Biological-age tests estimate different aspects of aging; a score change alone does not show that you have reversed aging, improved your health, or extended your life. The safer approach is to focus on established health measures—not chasing a test result or trying extreme diets and unproven products.
What “biological age” measures—and what it does not
Biological age is a broad research concept, not one universally agreed measurement. The NIH explains that it refers to the accumulation of molecular damage and physiological changes over time. Aging clocks use biomarkers to estimate aspects of that process, but different clocks can measure different things.

- An age estimate compares a person’s measurements with patterns seen across people of different chronological ages.
- A pace-of-aging score estimates how quickly aging-related changes are occurring. DunedinPACE, for example, was developed from DNA-methylation patterns associated with changes in 19 clinical biomarkers.
Faster DunedinPACE scores have been associated with worse health, chronic disease, and earlier death in other cohorts. That predictive association does not show that lowering the score itself improves health or extends life. The Dunedin Study followed more than 1,000 people born in New Zealand in 1972–73, with comprehensive evaluations at four points between ages 26 and 45; this work informed development of the measure. An overall score can also obscure differences between organs and tissues.
The National Institute on Aging (NIA) notes that DNA-methylation estimates can help predict chronic disease, cognitive impairment, functional limitations, and mortality in older adults. But demographic, socioeconomic, mental-health, and behavioral factors are comparable predictors and are often more robust. A clock score is not a diagnosis or a stand-alone reason to change medication, screening, or other treatment.

Can biological age be reversed?
There is no established evidence that lowering a commercial test score reverses aging in the everyday sense of restoring youth, preventing disease, or lengthening life. Scores may move because a measure responds to changes in health or stress, but a surrogate measure is not the same as a meaningful health outcome.
Short-term fluctuations illustrate the distinction. In a study summarized by the NIA, stress increased DNA-methylation estimates of biological age in studied humans and mice; the estimates decreased after the stress resolved. The long-term significance of such changes is uncertain. A lower reading after a stressful period, therefore, is not proof of lasting rejuvenation.

What the calorie-restriction trial actually found
The strongest specific intervention finding in the cited evidence comes from CALERIE, a randomized trial involving more than 200 healthy middle-aged volunteers. Participants assigned to calorie restriction were challenged to cut daily intake by 25% over two years and received dietary and behavioral strategies. On average, they reduced intake by 12.5% and lost 10% of body weight, mostly fat, according to the NIH summary.
The NIA’s summary of the DNA-methylation analysis reports that about a 12% calorie reduction over two years was associated with a 2–3% slowing in pace of aging on the measure studied. Importantly, DNA-methylation clock estimates of biological age did not change. This is a finding from a specific measure and selected trial population—not an individual target or a promise that a diet will lower your age score.

The NIH overview also reports a small decline in muscle tissue during the intervention, without a significant decline in muscle strength. That result does not establish safety for people with different ages, health conditions, nutritional needs, or medications. The cited sources do not provide an individualized safe calorie target. They also say larger trials with longer follow-up are needed to determine whether the pace-of-aging change leads to long-term health benefits or a longer lifespan.
These results do not justify starting restrictive dieting to change a test score. If you are considering a major dietary change, discuss your circumstances with a qualified clinician; the evidence cited here does not establish who should restrict calories or what level is safe for an individual.

What you can do safely to support health
The NIH overview on slowing aging recommends familiar actions for healthy aging. They can support health and reduce known risks, but they are not a guaranteed age-reversal protocol and may not change a commercial clock score.
- Be physically active and follow a healthy eating pattern suited to your needs.
- Avoid smoking, maintain a healthy weight where possible, and get good sleep.
- Receive recommended vaccines and age-appropriate preventive cancer screenings.
- Work with a clinician to treat known risk factors, including hypertension and high cholesterol.
These actions have health value independent of whether a biological-age test registers a change. Do not use a clock result alone to decide whether you need a screening or treatment.

How to interpret an aging-clock test
Before treating a result as meaningful, find out what the test estimates and what decisions it can actually support. The official sources summarized here do not compare consumer testing services, establish how repeatable their scores are, or show that changing a consumer score improves patient outcomes.
- Identify the measure: Is it an age estimate or a pace-of-aging score? Which biomarkers or tissue does it use?
- Check what has been validated: Association with health outcomes can make a measure informative without proving that changing it changes those outcomes.
- Put a change in context: A score may vary by clock, tissue, and time; stress-related changes can be transient.
- Keep clinical decisions clinical: Do not change medication, screening, or treatment based only on an aging-clock result. Discuss health concerns with a qualified clinician.
Why “age reversal” shortcuts are not a safe bet
Human evidence cited here does not establish supplements, extreme diets, prescription drugs for people without an appropriate indication, injections, or plasma products as safe ways for consumers to lower biological age. The NIA warns that many supplements and related products claim to “turn back the years” without scientific evidence. A score change or marketing promise is not evidence that a product improves health or lengthens life.
