The 2016 work described as “4D polymer printing” adds local chemical composition to the controls available in a three-dimensional polymer pattern. A University of Miami team used a flow-through microfluidic cell, an array of about 15,000 elastomeric tips and localized light to place polymer features, set their heights and change what each neighboring feature was made from.
This was a laboratory micropatterning demonstration, not a consumer 3D printer or a commercial manufacturing process. Under the reported conditions, the team produced features as small as 480 nanometres in diameter and polymer structures approaching 500 nanometres high.

What “4D” means in this polymer-printing experiment
In this study, “4D” does not mean that a printed object automatically changes shape over time, a common use of the term in popular descriptions of additive manufacturing. It refers to controlling an additional property of each patterned feature: its chemical composition.
A conventional three-dimensional description gives a feature a position and a height. The University of Miami method adds a local materials dimension. The operator can determine where a feature is placed, how tall it becomes and which monomer chemistry forms its polymer brush. Two features next to one another can therefore have different surface chemistries as well as different locations and dimensions.

That combination matters because a surface is experienced through both its geometry and its chemistry. Cells, proteins and other molecules can respond to the shape of a feature, the spacing between features and the chemical groups exposed at the surface. Ryan Chiechi, who led a nanofabrication research group at the University of Groningen, described the value as local control over how adjacent polymer features interact with cells, whose behavior can be strongly affected by both substrate chemistry and topology.
How the process creates chemically distinct features
A flow-through microfluidic cell
The reported apparatus places the polymerization reaction inside a flow-through microfluidic cell. Microfluidics confines small volumes of liquid and makes it possible to introduce successive ink solutions while the patterned substrate and tip array remain in the fabrication setup. The arrangement is important because the experiment depends on changing the monomer solution from one local pattern to the next.
The original paper describes localized light and thiol-acrylate photoinitiated brush polymerization. Light activates the photochemical reaction only where polymer growth is intended. The process grows polymer brushes from the substrate rather than depositing a preformed bulk object, allowing the chemistry at each patterned location to be selected through the solution flowing through the cell.

Chemistry World’s account of the work describes an array containing around 15,000 polydimethylsiloxane, or PDMS, pyramids. The tips were spaced 80 micrometres apart. Instead of addressing one point with a single probe, the array provided many potential writing locations in parallel.
Tip position determines where a feature is formed. Moving or positioning the array relative to the substrate therefore supplies the spatial part of the process. The elastomeric tips and the microfluidic cell work together to localize the reaction without requiring a high-energy beam.

Light, exposure and monomer selection
The experiment has three independent control ideas:
| Control | What it changes | Evidence from the reported setup |
|---|---|---|
| Tip position | The location of a polymer feature | The tip array was used to localize where individual features formed. |
| Exposure time | Polymerization and feature height | The original study reports that exposure time influenced polymer growth and height. |
| Monomer solution | The local chemical composition | Changing the monomer produced nearby patterns with different compositions. |
| Array layout | Parallel patterning opportunities | The reported array contained around 15,000 PDMS pyramids with 80 micrometre spacing. |
| Localized illumination | Where the photochemical reaction proceeds | Chemistry World reported sub-micrometre resolution without high-energy beams. |
These controls are related but not interchangeable. Moving a tip changes position, not the polymer’s identity. Increasing exposure can change growth and height, but it does not substitute for changing the monomer. To make neighboring regions chemically different, the team used successive ink solutions and optimized the photochemical polymerization conditions inside the reactor.

The fabrication sequence, step by step
The published description supports the following conceptual workflow. It explains the operating logic without implying that the apparatus is a ready-to-buy printer.
- Load a monomer-containing solution into the microfluidic cell. The cell supplies the reagents needed for thiol-acrylate photoinitiated brush polymerization.
- Position the elastomeric tip array over the substrate. The array’s tip positions define the locations at which polymer features can be written.
- Illuminate the selected reaction regions. Localized light initiates polymerization in the intended areas rather than across the entire substrate.
- Set the exposure conditions. Exposure time influences how far polymerization proceeds and therefore affects the resulting feature height.
- Change the ink solution for a different composition. Successive solutions allow nearby patterns to be made from different monomers and to expose different chemical functionality.
- Repeat the patterning sequence as needed. The combination of array position, exposure and monomer choice builds a surface in which geometry and chemistry vary from feature to feature.
The article does not present this sequence as a universal recipe. Flow rates, light intensity, initiator concentration, substrate preparation and other operating parameters were optimized for the particular laboratory reactor and polymer system.
What the 2016 experiment actually demonstrated
The quantitative results belong to the laboratory conditions reported by Liu, Zheng, Peurifoy, Kothari and Braunschweig in Polymer Chemistry in 2016. They should not be read as specifications for a current commercial printer.

