Co-ASS is an experimental aspirin-derived cobalt–alkyne complex, not ordinary aspirin with cobalt added. Laboratory studies report activity in cancer cell models, but they do not show that Co-ASS prevents or treats cancer in people. It has no established patient-use dose or clinical safety profile in the sources reviewed.
What is the aspirin–cobalt complex?
The name refers to Co-ASS, identified in a 2005 study as [2-acetoxy-(2-propynyl)benzoate]hexacarbonyldicobalt. It is an organometallic compound built from an aspirin-derived component and a cobalt–alkyne unit. It is chemically distinct from acetylsalicylic acid, the active ingredient in ordinary aspirin; it is not made by adding cobalt to an aspirin tablet. The researchers selected Co-ASS as a lead among the compounds they examined. Ott and colleagues’ 2005 study describes the compound and its preclinical investigation.

What did the cell studies find?
Co-ASS and breast cancer cell models
The 2005 study examined Co-ASS-type complexes in MCF-7 and MDA-MB-231 breast cancer cells. The researchers investigated cellular uptake, lipophilicity, DNA binding, nuclear drug content, glutathione reductase inhibition, cyclooxygenase (COX) inhibition, estrogen-receptor binding and apoptosis. The paper reports that its most antitumour-active compounds were potent COX-1 and COX-2 inhibitors. The authors proposed that COX inhibition probably plays a major role in the complexes’ action; this is a mechanistic interpretation of experimental results, not proof of how a drug would work in a person.
The authors called Co-ASS-type cobalt–alkyne complexes a new class of organometallic cytostatics and wrote that COX inhibition “probably plays a major role” in their mode of action. That statement summarizes preclinical work, not a clinical finding. The 2005 article in the Journal of Medicinal Chemistry provides the study context.

Chlorinated derivatives in additional cell lines
A 2018 study assessed chlorinated aspirin-derived cobalt–alkyne complexes, which are related compounds rather than interchangeable versions of Co-ASS. It reported antiproliferative activity in HT-29 and MDA-MB-231 cells, with the following IC50 ranges for the studied compounds:

IC50 is an assay measure of the concentration associated with 50% inhibition of the measured effect. These figures describe the named compounds in specific cellular systems: they are neither clinical concentrations nor doses for patients, and should not be treated as a direct potency comparison with the 2005 study. The 2018 article also reports no effect on growth of the non-tumorigenic HS-5 cell line under its assay conditions. That single cell-line result is not evidence of general safety in people. The Royal Society of Chemistry’s 2018 article reports these findings.

Co-ASS in mesothelioma models
A 2013 study examined Co-ASS in malignant pleural mesothelioma cell lines and reported stronger activity in sarcomatoid than in epithelioid models. Its authors suggested that Co-ASS might be better viewed as a carbon-monoxide-releasing NSAID than as an antitumour drug candidate. This is the authors’ interpretation of cell-model findings, not a settled clinical classification or evidence of patient benefit. The 2013 study abstract gives that model-specific context.
Does aspirin with cobalt treat cancer?
No. These studies concern experimental compounds and laboratory cell assays; they do not establish that taking ordinary aspirin with cobalt is useful, safe or equivalent to Co-ASS. The findings also do not show that Co-ASS treats or prevents cancer in people. The reviewed sources establish no human efficacy, patient-use dose, clinical approval or clinical safety profile for the compound.

Clinical research about ordinary aspirin is separate and cannot be used as evidence for Co-ASS. For example, the National Cancer Institute’s aspirin trials listing concerns aspirin, not the cobalt complex.

- Cell activity is an early-stage finding. Cancer-cell growth effects in laboratory assays do not establish effectiveness in animals or people.
- The compounds and models differ. The 2018 potency ranges apply to its chlorinated derivatives and named cell lines, while the 2005 and 2013 studies examined different experimental questions and models.
- Safety and drug behaviour in people are unresolved. The cited findings do not establish human pharmacokinetics, a safe dose or a clinical safety profile.
Accordingly, “anti-tumour promise” describes a research possibility, not a cancer treatment a reader can use. Co-ASS is a medicinal-chemistry research subject, and the cell studies are not a basis for changing cancer care or self-medicating.
