AFM Tip and Nano-Surfaces: What the Probe Actually Feels

An atomic force microscopy (AFM) tip does not feel a nanosurface like a finger feels texture. Forces between the probe and the sample bend or move a microscopic cantilever, and the instrument measures that response while the scanner changes the tip–sample position. Software converts the calibrated signal into a topographic map or, with an appropriate mode and model, estimates local mechanical, electrical, chemical or magnetic properties.

What the signal means is conditional: probe shape, cantilever calibration, operating mode, sample stiffness and adhesion, environment, scan settings and the analysis model all matter. A bright or dark contrast feature is not automatically a measurement of stiffness, and a force curve cannot be interpreted correctly without checking the contact mechanics behind it.

What an AFM tip actually “feels”

In an AFM, the probe is attached to a flexible cantilever. Interactions such as van der Waals attraction, electrostatic forces, capillary forces, adhesion and short-range repulsion alter the cantilever’s deflection or oscillation. A laser-and-photodetector system usually reads the cantilever motion, while a feedback loop moves the scanner to hold a chosen interaction signal or oscillation condition.

The instrument therefore senses a mechanical response, not a visual image of the surface. As the tip moves laterally, the feedback signal is recorded at each position and reconstructed as an image. In a topography channel, the recorded quantity is related to the vertical movement needed to maintain the setpoint. In a spectroscopy experiment, the tip is deliberately moved toward and away from one location and the response is plotted against displacement.

The resulting force–distance curve can show approach, contact, adhesion and retraction behavior. Its shape reflects the combined properties of the tip, sample and surrounding medium. A curve is evidence of an interaction; it is not a self-explanatory label for “hard” or “soft.”

AFM is capable of sub-nanometer resolution, as described in the 2020 NIST review by Gheorghe Stan and Sean King, but that capability is not a guarantee for every instrument, probe, specimen or operating condition. Vibration, thermal drift, contamination, feedback settings and tip radius can all limit the result. NIST’s review of AFM for nanoscale mechanical-property characterization describes the technique and its limits.

How a force measurement is made

Force mode separates the interaction measurement from a conventional area scan. The tip is positioned over a selected point, then moved vertically relative to the sample. The instrument records cantilever deflection (or an equivalent signal) against scanner displacement, producing a force curve after sensitivity and spring-constant calibration.

  1. Locate and prepare the site. Mount the sample securely, remove loose contamination where that is safe, and choose a region that will not be damaged by the probe.
  2. Calibrate the detector response. Convert photodetector voltage to cantilever deflection using the instrument’s sensitivity procedure.
  3. Calibrate the cantilever spring constant. Use a traceable or instrument-supported method appropriate to the probe; an assumed value can introduce a proportional force error.
  4. Set the approach and retract limits. Define travel, speed, trigger force and dwell time so the tip reaches the intended interaction without unnecessary loading.
  5. Collect approach and retract data. Record deflection versus displacement in both directions. Adhesion often appears as a negative force on retraction, while snap-in or snap-off events can indicate long-range attraction or capillary effects.
  6. Convert and model the curve. Apply the selected contact-mechanics model only after identifying the contact point, baseline, indentation and relevant geometry.

These steps produce a quantitative force curve only when the calibration and model assumptions are defensible. NIST’s discussion of the relationship between AFM force–distance curves and indentation load–penetration curves explains why the two are not automatically interchangeable: the force curve may require additional conversion and assumptions before indentation equations can be used.

What properties can be inferred?

Topography is the most familiar AFM output, but specialized probes and modes extend the measurement beyond shape. Stan and King state that detailed analysis of tip–sample contact mechanics can convert contrast into quantitative nanomechanical properties including elastic modulus, shear modulus, wear rate, adhesion and viscoelasticity. Their wording is important: the conversion depends on a contact-mechanics analysis, not merely on observing contrast in an image.

  • Elastic or Young’s modulus: resistance to reversible deformation, inferred from a model relating force and indentation.
  • Shear modulus: resistance to tangential deformation, often requiring lateral or dynamic measurements rather than a basic vertical force curve.
  • Adhesion: interaction energy or pull-off force estimated from approach/retraction behavior; humidity, contamination and dwell time can change it.
  • Viscoelasticity: time- and rate-dependent response, requiring controlled loading rate, dwell or oscillation and an appropriate model.
  • Wear rate: material loss or probe/sample damage assessed through repeated loading or scanning and a defined wear analysis.
  • Electrical response: current, potential or electrostatic-force contrast measured with conductive or electrically biased probes.
  • Magnetic response: force or resonance changes measured with a magnetically coated probe.
  • Chemical contrast: interaction differences associated with functionalized probes or spectroscopic extensions.

