Yes, Windows updates help protect against viruses by fixing security weaknesses malware could exploit, and Windows Update can also deliver threat information for Microsoft Defender scans. But an update is not an antivirus scan: it does not, by itself, establish that a PC is free of malware or remove an infection.
What Windows updates protect against
Windows security updates address vulnerabilities in Windows. Closing a weakness can prevent malware from exploiting that particular opening; Microsoft says security updates can help prevent virus and other malware attacks by closing possible security holes (Microsoft Support: Protect my PC from viruses).

That protection is preventive and specific to the weaknesses addressed by the fixes. It does not mean Windows Update has examined every file, checked every program, or cleaned an already infected PC. Does Windows Update update Microsoft Defender?
Windows Update can deliver Microsoft Defender security intelligence: information about current threats that Defender uses when a scan runs. Current intelligence helps scans recognize newer threats, but receiving it does not mean a scan has run or that anything has been removed (Microsoft Support: Stay protected with Windows Security).

In Windows 10 and Windows 11, you can request updated intelligence from Windows Security > Virus & threat protection > Protection updates > Check for updates. The label may vary by Windows version.
Updates, antivirus, firewall, and web protection do different jobs

Windows Security is the Windows 10 and Windows 11 app for viewing Microsoft Defender Antivirus and other protection settings. “Microsoft Defender” can refer to different products. Microsoft Defender Antivirus is built into Windows; the Microsoft Defender app included with some Microsoft 365 subscriptions is distinct from that antivirus (Microsoft Support).

- Open Windows Security > Virus & threat protection and review the current threat and protection information.
- Choose a Quick scan for an initial check. Use Full scan for a broader check or Custom scan to check a chosen location. Microsoft describes these scan options in its Windows Security guidance.
- If concern remains, choose Microsoft Defender Antivirus (offline scan). It restarts the PC into the Windows Recovery Environment and scans outside the normal Windows session. Save open work first.
- Under Protection updates, select Check for updates to request current security intelligence.
A scan result is useful evidence, but no update or single scan proves that every possible threat has been found. If Windows Security shows that another compatible antivirus is active, that product may turn off Microsoft Defender Antivirus automatically. Check Windows Security to confirm which antivirus is providing active protection rather than assuming both are scanning.
