How Can Dinosaur Collagen Last Millions of Years if Peptide Bonds Break Down in Water?

Peptide bonds can be vulnerable to water, but collagen’s repeating triple-helix structure may help shield them. A 2024 study proposes that interactions between neighboring carbonyl groups make water-driven bond cleavage harder. It offers a molecular piece of the puzzle—not proof that entire, unchanged collagen molecules survived fossilization.

How can dinosaur collagen last millions of years if peptide bonds break down in water?

Proteins are chains of amino acids joined by peptide bonds. In water, those bonds can undergo hydrolysis: water reacts with a bond and splits the chain. MIT’s 2024 report gives an ordinary peptide-bond half-life of about 500 years in the context described by the study. That figure is not a universal decay clock for collagen buried in every geological setting.

Collagen is not simply a loose chain of peptide bonds. Its three chains wind into a triple helix, and the 2024 proposal focuses on how that molecular arrangement could make some bonds less accessible to water.

What interaction might protect collagen’s peptide bonds?

The 2024 study describes an intramolecular n→π* interaction: an oxygen lone pair on one acyl group interacts with the antibonding orbital of a neighboring carbonyl. In shorthand, this is an O···C=O interaction. The authors report that these interactions recur along the collagen triple helix and can hinder water from reaching peptide bonds.

The proposed effect is local and chemical: it may make hydrolysis more difficult at bonds in the relevant structure. The study does not show that all bonds are protected equally, nor that a collagen molecule could remain untouched through tens of millions of years of burial.

What did the 2024 study actually test?

Researchers examined collagen and collagen mimics using chemical and computational work. In experiments comparing trans and cis forms of collagen mimics, the trans form—resembling the usual collagen structure—resisted water attack, while the cis form was more susceptible. The study therefore supports a bond-level protective mechanism in those systems; it did not recreate millions of years of fossilization.

MIT News quoted study author Ron Raines, Firmenich Professor of Chemistry at MIT: “We provide evidence that that interaction prevents water from attacking the peptide bonds and cleaving them. That just flies in the face of what happens with a normal peptide bond, which has a half-life of only 500 years.” The half-life comparison belongs to the study’s stated context, not every collagen bond in every environment.

Does the new interaction explain all of the collagen found in dinosaur bones?

No. The n→π* proposal offers one possible contribution to molecular survival. It does not by itself explain all reported fossil collagen evidence or establish that complete, native collagen tissue remains intact.

Different explanations act at different scales, and they need not be mutually exclusive:

Possible contributor What it could affect Evidence and limits
n→π* interactions Access of water to peptide bonds within the collagen triple helix. The 2024 study tested collagen or mimics through chemical and computational work; it did not reproduce long-term fossil burial. ACS Central Science study abstract (2024).
Physical shielding in fibrils Which regions of collagen are exposed or protected within tightly packed structures. A 2011 study mapped 11 peptide sequences extracted from dinosaur bone onto collagen fibril models. They localized to closely packed, shielded regions, consistent with selective survival rather than uniform preservation. PLOS ONE study (2011).
Burial conditions, including reduced water access The surrounding environment and the rate at which water can reach tissue. Reduced water access is a proposed contributor; the cited studies do not establish one set of conditions as a universal explanation for every fossil. MIT News (2024); Proceedings of the Royal Society B (2019).
Post-mortem chemical modification Protein chemistry, potentially including cross-linking or glycation. Iron-mediated chemistry and glycation have been discussed or tested as possible contributors. They are not established as a single explanation for all fossil collagen. MIT News (2024); Proceedings of the Royal Society B (2019).

What does “collagen found in a dinosaur bone” mean?

It matters whether a report concerns extracted peptide sequences, structural evidence, or intact native protein. These are not interchangeable. Finding collagen-derived peptides does not show that a complete collagen molecule or pristine tissue has survived unchanged.

The distinction applies to reports from fossils described as about 80 million years old and nearly 200 million years old in MIT’s 2024 account: those approximate specimen ages do not, by themselves, prove that entire collagen molecules remain intact. The 2011 mapping of 11 extracted peptide sequences supports selective survival in protected fibril regions, not uniform preservation of a whole protein.

Why this is a plausible contribution, not a complete answer

The 2024 proposal helps explain why collagen may behave differently from a simple estimate based on ordinary peptide-bond chemistry: neighboring groups in its characteristic structure may make water attack less effective. Physical shielding and burial chemistry could add further protection, while post-mortem modification may alter what survives. The available findings support multiple possible contributions, not one universal mechanism or a claim of unchanged collagen tissue persisting for millions of years.