Each property requires its own calibration, probe condition and model. A topographic image alone establishes surface geometry; it does not establish a modulus, conductivity or chemical composition.

Why the force-curve slope can mislead

A common mistake is to treat the slope of every contact segment as a direct measure of sample stiffness. NIST cautions that when the tip does not significantly penetrate the sample, the contact-region slope generally does not contain information about sample stiffness. In that situation, the measured response can be dominated by cantilever compliance, probe geometry, adhesion, substrate effects or an incorrect contact point.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Indentation equations also presume a defined relationship among force, indentation depth and contact area. If the AFM displacement has not been converted to true indentation, or if the tip shape is unknown, inserting a force-curve slope into an indentation formula can produce a precise-looking but unsupported modulus.

For thin films, cells, gels and nanostructures, the substrate or nearby features may influence the response before the tip has sampled enough material. For very stiff specimens, the cantilever may bend while the sample indentation remains negligible. For soft or sticky specimens, snap-in, hysteresis and rate dependence can violate a simple elastic model. Reported values should therefore include the model, fit range, probe geometry, calibration method and environmental conditions.

Probe geometry and calibration are part of the measurement

The tip is not an infinitesimal point. Its radius, cone or pyramid angle, aspect ratio, coating, wear state and contamination determine the contact area and the forces it can apply. A blunt or damaged tip broadens narrow features and changes the apparent mechanical response. A high-aspect-ratio structure can be inaccessible to a low-aspect-ratio probe even when the scanner has sufficient resolution.

NIST describes reference nanostructures and tip-on-tip methods for characterizing probe geometry. These approaches can reveal radius, shape or changes caused by wear, helping distinguish a sample feature from a probe artifact. Probe characterization is especially important when comparing measurements between cantilevers or instruments.

There is no universal AFM force scale or universally suitable probe. A soft biological sample may require a low-stiffness cantilever and low peak force; a hard ceramic may need a stiffer probe and a different contact strategy. Conductive, magnetic or chemically functionalized coatings add capability but also change spring constant, resonance, wear and contamination behavior.

Choose the AFM mode by the question

The mode determines how the tip interacts with the surface and what can be inferred. The following comparison uses the approaches identified by NIST; names and implementation details vary by instrument.

Reader goal Typical approach Interaction style Best suited to Key requirements and cautions
Surface shape Contact or amplitude-modulation (tapping/intermittent-contact) imaging Continuous contact in contact mode; intermittent interaction in tapping Samples where height and morphology are the primary outputs Feedback and tip shape control resolution; tapping reduces sustained lateral force but does not eliminate damage or adhesion.
Spatial mechanical contrast Force-volume or peak-force tapping Repeated force curves or controlled force peaks at each pixel Soft, heterogeneous or delicate samples requiring maps of adhesion or deformation Needs force and deflection calibration, a suitable probe and a contact model; acquisition is slower than a basic height scan.
Mechanical properties at resonance Contact-resonance or intermittent-contact-resonance AFM Continuous or intermittent contact with resonance-frequency and damping changes Surface and, in some configurations, subsurface mechanical-property contrast Requires resonance calibration, contact-stiffness analysis and control of contact conditions.
Pointwise force behavior Force–distance spectroscopy Vertical approach and retraction at selected points Adhesion, deformation, unfolding or rate-dependent response from soft gels through stiff ceramics Interpretation depends on contact point, indentation conversion, geometry, loading rate and environment.
Electrical properties Conductive AFM, electrostatic force microscopy or Kelvin probe force microscopy Electrical current, force or potential contrast, often with bias Conductive pathways, surface potential and electrostatic behavior Needs an appropriate conductive path, electrical calibration and control of stray fields and humidity.
Magnetic properties Magnetic-force extensions Longer-range magnetic interaction with a coated probe Magnetic domains and stray-field distributions Probe magnetization and lift height affect contrast; topography and magnetic signals must be separated.

NIST’s program pages describe these measurement families, including force spectroscopy, amplitude-modulation/tapping, force-volume, peak-force tapping, contact resonance, intermittent-contact resonance, conductive AFM, electrostatic force microscopy and Kelvin probe force microscopy: Atomic-force microscopy and Nanoscale Property Measurements by Atomic Force Microscopy.

Continuous versus intermittent interaction

Contact mode

The tip remains in contact while the scanner moves laterally. This can provide strong feedback on steep features and enables lateral-force measurements, but friction and shear can drag, deform or remove soft material. It is often practical for firmly attached, relatively robust samples.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Tapping or intermittent contact

The cantilever oscillates and touches the surface intermittently. Amplitude, phase and resonance changes provide feedback while reducing the duration of lateral contact. Oscillation does not make the method damage-free: excessive amplitude setpoint, poor tuning or strong adhesion can still deform a sample or contaminate the probe.

