope breach and recruits repair machinery. Whether the cell survives, pauses its cell cycle or accumulates lasting genome instability depends on the constriction, the cell type, the condition of its lamina and chromatin, and whether DNA damage occurred independently of a rupture.

Why a tight gap is a problem for the nucleus

The nucleus is commonly the largest and stiffest obstacle a migrating cell must move through. A 2026 review by Patel and colleagues reports typical nuclei of roughly 10–15 micrometres across, while tissue interstitial spaces can be as narrow as 1–10 micrometres. Those figures are contextual values from the review, not universal dimensions for every tissue or cell.

When the cell body enters a constriction, the nucleus cannot simply remain spherical. Actin, myosin and other cytoskeletal systems push and pull on the nuclear surface. The nucleus elongates, flattens or bends until it can follow the available path. This deformation concentrates stress at curved edges and narrow contact points, making the nuclear envelope the mechanical bottleneck even when the rest of the cell has already passed.

The nuclear envelope is made of inner and outer membranes. Underneath it, the nuclear lamina forms a supportive meshwork, while chromatin inside the nucleus also contributes to its mechanical behavior. Together, these materials let the nucleus resist force and distribute it rather than allowing one small region to bear all the load.

The nucleus uses several protective layers

Mechanical support from the lamina

Lamins reinforce the inner nuclear membrane and help the envelope withstand the compression and shear generated during migration. In the models used for the foundational 2016 study, reducing lamin levels increased the incidence of nuclear-envelope rupture under confinement. That result demonstrates a relationship between lamina strength and rupture risk in those tested cells and geometries; it does not establish one rupture threshold for every cell type.

Chromatin helps bear and distribute force

Chromatin is not only a storage medium for genes. Its organization and attachment to the nuclear interior affect how the nucleus deforms. During confined migration, chromatin can be pushed toward or through a damaged envelope, a process described as chromatin herniation. The 2016 study linked this phenomenon with DNA damage in confined cancer-cell migration models.

Cytoskeletal control of the passage

The cytoskeleton transmits force to the nuclear surface and controls how quickly the cell advances. A gradual, coordinated deformation gives the envelope more opportunity to redistribute stress. A severe or abrupt constriction can concentrate force and increase the chance of a tear. The precise balance depends on the geometry of the gap and on the cell’s mechanical state, so a result measured in one microfluidic or tissue model should not be generalized to all migration.

What happens as a cell squeezes through a constriction

  1. The cell encounters a size mismatch. The cell body can often narrow more readily than the nucleus, leaving the nucleus at the front of the constriction.
  2. Force is transmitted to the nuclear surface. Cytoskeletal structures push the nucleus forward and pull from behind, while contact with the surrounding material presses on its sides.
  3. The nucleus deforms. The lamina and chromatin resist the deformation, but the envelope changes shape to fit the available space. Nuclear transport and chromatin organization can change as this occurs.
  4. Small defects may appear. Under sufficiently high strain, the envelope can rupture. Nuclear and cytoplasmic contents can then mix, and proteins normally confined to one compartment can become mislocalized.
  5. The cell either restores the barrier or suffers continuing damage. Repair machinery is recruited to limit the breach. If the stress is too great, or if DNA lesions have already formed, migration can be followed by delayed cell-cycle progression, altered signaling, reduced survival or genome instability.

How an envelope rupture can threaten DNA

An intact nuclear envelope separates DNA from the chemically different cytoplasm. A rupture temporarily removes that separation. Cytoplasmic factors can contact chromatin, and nuclear proteins can diffuse outward. The 2026 review identifies the ER-associated exonuclease TREX1 as one route by which a breach can contribute to DNA damage. The mechanism is reported in specific studies, not as a guaranteed outcome of every rupture.

Physical deformation can also produce DNA damage without a rupture being detected. Extreme nuclear shape changes can interfere with replication and stall replication forks. This is important because an experiment that records no visible envelope tear cannot automatically conclude that the genome was unharmed. Conversely, observing a rupture does not prove that every DNA lesion in that cell came from the breach.

The 2016 confined-migration study reported chromatin herniation and DNA damage, with rupture incidence rising as confinement increased and as lamin levels were reduced. It did not provide a single rupture rate that can be applied to all tissues. Geometry, speed of migration, duration in the constriction and the cell’s lamina and chromatin properties all influence the result.

How cells repair a damaged nuclear envelope

Rapid sealing at the breach

Envelope repair is an active response. The 2026 review describes BAF and Lamin C appearing rapidly at rupture sites before the later recruitment of repair machinery. BAF can help protect exposed chromatin and organize the damaged region, while Lamin C contributes to rebuilding mechanical support around the opening.

ESCRT machinery closes the membrane gap

ESCRT proteins, including ESCRT-III, participate in restoring nuclear-envelope integrity. ESCRT-III can assemble at membrane disruptions and help drive membrane remodeling that seals the opening. The exact timing and molecular sequence vary among experimental systems, but the central point is consistent: the cell does not passively wait for a tear to disappear; it mobilizes a membrane-repair program.