| Reported quantity | What it describes | Qualification |
|---|---|---|
| 480 nm | Smallest feature diameter reported | Minimum diameter in the 2016 experimental study. |
| Nearly 500 nm | Polymer height approached | Height reached under the paper’s experimental conditions. |

Three-dimensional relief alone cannot specify how a surface binds a protein, presents a ligand or interacts with a cell. A chemically varied polymer pattern can provide those functions while the feature’s height and spacing provide physical cues.
The researchers and the Chemistry World report identified gene chips, protein arrays and stimulus-responsive surfaces as possible applications. In each case, the attraction is the ability to place chemically addressable regions close together while retaining control over their geometry.
| Possible use | Why the demonstrated control could matter | What the report establishes |
|---|---|---|
| Gene chips | Different surface chemistries could be patterned at defined locations for molecular capture and readout. | Named as a potential application, not shown as a completed product. |
| Protein arrays | Neighboring sites could present different binding environments with controlled feature shape. | Named as a potential application, not demonstrated as a commercial or clinical array. |
| Stimulus-responsive surfaces | Local polymer chemistry could be selected to respond differently to an external stimulus while topology supplies additional control. | Presented as a possible use in the 2016 report. |
| Engineered biological interfaces | Combining chemical and physical cues could help reproduce aspects of a cell’s surrounding surface. | Stated as a longer-term goal; the report says this remained a long way off. |
The distinction between a possible application and a demonstrated device is essential. The study did not report a finished gene chip, protein array, clinical instrument or commercial responsive surface.
What makes the method different from ordinary 3D printing
Consumer 3D printers generally select from a limited set of bulk materials and build objects at millimetre or larger scales. The Miami work instead patterned polymer brushes on a surface at sub-micrometre dimensions. Its central question was not how to make a large mechanical part, but how to give adjacent microscopic regions different chemical identities.

The process also differs from a single-beam writing system. The 15,000-tip array was designed for massively parallel localization, while the microfluidic cell supplied the changing chemistry. Chemistry World emphasized that the reported resolution was obtained without high-energy beams.
Calling this work “4D printing” can therefore mislead readers who expect a desktop machine, filament, vat resin or a shape-changing object. It is more precise to describe it as a specialized polymer-micropatterning strategy that combines spatial, height and chemical control.

The sources establish a research demonstration, not a product category. They do not establish commercial availability, pricing, production-scale throughput or present-day adoption. No retail printer, consumer accessory, replacement part or standard consumable is associated with the reported method.
The team’s longer-term ambition was to reproduce the architectural complexity and chemical properties of biological interfaces over larger areas. The Chemistry World report explicitly characterized that objective as still far away in 2016. Moving from a controlled laboratory pattern to a large, uniform surface would require the array, illumination and fluid chemistry to remain consistent across the expanded area.

How to read the result in context
The significance of the 2016 paper is architectural control. It shows that a polymer pattern can be designed so that position, height and chemical identity are selected locally in one fabrication concept. That is a different goal from maximizing the size, speed or mechanical strength of a printed object.
The work also illustrates why terminology needs qualification. “4D” captures the added materials dimension, but it does not by itself promise time-dependent motion, biological function or industrial readiness. The evidence supports a carefully controlled research platform whose potential lies in chemically patterned interfaces.
Adam B. Braunschweig summarized the motivation in the Chemistry World report: “We’re still a long way off, but that is the motivation for our work.” That remains the appropriate reading of the result: an important demonstration of local chemical and geometric control, with substantial development required before the idea could become a broadly deployed manufacturing technology.

4D polymer printing, as demonstrated by the University of Miami team, adds chemical composition to the spatial and height control of polymer micropatterning. A flow-through microfluidic cell, localized photochemistry and a roughly 15,000-tip PDMS array produced sub-micrometre features, including reported diameters down to 480 nanometres and heights approaching 500 nanometres. The method points toward chemically sophisticated biological interfaces, but the cited 2016 work remains a laboratory research platform rather than a commercial 3D printer.
