A Drug Extended Lifespan in Three Species—but Not in Mice or Humans

An experimental compound that directly activates AMPK extended lifespan in fission yeast, nematode worms and fruit flies, according to a study published in Aging Cell on 29 September 2026. UK Research and Innovation says some results exceeded 25%; that figure does not apply uniformly to every species or experiment. The study did not show longer lives in mice or people.

What the study found

The compound, called 991, directly activates AMP-activated protein kinase (AMPK), an enzyme involved in sensing a cell’s energy state. Researchers reported lifespan extension in three model organisms: fission yeast (Schizosaccharomyces pombe), nematode worms (Caenorhabditis elegans) and fruit flies (Drosophila melanogaster). The primary study was published in Aging Cell on 29 September 2026 (study).

UK Research and Innovation described lifespan extension as “in some cases by over 25%” in its 30 September 2026 summary (UKRI summary). This is a qualified description of selected results, not a shared percentage for all three organisms, doses or experimental conditions. The available headline-level figure does not establish one universal effect size.

Why directly activating AMPK matters

AMPK is often described as an energy sensor because it responds to changes in cellular energy availability. Compound 991 gave researchers a way to activate AMPK directly and test the effects of that target in living organisms.

That is not the same intervention as taking metformin. Metformin acts through a more complex mechanism and can affect other metabolic pathways; the study’s authors distinguish it from direct AMPK activation by 991. The compounds should not be treated as interchangeable longevity treatments.

What supports the AMPK link—and what complicates it

In worms and yeast lacking functional AMPK, the lifespan effect disappeared. That result supports the interpretation that AMPK is necessary for the observed effect in those organisms. It does not by itself show that activating AMPK will extend lifespan in other species.

Dose also mattered: Queen Mary University of London’s 1 October 2026 account reports that higher doses shortened lifespan in some experiments (Queen Mary University summary). The results therefore do not support the assumption that stronger AMPK activation is always better.

What the mouse experiments did—and did not—show

Mice received treatment for three weeks. Researchers observed changes in liver proteins consistent with AMPK activation, including changes associated with ATP production, mitochondrial biogenesis and suppression of the growth-promoting mTOR pathway. These are molecular findings, not evidence that the mice lived longer or experienced improved health.

As Professor David Carling, who leads the Cellular Stress Group at the MRC Laboratory of Medical Sciences, put it in UKRI’s account: “The next step will be to show that we can also improve health and extend lifespan in mice.” Longer-term mammalian studies are needed before making that claim.

Does this mean AMPK activation can extend human lifespan?

No human longevity benefit has been established by this study. Lifespan extension was reported in yeast, worms and flies; the mouse work measured molecular changes after three weeks, not lifespan. Those findings do not demonstrate that 991—or another way of activating AMPK—extends human life.

Nor is 991 an established longevity medicine or a basis for choosing a supplement. Study co-author Dr Charalampos Rallis, a reader at Queen Mary University of London, cautioned: “I would not want anyone to reach for a supplement on the back of this.”

How to interpret longevity claims from this study

  • Check the organism: a result in yeast, worms or flies is not a mouse or human outcome.
  • Check what was measured: lifespan is different from a change in proteins or pathway activity.
  • Check the dose and conditions: the response was not uniformly beneficial at higher doses.
  • Check the intervention: direct activation by 991 is not equivalent to indirect effects from metformin or a supplement.
  • Read percentages narrowly: “over 25%” refers to some reported cases, not every experiment.
