A 2025 preclinical study tested a CRISPR-Cas9 strategy designed to disrupt cancer-linked KRAS mutations in lung cancer models. The results included reduced cancer-cell viability and slower tumour growth in the models studied, but they do not show that the approach works in people or is ready to treat patients. The study is a plausible match for the supplied headline, although that headline’s original source could not be confirmed.
What the study tested
Published in Nature Communications on 1 September 2025, “High-fidelity Cas9-mediated targeting of KRAS driver mutations restrains lung cancer in preclinical models” examined gene editing aimed at two KRAS mutations: G12C and G12D. The work focused on non-small-cell lung cancer and used cell cultures, patient-derived organoid models and preclinical tumour models. Read the Nature Communications paper.

KRAS mutations can help drive cancer growth. The paper’s introduction cites earlier literature estimating that about 32% of lung adenocarcinomas have KRAS mutations. It also cites an estimate of about 88% for pancreatic adenocarcinomas. Those figures provide context about KRAS, not evidence that this study’s approach works against pancreatic cancer.
How mutation-directed CRISPR is meant to work
Unlike a drug that blocks a protein after a cell has made it, this strategy aims to edit the DNA instructions for mutant KRAS. The researchers paired high-fidelity Cas9—a gene-editing enzyme selected to reduce unintended cutting—with guide RNAs designed to recognise the G12C or G12D mutant sequence rather than wild-type KRAS.

The aim is to disrupt the cancer-associated version of the gene while sparing the normal version. The study reports mutant-specific editing in the models it tested; it does not establish that healthy tissue would be unharmed in a person. Targeted therapies generally act on specific molecular features involved in cancer growth, and biomarker testing can help guide the use of some such treatments. This experimental gene-editing approach should not be confused with an approved targeted therapy. The National Cancer Institute explains targeted cancer therapies.
What the results do—and do not—show
In the tested cell and organoid models, the researchers reported mutant-specific editing and reduced viability. In preclinical tumour models, they reported tumour-growth suppression after delivering the editing tools with adenoviral particles. These findings support further investigation, but they are not clinical evidence: the study did not show that patients lived longer, that tumours were cured, or that the strategy is safe and effective in people.

The authors also reported activity in some preclinical settings involving resistance to sotorasib, a drug that inhibits the KRAS G12C protein. The two approaches act at different levels: sotorasib targets the protein, while CRISPR aims to disrupt mutant KRAS DNA. The reported results do not mean gene editing overcomes every form of resistance, is superior to sotorasib in patients, or should replace established care.
Why delivery and safety remain major hurdles
Editing tools must reach enough tumour cells, at the right place and in a usable form. The study used adenoviral particles for in vivo delivery, but a result in a preclinical model does not prove that the same delivery method would work reliably or safely in people. Unintended edits, effects on normal tissue and the durability of any effect also require careful assessment.

GENyO’s summary describes the work as proof of concept and says delivery needs further optimisation before clinical trials can be considered. Study lead Pedro P. Medina cautioned: “It is essential to emphasise that we are not talking about a therapy that is ready for patients. The evidence here concerns KRAS G12C and G12D in preclinical non-small-cell lung cancer models. It does not establish benefit for people with lung cancer, for other KRAS mutations, or for patients with pancreatic or other cancers. The prevalence of KRAS mutations in another cancer type is not proof that this particular intervention will work there.

This is not a treatment patients can currently seek based on the reported study. The available sources do not establish a clinical trial or patient availability for this approach. Anyone making treatment decisions should discuss their diagnosis and options with their oncology team; experimental gene editing should not be treated as a substitute for prescribed care.
