Are Plastic-Eating Enzymes Safe for People and the Environment?

Not yet for unrestricted use. PET-degrading enzymes such as PETase and MHETase are promising tools for recycling polyethylene terephthalate (PET) in contained reactors, but current studies do not establish that they are safe to put in drinking water, use in the human body, pour into household waste, or release into soil, rivers, oceans, or wildlife habitat. A 2024 laboratory study using human serum found no cell-viability effect under its tested conditions, yet that result is an early proof of concept, not a clinical safety assessment.

What “plastic-eating” enzymes actually do

The phrase is shorthand for enzymes that break particular polymers into smaller molecules. It does not mean one enzyme can digest every plastic. The best-established work concerns polyethylene terephthalate, or PET, the polyester used in many beverage bottles, food containers, textile fibers, and films. Polyethylene (PE), polypropylene (PP), polystyrene (PS), polyvinyl chloride (PVC), and multilayer packaging have different chemical structures and are not covered by the same evidence.

PETase starts the hydrolysis

PETase attacks the ester bonds in PET. Its reaction produces shorter fragments, including mono(2-hydroxyethyl) terephthalate (MHET), along with other soluble intermediates. Performance depends on the enzyme’s sequence, engineering, surface area of the plastic, temperature, pH, mixing, and the presence of dyes, coatings, food residue, or other contaminants.

MHETase converts an intermediate

MHETase can further process MHET toward terephthalic acid (TPA) and ethylene glycol. Recovering those monomers is important because they can potentially become feedstocks for new materials. Stopping at partially degraded fragments would not deliver the same recycling benefit and could complicate waste treatment.

What recent laboratory systems showed

A 2025 ACS Synthetic Biology study displayed engineered PETase and MHETase on yeast cells. It reported an optimum around pH 9 and 30 °C, stability and reuse in the tested system, and conversion of drinking-water bottles into value-added products. A 2024 Journal of Hazardous Materials study examined engineered PET hydrolases and a two-enzyme system over more than 30 days. These are controlled-process results, not proof that the enzymes will behave the same way in a river, landfill, wastewater plant, or human body.

What is known about safety for people?

The human-serum result is limited but relevant

The most directly relevant publication is the 2024 Faraday Discussions paper titled Degradation of PET microplastic particles to monomers in human serum by PETase. Researchers tested PETase against microplastic-like PET substrates at 37 °C in human serum and reported that applying PETase did not influence cell viability in vitro. That finding indicates that, under those experimental conditions, the enzyme did not produce an immediately detectable loss of viability in the cells used for the assay.

Why that does not establish clinical safety

A cell-viability assay is only one endpoint. It does not determine a safe dose, repeated-exposure effects, immune reactions, allergy or sensitization, absorption through the gut or lungs, distribution to organs, metabolism, excretion, or interactions with medicines. It also does not show that PETase reaches plastic particles in a living person, that all reaction products are harmless, or that treatment would provide a medical benefit. No human clinical trial or regulatory approval for treating people with plastic-eating enzymes was identified in the available evidence.

Can PETase be put in drinking water?

There is no basis in these studies for adding PETase, MHETase, engineered yeast, or another plastic-degrading preparation to drinking water. Drinking-water safety requires validated identity and purity, dose limits, stability data, toxicology, control of impurities, and regulatory review. None of those requirements is supplied by a laboratory demonstration of PET breakdown.

Is it safe to touch?

The cited PET studies do not establish skin, eye, inhalation, or occupational exposure limits. A purified enzyme may be a protein preparation, while a commercial or laboratory mixture can also contain buffers, preservatives, residual organisms, or reaction contaminants. Until a product has a specific safety data sheet and exposure assessment, handle it as a laboratory reagent: avoid direct contact and aerosols, use appropriate gloves and eye protection, work with ventilation, and follow the supplier’s instructions. Do not infer that “natural” or biodegradable means non-allergenic.

Could the enzymes harm ecosystems?

Environmental release is an open question rather than a demonstrated benefit. Enzymes optimized in a buffered reactor encounter very different conditions outdoors: changing temperature and pH, sunlight, dilution, salt, suspended sediment, biofilms, competing microbes, and irregular plastic surfaces. A 2024 review of microbial and enzymatic plastic biodegradation identified restricted substrate specificity, poor activity under fluctuating environmental conditions, slow reaction rates, and inadequate study of nanoplastics.

Reaction products and additives need tracking

Breaking PET into TPA and ethylene glycol is not the same as making a contaminated item environmentally harmless. Bottles and textiles can contain dyes, coatings, catalysts, stabilizers, plasticizers, flame retardants, inks, adhesives, and food residues. Enzymes may leave some additives untouched, and incomplete conversion can leave MHET or other oligomers. A responsible process therefore measures the feedstock and the liquid and solid outputs rather than assuming that disappearance of visible plastic equals detoxification.

What happens in seawater, soil, or wastewater?

No cited study establishes reliable rates, persistence, transport, or ecological effects for PETase or MHETase in those settings. Salinity, organic matter, minerals, and microbial communities can reduce activity or change where an enzyme binds. If degradation is slow or incomplete, particles can remain available to organisms while intermediate chemicals accumulate locally. Conversely, rapid conversion in a confined site could produce concentrated dissolved products that still require treatment and recovery.