Peak-force and force-volume mapping

These modes acquire a force interaction repeatedly across a grid. They can map adhesion, deformation or other fitted parameters alongside height. The map inherits every uncertainty in the force curve, including drift, calibration error, tip shape and model choice. A visually smooth parameter map is not proof that the fitted property is accurate.

Sample, environment and setup choices

Humidity can create a water meniscus that increases adhesion. Liquid operation changes damping, hydrodynamic forces and resonance behavior. Temperature drift moves the apparent surface during long scans. Electrical measurements require grounding and a defined bias path; magnetic measurements require control of lift height and magnetic history.

  • Use a rigid mounting method that prevents sample movement but does not introduce a compliant adhesive layer into the measurement region.
  • Choose scan size and speed so feedback can follow real features without ringing or flattening.
  • Record setpoint, oscillation amplitude, scan rate, environment, probe identification and calibration values with every dataset.
  • Check a reference sample or repeat a line in reverse direction when drift, hysteresis or tip artifacts are suspected.
  • Inspect the tip after measurements that involve high load, sticky materials or repeated wear testing.

Common failure modes and how to diagnose them

Everything looks wider than expected

A finite or contaminated tip can broaden features, especially narrow pillars and trenches. Image a reference nanostructure, clean or replace the probe as appropriate, and compare features with known dimensions. Do not “correct” the width numerically without a defensible tip-shape estimate.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The force curve has no clear contact point

Long-range attraction, drift, vibration, poor deflection sensitivity or a dirty sample can obscure contact. Reduce approach speed, stabilize temperature, improve isolation and verify detector calibration. A contact point chosen by eye should be tested against the baseline and the physical behavior of the sample.

Fitted modulus changes with loading rate

The sample may be viscoelastic, or the changing rate may be altering adhesion and hydrodynamic forces. Measure at controlled rates, report the rate explicitly and use a viscoelastic analysis rather than forcing a purely elastic fit.

Mechanical contrast follows topography

Steep slopes alter contact area and feedback, while a damaged tip can make every region look similar. Compare the property channel with height, reduce force, verify tip geometry and test a flat reference before assigning chemical or mechanical meaning.

Images show streaks, jumps or duplicated edges

These artifacts commonly indicate feedback gains that are too low or high, scan speed that is too fast, loose particles, thermal drift or intermittent tip contact. Adjust one parameter at a time and repeat a small area rather than interpreting the artifact as nanoscale structure.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

A defensible workflow for interpreting “what the tip felt”

  1. State the measured signal. Identify whether the channel is height, deflection, amplitude, phase, resonance, current, potential or another quantity.
  2. Describe the interaction. Say whether the probe was in continuous, intermittent or pointwise contact and give the relevant setpoint or force range.
  3. Document the probe. Record cantilever type, nominal or measured spring constant, resonance, coating, radius estimate and any tip characterization.
  4. Check calibration. Confirm detector sensitivity, spring constant, scanner calibration and electrical or magnetic calibration where applicable.
  5. Select a model that matches the sample. Consider adhesion, substrate, geometry, viscoelasticity, indentation depth and loading rate.
  6. Test robustness. Repeat points, vary the loading rate or setpoint within a safe range, scan in both directions and compare with a reference.
  7. Report uncertainty and scope. Distinguish directly measured channels from inferred properties and state which assumptions limit the result.

This workflow turns the metaphor of “feeling” into a traceable measurement: a calibrated interaction, a defined model and an uncertainty that a reader can evaluate.

Frequently Asked Questions

Does every AFM image measure stiffness?

No. A conventional topography image records surface geometry or the feedback signal needed to follow it. Stiffness requires a suitable mechanical mode, calibrated force data and a contact-mechanics model.

Is an AFM tip always touching the sample?

No. Contact modes maintain continuous contact, while tapping and other intermittent-contact modes touch periodically. Resonance, electrical and magnetic methods can also use controlled separation or lift heights.

Can a force-curve slope be converted directly to Young’s modulus?

Not generally. The conversion requires a valid contact point, indentation estimate, probe geometry, calibration and assumptions about adhesion, substrate and material behavior. If penetration is negligible, the contact slope may contain little or no sample-stiffness information.

What’s actually slowing this PC down?

Pick the symptom - the matching free tool is one click away.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Why does probe shape matter at nanoscale?

The tip has a finite radius and shape, so it sets the contact area and can broaden or distort features. Wear and contamination change both image dimensions and inferred mechanical properties.

Which AFM mode should be used for a soft sample?

There is no universal choice. Intermittent-contact, peak-force or force-volume approaches can limit sustained lateral loading, but the appropriate mode depends on adhesion, stiffness, thickness, environment and the property being measured. Calibration and validation remain necessary.