Envelope repair is not DNA repair

Sealing the envelope re-establishes compartmental separation. It does not automatically reverse DNA breaks, stalled replication forks or other lesions created while the nucleus was deformed or open. A study can therefore find successful envelope repair alongside persistent DNA-damage signals. These are separate outcomes and should be measured separately.

DNA damage can occur even when the envelope stays closed

Mechanical strain changes the internal arrangement of chromatin and can disturb the processes that copy and read DNA. Replication forks that encounter an unusually deformed or mechanically stressed nucleus may stall. A stalled fork can require additional processing before replication resumes, creating an opportunity for lesions or rearrangements even when microscopy shows no rupture.

Deformation also affects nuclear transport and signaling. Proteins may enter or leave the nucleus at different rates, and altered chromatin organization can change which genes are accessible. These effects are not synonymous with DNA breaks, but they can change how a cell responds to stress, repairs lesions and decides whether to continue migrating.

Why outcomes differ between cells and experiments

There is no universal “squeeze limit” for cells. Comparisons are meaningful only when several variables are kept in view.

Context reported in the literature Observation Interpretation
Confined cancer-cell migration models in the 2016 primary study Chromatin herniation and DNA damage were observed; rupture became more frequent with stronger confinement and lamin depletion. The lamina and the degree of confinement influence nuclear-envelope failure in the tested models.
Developing neurons in a 2026 Nature study DNA damage associated with migration was reported as non-lethal. Mechanical migration stress is not exclusively a cancer-cell problem, and damage does not inevitably kill the cell.
Fibroblasts and dendritic cells discussed in reviews They appear in distinct biological settings with different migration demands and nuclear properties. Cell type and tissue context must be specified before comparing damage, repair or survival outcomes.

Six questions are especially useful when evaluating a result: How narrow and long was the constriction? What cell type was tested? How strong and how organized were its lamina and chromatin? Was an actual rupture observed? Could the DNA damage have arisen independently through replication stress or deformation? Were envelope repair, DNA repair and cell fate measured as separate endpoints?

What the nucleus does after the immediate stress

The nucleus is not merely cargo that survives a passage. The 2026 review describes it as both a mechanical obstacle and a mechanoresponsive organelle. Deformation can reorganize chromatin, alter nuclear transport and change signaling. Those changes feed back on migration: a cell may alter its speed, direction, adhesion or persistence after encountering a constriction.

Some consequences appear quickly, while others emerge later. A cell that reseals its envelope may continue moving but show delayed cell-cycle progression, a response discussed in a 2019 review. Another cell may tolerate a lesion and remain alive, as in the developing-neuron findings, yet carry altered genome or signaling states. Severe or repeated damage can instead reduce survival or promote genome instability.

Can squeezing through a narrow space damage DNA?

Yes, but the probability and consequence depend on the circumstances. Strong confinement can rupture the envelope and expose chromatin to cytoplasmic factors. Independent mechanical deformation can stall replication forks or otherwise injure DNA without a recorded rupture. The protective lamina, chromatin organization and active envelope-repair systems lower risk; they do not guarantee that no lesion will occur.

The most accurate conclusion is conditional: cells are equipped to deform their nuclei, limit envelope breaches and restore the barrier, yet migration through a tight space can still leave DNA damage or altered nuclear function. Evidence from one cell type, constriction geometry or experimental time point cannot be converted into a universal damage rate.

What this means for interpreting migration studies

  • Do not treat a closed envelope as proof of an undamaged genome. Replication-fork stalling and other deformation-linked mechanisms can operate without a visible tear.
  • Do not treat a rupture as proof of irreversible failure. BAF, Lamin C and ESCRT-III can restore envelope integrity, and some cells survive with non-lethal damage.
  • Keep compartments and endpoints separate. Envelope sealing, removal of DNA lesions, cell-cycle recovery, migration and survival are different measurements.
  • Report the biological setting. Cancer cells, fibroblasts, dendritic cells and developing neurons do not share identical nuclear mechanics or consequences.
  • Match claims to the tested geometry. A 1–10 micrometre interstitial-space range and 10–15 micrometre nuclear-size range provide context, not a universal rule for every organism or tissue.

The practical takeaway

Cells protect DNA during confined migration through a layered system: the lamina and chromatin provide mechanical reinforcement, the cytoskeleton manages deformation, and BAF, Lamin C and ESCRT machinery help reseal a ruptured envelope. That protection is effective but limited. A cell can repair the barrier while retaining DNA lesions, and it can sustain DNA damage even when no rupture is detected. The outcome is therefore determined by the severity and geometry of confinement, the cell’s nuclear materials and the distinction between repairing the envelope, repairing DNA and surviving the journey.