Engineered microbes raise an extra containment issue

Some systems display enzymes on engineered yeast or use other modified cells. The enzyme’s chemistry and the organism’s biological behavior are separate risk questions. A contained reactor must prevent escape of viable engineered cells, control waste streams, and verify that no organism is released with the treated water or solids. An open-environment application would require ecological, persistence, gene-transfer, and organism-level assessments that are not provided by the PET degradation studies summarized here.

What the microplastic evidence does and does not show

The human-serum experiment is often interpreted as evidence that PETase can safely remove microplastics from people. That conclusion goes beyond the study. It used microplastic-like PET particles in an in-vitro serum system, measured cell viability, and reported no effect under the tested conditions. It did not test a living human, repeated dosing, organs, immune responses, long-term exposure, or every type of microplastic.

Microplastics are chemically diverse. Weathered particles can carry biofilms, metals, and organic pollutants; nanoplastics have a much smaller size range and different surface behavior. The 2024 review specifically noted that nanoplastic biodegradation remains inadequately studied. A result for PET particles therefore cannot be generalized to particles made from other polymers or to environmental mixtures.

Why a successful reactor is not an environmental treatment

Process conditions are part of the result. The engineered-yeast study reported its best performance at pH 9 and 30 °C, while the serum assay used 37 °C. Natural waters and soils rarely hold either temperature or pH constant, and most are not adjusted to the enzyme’s optimum. The 2024 Journal of Hazardous Materials work ran degradation tests for more than 30 days, illustrating that residence time can be substantial.

A 2025 review of PET biocatalysis also described inhibition by the intermediate MHET and unresolved problems in efficiency, separation, and scale-up. In practice, engineers must keep the enzyme active, remove or manage inhibitory products, separate monomers from solids and impurities, and demonstrate that conversion is complete enough for the intended product. Those requirements are easier to enforce in a closed vessel than in a lake or landfill.

What responsible industrial use would require

  1. Characterize the incoming plastic. Identify PET content, multilayer structures, pigments, coatings, labels, metals, food residues, and other additives. Do not assume a clear bottle represents all PET waste.
  2. Define and control operating conditions. Measure pH, temperature, mixing, solids loading, enzyme concentration, and residence time. Compare actual performance with the validated range rather than relying on a nominal optimum.
  3. Contain engineered organisms. If yeast or another modified cell is used, use physical containment, validated inactivation, closed transfers, and monitoring of liquid and solid waste for viable cells.
  4. Measure conversion products. Quantify TPA, ethylene glycol, MHET, residual oligomers, unreacted PET, and relevant additives. Visual disappearance of a bottle fragment is not a mass balance.
  5. Recover useful monomers and treat residuals. Separate and purify TPA and ethylene glycol for the planned application. Send contaminated liquids, solids, and spent biomass through appropriate waste treatment.
  6. Verify performance over time. Test enzyme reuse, activity loss, contamination tolerance, and batch-to-batch variation. The useful question is not whether degradation occurred once, but whether the process remains predictable at scale.
  7. Monitor releases. Establish sampling for air, water, solids, workers, and nearby ecosystems before operation. Stop and investigate if engineered cells, unexpected intermediates, or additives leave the controlled boundary.

How to evaluate a claim that a plastic-eating enzyme is safe

Question What a credible claim should specify What remains unestablished in the current evidence
Which plastic? The exact polymer, additives, particle size, and contamination level. General digestion of plastics beyond PET.
Which catalyst? Enzyme identity, sequence or engineering, purity, and whether living cells are present. A universally safe PETase or MHETase preparation.
Under what conditions? pH, temperature, mixing, solids loading, and reaction time. Predictable activity in unbuffered soil, seawater, rivers, or household drains.
How complete is conversion? Measured TPA, ethylene glycol, MHET, oligomers, residual PET, and additives. That all products from real waste are harmless.
How is exposure assessed? Dose, route, repeated exposure, toxicology, immune effects, and worker controls. Human clinical safety or a safe drinking-water dose.
How is the process contained? Closed equipment, engineered-organism controls, waste treatment, and release monitoring. Ecological safety after open release.
Can the products be recovered? Separation, purification, reuse, and a verified mass balance. Economical, complete recovery at broad commercial scale.

Should you buy a home plastic-digestion kit?

No validated consumer product is established by the cited evidence for safely digesting household plastic. Home users cannot reliably identify polymer blends, maintain the pH and temperature needed by a particular enzyme, separate TPA and ethylene glycol, or test for residual intermediates and additives. Pouring an enzyme mixture into a sink, septic system, compost bin, or outdoor waterway would move an unvalidated process into an uncontrolled environment.

For household waste, use the collection and recycling route specified by your local authority, and keep PET separate when that system requests it. A laboratory or industrial demonstration should not be treated as permission to experiment with drinking containers, food-contact surfaces, drains, or wildlife habitat.

Bottom line for people and the environment

PETase and MHETase are credible research tools for controlled PET depolymerization, not proven all-purpose plastic cleaners. The 2024 human-serum study found no cell-viability effect in vitro at 37 °C, but it did not establish clinical safety. Environmental reviews continue to identify narrow substrate range, slow or condition-sensitive reactions, limited nanoplastic data, and uncertain product and additive fate. The defensible near-term boundary is a contained industrial or research process with characterized feedstock, controlled conditions, engineered-organism containment where relevant, measured products, and recovery of TPA and ethylene glycol. Open-environment release and household “plastic-eating” treatments are not supported by the available safety evidence.
