An AI model helped researchers search a database of more than 107 million molecules for possible antibacterial activity. The researchers did not test all 107 million in a lab: they experimentally tested 23 predictions from that collection, identifying eight antibacterial compounds. Two showed potent broad-spectrum activity in the reported experiments, including activity against multiple resistance determinants in E. coli.
What did the AI screen?
The 2020 study by Jonathan Stokes and colleagues used machine learning to predict which molecules might have antibacterial activity. For this search, the team drew on more than 107 million molecules curated in the ZINC15 database. The model ranked candidates for follow-up; the database’s full collection was not a laboratory test set.

The paper, published in Cell on February 20, 2020, describes the approach as using a model to prioritize compounds for experimental evaluation. The scale of the computational search made it possible to look across a very large chemical collection, but experimental evidence came from a much smaller selected group.

Were all 107 million molecules tested?
No. Researchers experimentally tested a discrete set of 23 predictions from the ZINC15 search. Eight of those tested compounds showed antibacterial activity. In other words, “107 million” refers to the molecules considered computationally, while 23 is the number of model predictions from that collection that were empirically tested in the reported work.

Two of the eight compounds showed potent broad-spectrum activity and overcame multiple resistance determinants in E. coli in the study’s experiments. These findings establish promising laboratory activity, not effectiveness in people or approval as medicines.
How does halicin fit into the findings?
Halicin was a separate result from the same study, not one of the eight antibacterial compounds identified from the 23 ZINC15 predictions. The researchers found halicin using a different collection, the Drug Repurposing Hub.

The study reported that halicin was active against a range of pathogens, including Mycobacterium tuberculosis and carbapenem-resistant Enterobacteriaceae. It also reported treatment effects in mouse models of C. difficile and pan-resistant Acinetobacter baumannii infection. Those animal-model findings are preclinical evidence; they do not show that halicin treats infections in humans.

What the study shows—and what it does not
The work demonstrates how machine learning can help prioritize chemical candidates for early antibiotic discovery: the model searched a large collection computationally, then researchers tested selected predictions in experiments. The reported results support the value of that screening strategy, while also showing why a large prediction pool should not be confused with a large-scale lab trial.

The study does not establish clinical benefit, regulatory approval, or present availability for the compounds. Its strongest conclusions concern antibacterial activity in the experiments and models reported by the authors. A later paper summarizes this research line as including halicin and abaucin from the Drug Repurposing Hub as well as antibacterial compounds from the roughly 107-million-molecule ZINC15 collection; that retrospective summary does not change the specific counts reported for the 2020 ZINC15 experiments.
