Xbox describes E-Day as a new mainline entry set earlier in the series timeline: it takes place fourteen years before the original Gears of War. The developers also frame the Locust as a new threat encountered for the first time by Marcus, Dom and those around them. That makes it reasonable to expect newcomers to understand the central premise alongside the leads. It is an inference from the story’s framing, not an explicit guarantee that prior knowledge could never help.

What earlier games add
Returning players may recognize the significance of Marcus and Dom’s relationship and have more context for the history connecting them. Xbox Wire notes that Dom’s brother Carlos died alongside Marcus, and positions E-Day at the beginning of the pair’s bond. That background can make the characters’ early relationship more resonant, but the official material does not say you must know it to follow E-Day.
