The “bloody solution” was hemoglobin acting as an electron sink—not a test of blood. The work demonstrated DNA sensing and restriction-enzyme detection on an electrode, not a validated diagnostic performed directly on whole blood.
What the “bloody solution” means
The phrase comes from a 2012 Chemistry World headline. Hemoglobin—the oxygen-carrying protein in red blood cells—was used as a component of an electrochemical DNA assay. It was not evidence that adding a patient’s blood makes an ordinary DNA sensor work.

In their PNAS paper, Catrina G. Pheeney, Luis F. Guerra, and Jacqueline K. Barton describe “the use of hemoglobin as a robust and effective electron sink for electrocatalysis in DNA sensing on low-density DNA films.” The phrase describes a laboratory signal-amplification method, not a clinical blood test.

How hemoglobin amplified the DNA signal
The researchers attached a redox reporter, methylene blue (MB), to DNA through a flexible alkyl linkage and placed the DNA on an electrode. The DNA base stack mediates charge transport between the electrode and the reporter. When the reporter is reduced, hemoglobin accepts electrons and enables catalytic turnover, amplifying the electrochemical signal.

The authors report that protein shielding around hemoglobin’s heme redox center minimizes direct reduction at the electrode surface. That matters because the intended signal is tied to charge transport through the DNA, rather than an unrelated direct electrode reaction.
What the experiment demonstrated
- Signal amplification: The paper reports at least a 100-fold improvement in sensitivity for methylene-blue DNA reduction with hemoglobin electrocatalysis.
- Small DNA samples: The authors report sensitivity sufficient for femtomole DNA sampling.
- Mismatch detection: A single cytosine-adenine (CA) mismatch consistently suppressed the redox signal. The authors interpret this as evidence that both amplified and unamplified signals arise through DNA-mediated charge transport.
- Restriction-enzyme detection: The study also demonstrated detection of restriction-enzyme activity.
The approach addressed a limitation of earlier electrocatalytic DNA-sensor strategies, which the authors say required high-density DNA films and strict assembly and passivation. Their hemoglobin approach worked on low-density DNA films. The paper describes the electrocatalysis as stable to pH and temperature variation, but that statement does not by itself establish a general-purpose assay protocol; performance depends on the experimental conditions.

Why this was not a whole-blood diagnostic
The experiments establish signal amplification on DNA-modified electrodes, with mismatch and restriction-enzyme demonstrations. They do not establish clinical diagnostic performance, patient results, regulatory clearance, or routine testing directly from whole blood. The sources report no clinical cohort or diagnostic sensitivity and specificity.
In a 2012 Chemistry World report, Alon Gorodetsky of the University of California, Irvine, said the result “raises the possibility” of detecting biomarkers such as RNA, DNA, and proteins directly from whole blood. That was a prospective comment about a possible application, not a sample type demonstrated in the study.

What researchers might take from the result
The work is relevant as a method for amplifying electrochemical signals from DNA on low-density films. Its reported mismatch response and restriction-enzyme detection also point to ways DNA-mediated charge transport could be used to study interactions involving DNA and proteins. Emil Palecek, quoted by Chemistry World, said: “The reported 100-fold increase of sensitivity combined with low density DNA films opens the door for a new type of DNA–protein interaction analysis.”
Reproducing this kind of experiment requires laboratory electrochemical measurement and appropriately prepared DNA-modified electrodes; the paper identifies materials including hemoglobin, methylene-blue DNA probes, and gold electrodes. An electrochemical workstation or USB potentiostat is equipment for laboratory measurement, not a turnkey blood test. The cited sources do not identify a marketed sensor based on the study.
