Could Nanopore Tests Identify Misfolded Proteins in Alzheimer’s and Parkinson’s?

Nanopore methods have detected protein targets linked to Alzheimer’s and Parkinson’s in separate research studies, but there is no established combined clinical test in the evidence described here. The experiments differ in their targets, samples and techniques: some examine α-synuclein oligomers relevant to Parkinson’s, while another measures Alzheimer’s-related Aβ and Tau biomarkers. These are promising laboratory approaches, not validated routine diagnostics or home tests.

What a nanopore test does

A nanopore is a tiny opening through which molecules pass. Researchers can study changes in the electrical signal as a molecule or tagged molecular carrier moves through the pore. In protein research, this can help detect or characterize targets that are difficult to distinguish in a complex biological sample.

The challenge is selectivity: target proteins may be scarce and vary in size and shape, while other molecules in a sample can contribute background. Amyloid particles also take multiple forms and can interconvert, complicating measurement. Reviews describe nanopore sensing as an emerging analytical approach, with limitations still to address (2018 review; 2023 Parkinson’s study).

What the Alzheimer’s and Parkinson’s studies found

The studies investigate different targets and are not head-to-head comparisons. Their results should not be read as evidence of one test that diagnoses both diseases.

Study and target Method and sample What it reports
2023 study of misfolded protein oligomers, including α-synuclein Solid-state nanopores with multiplexed DNA barcoding; the study examines α-synuclein oligomers in the presence of small-molecule inhibitors. A single-molecule method to detect and characterize oligomeric species. The paper describes its approach as “a single-molecule approach for the detection and quantification of oligomeric species.” This is a research method, not a clinical diagnosis. (JACS, published November 16, 2023)
2023 Parkinson’s study of α-synuclein oligomers Nanopore sensing with aptamer-modified DNA carriers; clinical samples from Parkinson’s patients and healthy controls. Reports detection of α-synuclein oligomers and differentiation between the patient and control cohorts. Cohort differentiation in a research study does not establish validated routine diagnostic performance. (PubMed record)
2025 Alzheimer’s biomarker study Label-free nanopore detection of Aβ42, Aβ40, APP(669–711) and Tau-related targets in biological samples; includes CSF and Alzheimer’s mouse models. Reports detection of Aβ42 in cerebrospinal fluid and age-dependent Aβ changes in mouse models. Animal-model findings do not establish accuracy or clinical usefulness in people. (PubMed record)

How to interpret the reported detection limits

Liu and colleagues’ 2025 Alzheimer’s biomarker study reports analytical detection figures in serum: 2.1 pM for Aβ42, 1.5 pM for APP(669–711), and 627 fM for Aβ40. These are detection limits reported for that study’s analytical method; they are not clinical sensitivity or specificity figures, and they do not demonstrate that the method is an approved or routinely available diagnostic. The study also reports comparison with ELISA, but that does not by itself establish clinical diagnostic utility.

What remains unproven

  • A combined Alzheimer’s–Parkinson’s test: The cited studies examine distinct targets and methods; they do not establish a single test that diagnoses both diseases.
  • Routine clinical availability or approval: The cited sources do not establish regulatory clearance or routine clinical use.
  • Screening accuracy: Analytical detection and differentiation between study cohorts are not enough to establish performance as a validated screening tool.
  • Home use: The studies describe experimental laboratory methods, not consumer tests.

For a diagnostic claim, researchers would need evidence that the approach reliably identifies disease in the intended population and setting. The results described here support continued investigation of nanopore sensing, but do not show that it can replace established clinical evaluation.
