A gold nanogap is a nanoscale separation between plasmonic metal surfaces, usually at the junction of two nearby gold particles. Its purpose is often to concentrate electromagnetic energy into a local hotspot for surface-enhanced Raman scattering (SERS) or related sensing. A chiral plasmonic nanoparticle is defined by handedness: its shape, molecular environment, or particle arrangement interacts differently with left- and right-circularly polarized light.
They are not mutually exclusive categories. A chiral assembly can contain several interparticle gaps, and a nanogap can host chiral molecules whose optical response is enhanced. The correct choice depends on whether the experiment primarily needs a strong local field, a handed optical signal, or both.

What each term means
Gold nanogap: a geometry and a hotspot
A nanogap is the very small space between neighboring plasmonic features. In a common model, two gold nanoparticles sit close enough for their conduction electrons to oscillate as a coupled system. The junction between them can support a much stronger electromagnetic field than the incident light alone.

It does not, by itself, say anything about mirror-image symmetry, circular polarization, or molecular handedness. Two identical gold spheres separated along a straight line can form a powerful but achiral dimer.

Hotspot behavior is conditional. The incident wavelength must overlap the coupled plasmon resonance, the particle shape must support the desired mode, and the analyte must be located in the high-field region. A molecule several nanometers away from the junction may experience far less enhancement than one sitting directly in the active volume.
Chiral plasmonic nanoparticle: a handed optical structure
Chirality means that a structure cannot be superimposed on its mirror image. In plasmonics, the handedness can originate in several ways:
- An individual metal particle can have an intrinsically twisted or otherwise asymmetric shape.
- Chiral molecules can interact with an otherwise achiral gold particle and induce a handed optical response.
- Achiral particles can be arranged into a left- or right-handed assembly using molecular linkers, templates, or systems such as DNA origami.
Here, “chiral” describes the symmetry and optical response of the object or assembly, not the size of a gap. A chiral particle may have no narrow junction at all, while a chiral assembly may contain many nanogaps.
The central difference: spatial separation versus handedness
| Comparison axis | Gold nanogap | Chiral plasmonic nanoparticle or assembly |
|---|---|---|
| What the term describes | A nanoscale separation or junction between plasmonic metal features | Handed particle geometry, molecularly induced chirality, or a handed arrangement of particles |
| Main optical design aim | Localized electromagnetic-field enhancement at a coupled plasmon hotspot | A handed optical response, commonly differential extinction of circular polarizations |
| Typical measurements | Surface-enhanced Raman scattering and other hotspot-based spectroscopy or sensing | Circular-dichroism spectroscopy, enantiomer-sensitive analysis, and biomolecule sensing |
| Important controls | Gap width, particle shape, resonance matching, illumination, reproducibility, and analyte location | Source and handedness of chirality, assembly structure, wavelength, circular-dichroism convention, scattering, and stability |
| Can the features coexist? | Yes. A gap can be one junction in a chiral assembly. | Yes. A chiral structure can contain optically active interparticle gaps. |
This is a qualitative comparison, not a performance ranking. The available reviews do not report a standardized head-to-head experiment that would establish one platform as universally more sensitive or more useful.
How a gold nanogap creates a plasmonic hotspot
Coupled plasmon modes
When light drives the conduction electrons in two neighboring gold particles, the particles’ plasmon modes interact. The coupled mode can concentrate the electric field in the narrow junction. For Raman spectroscopy, the local field can increase both the excitation of a molecule and the radiation of its Raman signal, producing a large apparent SERS enhancement.
The effect depends on the full electromagnetic configuration rather than gap width alone. Particle diameter and shape, the dielectric environment, polarization direction, excitation wavelength, and the position and orientation of the analyte all matter. A nominally identical gap can produce different results if the particles are slightly different or the molecule sits outside the mode maximum.
Model-specific enhancement values
A 2019 Applied Sciences review discussed a calculated gold-dimer example in which the SERS enhancement was approximately 5 × 105 for a 10 nm gap and approximately 3 × 109 for a 2 nm gap. These are calculated values for a particular modeled geometry, not universal experimental specifications or guarantees for every gold nanogap.

