Could Perseanol Become an Eco-Friendly Insecticide? What the 2019 Synthesis Shows

Not yet. A 2019 chemical synthesis of (+)-perseanol gives researchers a way to study a natural product with reported insecticidal activity, but it does not establish a safe, practical or approved pesticide. The result revives a research avenue around ryanodane compounds—not a ready-to-use alternative to conventional insecticides.

What is perseanol?

(+)-Perseanol is an isoryanodane diterpene isolated from the tropical shrub Persea indica. It has reported antifeedant and insecticidal properties, meaning it can discourage feeding and affect insects. The 2019 Nature paper describes the compound as structurally related to (+)-ryanodine, a high-affinity ligand and modulator of ryanodine receptors.

Ryanodine receptors regulate calcium release in cells, and this process also occurs in mammals. The Nature paper notes preliminary evidence that ryanodane and isoryanodane compounds without a pyrrole-2-carboxylate ester—including perseanol—may act selectively in insects. That is an early indication, not a demonstrated safety profile for people, wildlife or ecosystems.

What did the 2019 synthesis achieve?

Arthur Han, Yujia Tao and Sarah E. Reisman reported the synthesis of (+)-perseanol in a paper published in Nature on 25 September 2019. Their route took 16 steps from commercially available (R)-pulegone. A two-step annulation rapidly assembled the molecule’s tetracyclic core from accessible cyclopentyl building blocks; the authors also used convergent fragment coupling and strategic oxidation tactics. Garden Safe Insecticidal Soap Insect Killer Spray, 32 Oz
  • READY-TO-USE CONTACT SPRAY: No mixing required. Thoroughly spray insecticide on all plant parts, including the underside of leaves. Pests must be sprayed directly to achieve control
  • FOR ORGANIC GARDENING: This product can be used indoors, outdoors and in greenhouses on vegetables, fruit trees, ornamentals, shrubs, flowers and gardens
  • USE UP TO DAY OF HARVEST: This product may be applied to edibles up to and including the day of harvest
  • KILLS BUGS THROUGH CONTACT: Controls aphids, mealybugs, mites, leafhoppers, psyllids, scale insects, thrips, whiteflies and other listed pests.
  • CONTAINS FATTY ACID SALTS: The formula contains specifically selected soaps (fatty acid salts)

This is a total-synthesis result: it provides a laboratory route to a complex natural product so its chemistry and biology can be studied. The paper does not establish manufacturing cost, agricultural-scale production or commercial feasibility. Chemistry World reported that the molecule’s complexity makes it unsuitable as an agrochemical. Read the Chemistry World report.

How might it affect insects?

The relationship to ryanodine makes ryanodine receptors a plausible line of investigation, but the exact mechanism was still an open question in the contemporaneous reporting. Reisman described perseanol as a probe for finding out whether it acts on that receptor or has insecticidal properties through another mechanism. The report said biological testing was beginning; it does not establish what later tests found.

Sevin Insect Killer Spray, Ready-to-Use 2, 1 Gallon
  • Plant insecticide to protect your roses and edibles from listed damaging pests
  • This insecticide sprayer won't harm plants or blooms
  • Kills on contact - This plant bug spray kills over 700 listed insects
  • This garden insect spray also kills ants, Japanese beetles, whiteflies, aphids and many more pests
  • Simply shake and spray this highly effective plant bug killer

That distinction matters: observing insecticidal activity does not by itself identify a molecular target, show how reliably the effect occurs, or establish whether the compound harms non-target organisms.

Does this revive an eco-friendly pesticide class?

It may renew scientific interest in a related family of natural products and help researchers investigate alternative modes of action, potentially including approaches relevant to insecticide resistance. It does not show that perseanol itself is an eco-friendly replacement. The available evidence does not establish comparative efficacy, resistance performance, environmental persistence, effects on beneficial species, mammalian safety or regulatory approval.

Chemistry World described ryanodine as a once-popular pesticide whose use declined as cheaper synthetic insecticides became available. The same report said ryanodine production reached 200 tonnes per year in the early 1950s; that historical figure is reported by the article, not independently verified here against an original production record. It also cited annual sales exceeding $1 billion for synthetic insecticides acting in a similar way. That is 2019 market context, not a current market estimate or evidence that perseanol can compete with those products.

What the result means—and what it does not

  • Established by the synthesis: researchers reported a 16-step route to (+)-perseanol from commercially available (R)-pulegone.
  • Reported biological properties: the compound has antifeedant and insecticidal activity; possible selectivity for insects is preliminary.
  • Still unresolved in the cited contemporaneous coverage: whether perseanol acts through ryanodine receptors, how selective or effective it is in relevant settings, and whether it has a viable path to use.
  • Not demonstrated: a market-ready pesticide, broad environmental safety, commercial production or regulatory approval.
