A light-triggered labeling method has identified hexokinase-1 (HK1), a metabolic enzyme, as a candidate partner of G-quadruplex DNA—folded structures formed by guanine-rich sequences. Follow-up tests with purified protein support selective HK1 binding to the human telomere G-quadruplex in vitro. The study does not show what HK1 does at these structures inside living cells.
What the researchers labeled
G-quadruplexes, often shortened to G4, are folded DNA structures that can form in guanine-rich sequences. The team used a human telomere G4 sequence as a target, attaching either a ruthenium-complex or BODIPY photocatalyst to the DNA. After adding the light-activated labeling reagent MAUra, they illuminated the samples so nearby proteins could be tagged, enriched and identified by quantitative proteomics. The study was published in Communications Chemistry on 27 August 2026; the paper describes the method and experiments.

The approach is designed to label proteins near the DNA probe rather than first occupying a G4 binding pocket with a small-molecule ligand. That distinction may help avoid displacing some native interactors, but proximity labeling detects nearness, not necessarily a stable or functional protein–DNA complex.

The proteomic screen produced a broad set of candidate proteins. A Tohoku University-authored summary reported more than 1,000 candidates, a count of proteins flagged by the method rather than 1,000 independently confirmed G4 binders. Each candidate requires further testing; the summary reported by Phys.org also describes the study’s discovery.

HK1 stood out in follow-up tests
HK1 ranked highly in comparisons between the G4 probe and controls. In electrophoretic mobility shift assays with purified protein, it showed binding to G4 DNA but not to the double-stranded DNA tested. Microscale thermophoresis (MST) measured an apparent dissociation constant of 11.8 ± 2.0 nM (n=3) for HK1 binding to Texas Red-labeled human telomere G4 DNA under the paper’s assay conditions. This is a result for that specific probe and experimental setup, not a universal affinity for all G4 sequences or cellular conditions. The measurement and protocol details are in the primary paper.
What remains unproven
The purified-protein assays support a direct HK1–G4 interaction in vitro. They do not establish that the interaction occurs in living cells, explain its biological function, show an effect on gene regulation, or demonstrate a causal connection to cancer or metabolic disease. Those are open questions, not outcomes shown by this study.

Why probe placement and controls matter
The authors report different capture profiles when the photocatalyst was positioned at a G4 terminus versus in a loop. The local structure and labeling conditions therefore influence which nearby proteins are detected. A candidate list should be interpreted in light of the probe design and sample, then narrowed using validation such as purified-protein binding assays and orthogonal tests.

This method also has trade-offs relative to approaches that use a G4-binding ligand: avoiding a ligand that occupies a binding site may reduce competition with some native interactors, while the photocatalyst’s position and the labeling reaction can shape the result. Neither a proximity signal nor enrichment alone proves direct binding; in this case, the separate HK1 assays provide additional support for that particular interaction.

What the blue-light protocol means
In the paper’s purified-protein photolabeling protocol, the researchers used 455 nm LED irradiation for 30 seconds. That is a defined laboratory protocol detail, not evidence that an arbitrary blue light or consumer LED will reproduce the result. The experiment also requires modified oligonucleotides, photocatalyst, MAUra reagent, controlled reaction conditions and proteomic analysis; the illumination time alone is not a usable replication recipe. The paper’s methods give the experimental context.

The work adds a way to profile proteins near a selected folded DNA structure and identifies HK1 as an unexpected candidate for further investigation. Its strongest conclusion is molecular and limited: purified HK1 can bind the tested human telomere G4 under the reported in-vitro conditions. Whether that interaction has a role in cells remains to be determined.