The numbers illustrate why researchers try to reduce the separation: a smaller junction can greatly increase the classical electromagnetic field. They should not be used to predict the response of a different particle shape, substrate, wavelength, or fabrication method without a new calculation or measurement.
Why sub-nanometer gaps do not follow an unlimited classical trend
At separations below roughly 1 nm, quantum effects become important. Electrons can tunnel across the junction, changing the charge distribution and weakening or reshaping the classical field concentration. The simple assumption that every reduction in gap width produces another orders-of-magnitude increase therefore breaks down. Molecular layers, atomic-scale roughness, and chemical bonding can also determine what the junction actually is.
How chiral plasmonic nanoparticles produce a handed signal
Chirality sources and fabrication approaches
Gold chiral structures are commonly produced through wet-chemical, seed-mediated growth or through controlled assembly. Chiral molecules, linkers, and templates can bias the orientation of otherwise achiral particles. DNA-origami-based templates are one example of a method for arranging particles into a prescribed three-dimensional handed geometry.
Small changes in particle orientation, linker length, spacing, or twist can alter the optical activity of the assembly. That sensitivity is useful for sensing, but it also makes reproducibility and structural characterization essential. A sample that contains a mixture of left- and right-handed objects can show a reduced net signal even when each individual object is strongly chiral.

Circular dichroism and plasmonic circular dichroism
Circular dichroism (CD) is the difference in a sample’s interaction with left- and right-circularly polarized light. In a 2023 review, Probing Interactions between Chiral Plasmonic Nanoparticles and Biomolecules, the principle is stated as follows: “The difference in the extinction of left and right circularly polarized light through a sample is known as circular dichroism (CD), and the corresponding spectrum is an excellent tool for detecting interactions of nanoparticles with biomolecules.”
For plasmonic nanoparticles, the measured extinction can include both absorption and scattering. Scattering becomes especially relevant for larger particles and assemblies. A change in the CD spectrum can therefore reflect a structural change, a molecular binding event, a change in aggregation, or a change in the balance between absorption and scattering.
Why CD values cannot be compared casually
Different studies may report ellipticity, a CD intensity, or a dissymmetry factor. They may normalize by concentration, path length, extinction, or another quantity; measure a suspension, a film, or an immobilized assembly; and use different wavelength ranges and particle concentrations. Before comparing two reported values, check the definition, normalization, wavelength, sample state, and whether the signal is dominated by absorption, scattering, or both.

What each platform is normally used for
Nanogaps for SERS and local-field sensing
Nanogaps are attractive when the measurement benefits from placing a target molecule in a very small, intense electromagnetic mode. Typical goals include SERS, hotspot-based chemical identification, and sensing schemes in which the Raman signal changes when an analyte enters or leaves the junction.
The practical challenge is not merely making one spectacular junction. A useful substrate must produce enough equivalent junctions, with controlled dimensions and stable optical behavior, that measurements can be repeated. Fabrication variation, particle aggregation, contamination, and uncertain analyte placement can all make a nominally high-enhancement design unreliable.
Chiral structures for enantiomer-sensitive and biomolecular measurements
Chiral plasmonics is investigated for distinguishing or sensing molecular handedness, monitoring biomolecule interactions, and studying polarization-dependent optical effects. A molecule that binds to, reorganizes, or changes the environment around a chiral assembly can alter its CD spectrum.
These uses remain research directions in the cited reviews. They should not be presented as established clinical diagnostics or routine commercial tests without evidence for a specific assay, validation population, and instrument workflow.
When a design uses both chirality and nanogaps
The two concepts can reinforce one another. A helical or otherwise handed assembly may contain several narrow junctions, giving it both a chiroptical response and local electromagnetic hotspots. Conversely, a nanogap can be functionalized with a chiral molecule. The gap may then enhance a molecular signal while the surrounding structure determines how that signal couples to polarization.
In a combined design, there are two distinct questions:
- Does the junction provide a sufficiently strong and accessible local field for the target molecule?
- Does the complete particle geometry or molecular environment produce a measurable difference between the two circular polarizations?
A strong answer to one question does not guarantee a strong answer to the other. A symmetric dimer can have excellent SERS enhancement but negligible CD, while a visibly chiral assembly can show a clear CD spectrum without placing the analyte at the maximum SERS hotspot.

Choosing the right architecture for an experiment
Choose a nanogap-first design when the readout is local enhancement
Start with a nanogap when the primary signal is Raman intensity or another process that benefits from a concentrated near field. Define the target excitation wavelength, estimate the required resonance, and design the particle size and separation around that optical condition. Then determine how the analyte will be delivered to the junction; a gap that is inaccessible to the sample is not useful regardless of its simulated field.
Choose a chiral-first design when the readout is polarization contrast
Start with a chiral architecture when the experiment asks whether a structure or molecular interaction treats left- and right-circularly polarized light differently. Specify the intended handedness, the method for producing it, and the CD metric before collecting data. Include controls for achiral particles, opposite-handed assemblies when available, and concentration or aggregation changes that could alter scattering.
Choose a hybrid design when both signals answer the question
A hybrid platform is justified when the experiment needs a local spectroscopic enhancement and a polarization-dependent signature. Keep the two performance criteria separate during optimization. Measure the gap-dependent hotspot response and the CD response with controls that can distinguish a genuine chiral change from simple aggregation or concentration drift.

