Microneedles and nanoneedles are not simply smaller versions of an ordinary injection needle. Microneedles are micron-scale projections designed to get medicines past the skin’s outer barrier; nanoneedles are an emerging research approach to navigating biological membranes. Some microneedle designs are studied with nanoscale medicines, but that combination—and nanorods in particular—should not be mistaken for a routinely approved treatment. Whether any device is ready for patients depends on the specific product and its evidence.

Why change how a medicine crosses the skin?

A conventional hypodermic needle places a dose through the skin. Transdermal delivery aims to get a medicine through the skin without using that familiar injection route, but intact skin’s outer barrier blocks many substances. Microneedles are one way researchers try to cross or disrupt that barrier so a medicine can reach tissue beneath it.

The approaches differ in more than size. They may target different tissues, carry different kinds of payloads, and release them in different ways. A device’s scale alone does not establish what it delivers, how well it works, or whether it is available as a treatment. Reviews describe several microneedle designs and research into using them with nanoscale formulations (microneedles for transdermal delivery; microneedle-mediated nanomedicine).

How do microneedles deliver medicine?

Microneedles are micron-scale projections intended to pass the skin’s outer barrier, often without reaching deeper structures. Their design determines where the medicine sits and how it is released. The main formats are solid, coated, dissolving, and hollow.

Design How it delivers What the design means
Solid The projections create tiny pathways through the outer skin barrier; medicine is then applied to the treated area. Delivery depends on the pathways and the formulation applied afterward.
Coated Medicine is carried on the projections’ surface and deposited as they enter the skin. The coating has to hold the intended payload and release it during application.
Dissolving The projections contain medicine and dissolve after insertion, releasing their payload. The material and formulation affect how the projections dissolve and release the dose.
Hollow The projections contain a channel through which a liquid formulation can be delivered. The design must accommodate a fluid payload and its passage through the device.

These are distinct delivery mechanisms, not interchangeable labels for one device. A suitable design depends on the payload, intended tissue, material, release requirements, and application method. Reviews of transdermal and non-transdermal microneedles discuss a range of designs and uses; they do not establish one type as best for every medicine (transdermal microneedle review; non-transdermal microneedle review).

What does “microneedle plus nanomedicine” mean?

It describes a combination: a microneedle device helps deliver a formulation that uses nanoscale materials or structures. The device and the formulation each play a role. The microneedle may help the payload get past the skin barrier, while the formulation’s properties affect how the medicine is carried and released. A review of microneedle-mediated nanomedicine describes this as a research and development strategy, not a blanket clinical capability (2024 review).

Combining the two does not remove the need to assess either one. Developers must characterize the finished formulation and device, understand how the payload is released, and evaluate safety, effectiveness, and manufacturing for the particular product. A result for one material, medicine, or device cannot automatically be generalized to another.

Are nanoneedles or nanorods established treatments?

Nanoneedles are an emerging research area, with work exploring how nanoscale or nanostructured systems might navigate biological membranes and support intracellular delivery. That is a different concept from a conventional injection needle and does not mean nanoneedles are established therapies. A 2023 review surveys this research area, but the existence of research is not evidence that a particular product is approved or routinely used (review of nanoneedles in drug delivery).

“Nanorod” describes a rod-like nanoscale structure; it is not, by itself, the name of an established drug-delivery treatment. The reviews cited here address nanoneedles and microneedle-mediated nanomedicine, not a verified clinical nanorod delivery product. Claims about a particular nanorod system therefore need product-specific evidence rather than an inference from the word “nano.”

What does the evidence show—and what does it not show?

A 2025 systematic review of preclinical studies on dissolving microneedles for nucleic-acid delivery reported that 83% of the included studies showed equivalent or superior efficacy compared with their comparator administration routes (2025 systematic review). That percentage is a share of studies in a preclinical evidence synthesis. It is not a clinical response rate, the probability that a patient will benefit, or a general estimate of microneedle effectiveness.

Preclinical findings can justify further study, but they do not establish human outcomes. A review of microneedle-enabled therapeutics and biosensing in clinical trials addresses clinical-trial activity; trial participation alone does not establish approval or routine availability (2023 review of clinical trials). To judge readiness, look for the named product, its tested indication, the phase and results of relevant human studies, and its regulatory status in the country where it would be used.

How does the FDA assess drug products containing nanomaterials?

In the United States, FDA’s April 2022 guidance addresses human drug products, including biological products, that contain nanomaterials in the finished dosage form. It discusses product characterization, controls, testing, and qualification, including cases in which nanomaterials are active ingredients or inactive carriers (FDA guidance, April 2022). The guidance is not an approval of nanomedicines as a category.

FDA’s June 2014 guidance discusses dimensional characteristics and size-dependent properties when considering whether a regulated product involves nanotechnology (FDA guidance, June 2014). These documents concern regulatory assessment; they do not establish a universal classification for every microneedle or determine the status of an individual product. A nanomaterial’s presence alone does not establish that a drug is safe, effective, or approved.

How to judge a claim about a new delivery device

  • Identify the route and target: Is the product intended to cross the skin, reach another tissue, or deliver a payload inside cells?
  • Check the physical design and release method: Is it solid, coated, dissolving, hollow, or a different nanostructured system?
  • Ask what medicine it carries: Evidence for one payload or formulation does not automatically apply to another.
  • Look for the evidence stage: Laboratory and animal results, human trials, and an authorized product are different milestones.
  • Verify the specific product and location: Approval or availability depends on the product, indication, and relevant regulator—not on a technology label alone.
