A 2025 Friends of Murdoch Children’s Research Institute (MCRI) gathering in New York introduced supporters to Decoding Broken Hearts, a global MCRI–Gladstone Institutes program developing precision treatments for childhood heart disease. The program’s work now includes a 2026 preclinical result in which a single injection delivering a healthy ALPK3 gene copy restored heart-muscle function in laboratory-grown patient tissue and mouse models.

The therapy has not been tested in children and is not available to families. Any benefit for patients remains contingent on further studies showing that the approach can be translated safely and effectively to people.

What happened at the New York event

MCRI held its first “Friends of MCRI” event in New York City in April 2025 at Old Mates, a venue co-founded by Nicholas Stone, a member of MCRI’s Global Advisory Board. The gathering brought together Australian expatriates and their networks for a supporter-focused discussion of congenital heart disease and the institute’s research priorities.

The program included a fireside conversation between Stone and Professor Enzo Porrello, who leads MCRI’s Decoding Broken Hearts initiative. The event was intended to explain why new treatment strategies are needed and how supporters could help advance them.

MCRI’s 2025 event article said congenital heart disease affects 1 in 100 live births, equating to one child born with a heart defect every 15 minutes in the United States. That statistic is MCRI’s published estimate and is presented here with its 2025 attribution.

How Decoding Broken Hearts approaches childhood heart disease

Decoding Broken Hearts is a global collaboration between MCRI and Gladstone Institutes. It combines stem-cell technologies, computational analysis and artificial intelligence to study heart disease in human tissue and identify potential precision therapies.

Thousands of miniature human heart tissues

MCRI says the platform can produce thousands of miniature human heart tissues from stem cells. These models allow researchers to examine how disease-associated genetic changes affect human heart muscle and to test candidate treatments in a controlled laboratory setting.

Why the program focuses on new options

Children whose heart disease progresses toward heart failure can face limited treatment choices. A transplant may be constrained by the availability of donor hearts and requires lifelong immunosuppression to reduce the risk of rejection. Decoding Broken Hearts is designed to find treatments that address the underlying biology instead of relying only on late-stage interventions.

The New York event article reported an event-era funding request of US$4 million to advance the work. That figure described the need presented in 2025; it should not be read as a current campaign balance or a still-active total without checking MCRI’s latest information.

What the 2026 ALPK3 study found

In a release dated 20 August 2026, MCRI described a world-first preclinical study led by the institute and published in Nature Cardiovascular Research. Researchers delivered a healthy copy of the ALPK3 gene in a single injection.

The disease model was cardiomyopathy, in which the heart becomes enlarged and pumps weakly or irregularly. MCRI reported that the treatment reversed heart-muscle disease in two experimental settings:

  • Patient-derived heart tissue: laboratory-grown human heart tissue made from patients with the disease showed restored function after ALPK3 gene replacement.
  • Mouse models: mice with the modeled disease also showed reversal after the single injection.

Dr James McNamara, MCRI Team Leader in Heart Disease, described the potential reach of the result:

“If this success translates to patients, the gene therapy could become the first targeted treatment for a range of inherited heart diseases, offering families a future without progressive heart failure or the eventual need for a heart transplant.”

The conditional wording matters. The experiments were preclinical, and the cited material does not report treatment of children or any clinical trial.

What the evidence does and does not establish

Evidence setting Reported result Evidence stage What remains unresolved
Laboratory-grown patient heart tissue Healthy ALPK3 gene delivery reversed heart-muscle disease in the modeled tissue Preclinical human-tissue study Whether the effect, dose and safety translate to children
Mouse models A single injection reversed the modeled cardiomyopathy Preclinical animal study Human delivery, immune responses, durability and clinical benefit
MYH7 and TTN truncating variants MCRI said the approach may be adaptable to these cardiomyopathies Prospective extension No results for these targets were reported in the cited release

The study therefore supports a promising research direction, not a proven treatment. It also does not establish a head-to-head advantage over other gene therapies, medicines or transplant strategies. The relevant comparisons for future studies will include the biological model used, the genetic target, clinical versus preclinical evidence, delivery and safety, the number of mutations that can be addressed, and whether treatment reduces the need for transplantation or lifelong therapy.

Why the result matters beyond one gene

Congenital heart disease includes structural defects present at birth, while inherited cardiomyopathies can damage the heart muscle over time. Both can lead to serious disease in childhood, but they are not the same condition and should not be treated as interchangeable.

An ALPK3-focused therapy would initially apply only to disease caused by relevant ALPK3 changes. MCRI’s mention of MYH7 and TTN indicates a possible platform strategy, not proof that one injection already treats every inherited cardiomyopathy. Broader use would require separate validation for each mutation and clinical evidence in the appropriate patients.

How the New York gathering fits MCRI’s continuing US work

The 2025 New York event was part of a wider effort to connect MCRI’s research with supporters in the United States. In April 2026, MCRI announced that it had joined Anthropic’s AI for Science program. The institute said in-kind support would help a stem-cell medicine pilot identify therapeutic targets for heart disease, building on Porrello’s ability to produce human heart tissue at scale.

On 5 October 2026, MCRI and Gladstone held an inaugural symposium in San Francisco. More than 80 guests heard about stem-cell models, AI tools and novel therapies for childhood heart disease and heart failure. The symposium shows that the New York gathering was an early supporter event within continuing scientific and philanthropic outreach, rather than the announcement of a finished clinical product.

Can families receive the gene therapy now?

No. The cited MCRI material establishes laboratory and mouse results only. It does not establish clinical availability, a completed human trial, an approved dose or a pathway for families to request treatment. Parents should not interpret the release as a reason to delay established care or pursue unapproved gene-therapy products.

Future translation would ordinarily require additional testing of delivery, safety, immune reactions, durability and clinical benefit before treatment could be evaluated in children. The timing and design of those steps are not provided in the cited announcements.

How readers can support the work

Readers interested in helping should check MCRI’s current official donation and partnership routes rather than relying on the US$4 million figure reported for the 2025 event. A contribution or research partnership should specify which program it supports, whether the commitment is recurring, and how the institute describes the work’s current stage.

For families affected by congenital heart disease or inherited cardiomyopathy, the practical step today is to continue working with qualified pediatric cardiology teams. The ALPK3 result is an important preclinical signal, but it is not yet a treatment option.