Gap-width uncertainty
“Two-nanometer gap” can mean different things depending on whether the value comes from an electron-microscopy image, a lithographic design, a molecular spacer, or an electromagnetic fit. Surface roughness and particle curvature can make the local separation vary across the junction. Report how the gap was defined and characterized rather than treating one nominal number as exact everywhere.
Hotspot occupancy
SERS enhancement is highly position-dependent. If analyte molecules adsorb randomly, the measured spectrum may be dominated by a small fraction that happen to occupy the junction. Surface chemistry, linker design, incubation time, and washing conditions can therefore affect reproducibility as much as the optical design.
Resonance mismatch
A narrow gap does not automatically place the coupled resonance at the laser wavelength or at the spectral region needed for a particular Raman band. Calculate or measure the extinction spectrum of the actual substrate, including its environment, before interpreting a weak signal as a failure of the nanogap concept.

Opposite-handed particles, incompletely assembled structures, or a large achiral background can cancel the net CD signal. Structural characterization should verify that the intended handed arrangement was produced and remains stable during the measurement.
Best Value
Scattering artifacts in CD
For larger particles and assemblies, scattering can contribute substantially to extinction. A CD feature should be interpreted alongside concentration, size distribution, absorption and scattering spectra, and the sample’s physical state. Comparing a suspension spectrum with a film spectrum without accounting for these differences can lead to incorrect conclusions.
Aggregation and environmental sensitivity
Salt concentration, pH, solvent composition, biomolecule binding, and storage time can change interparticle spacing or assembly structure. Such changes may shift a plasmon resonance, create new gaps, or alter the CD spectrum. Record the sample conditions at the time of measurement and use time-dependent controls when stability matters.
A practical workflow for comparing candidate structures
- Define the readout. Decide whether the primary observable is Raman enhancement, CD, a dissymmetry factor, fluorescence modification, or another signal.
- Specify the optical conditions. Record the excitation or probe wavelength, polarization state, incident angle if relevant, and the medium surrounding the particles.
- Map the structure. Measure particle size, shape, orientation, assembly state, and gap distribution using an appropriate structural method. Treat nominal synthesis dimensions as targets, not proof of the final geometry.
- Model or measure the optical response. For nanogaps, examine the coupled resonance and near-field distribution. For chiral structures, measure the CD spectrum and establish the sign convention for the reported handedness.
- Control the chemistry. Verify where the analyte binds and whether the surface treatment changes the gap, the assembly, or the refractive index.
- Separate signal sources. Use achiral and no-analyte controls, opposite-handed controls where possible, and measurements that distinguish absorption from scattering.
- Report uncertainty. State the gap definition, particle-size distribution, concentration, wavelength, normalization, sample state, and whether an enhancement value is calculated or experimentally measured.
How to read claims about performance
Enhancement factors and CD amplitudes are not interchangeable currencies. A calculated SERS enhancement of 3 × 109 for one modeled 2 nm dimer does not predict the CD of a chiral assembly, and a large CD signal does not prove that a molecule experiences a high SERS hotspot. They arise from different observables and are influenced by different parts of the structure.

Likewise, a higher signal is not always a better sensor. A design with extreme local enhancement but poor junction-to-junction uniformity may be less useful for quantitative measurements than a lower-field substrate with controlled and repeatable gaps. A chiral assembly with a strong CD response may still be unsuitable for enantiomer analysis if the response is not selective, stable, or calibrated against the target concentration.
What is established—and what remains application-specific
The reviewed literature establishes the conceptual distinction between nanogap geometry and plasmonic chirality, the use of coupled gaps for localized fields, and the use of CD to study handed optical responses and biomolecule interactions. It also documents synthesis and assembly strategies for gold chiral structures and the importance of quantum effects at sub-nanometer separations.
It does not establish a universal winner, a single best gap size, a cross-platform sensitivity score, or routine clinical deployment. The 2025 review surveys synthesis methods and application directions but does not by itself demonstrate commercialization. Any claim about a particular assay, instrument, or product must therefore be tied to that assay’s own validation data.
Bottom line for experimental design
Use the word nanogap when the decisive feature is a nanoscale junction that concentrates a local electromagnetic field. Use chiral plasmonic nanoparticle when the decisive feature is handed geometry, handed molecular interaction, or handed particle arrangement that creates a polarization-dependent optical response. If your structure has both, describe and test both properties separately. That distinction prevents a common category error: treating a powerful hotspot as automatically chiral, or treating a strong CD spectrum as proof of superior local-field enhancement.
