A Colour Is Worth a Thousand Spectra: Screening Photocatalysis with Color

A colour change can help chemists quickly screen photocatalytic reactions—but it is a proxy, not a full chemical analysis. In a 2016 study, Michal Poznik and Burkhard König used indicators to detect common reaction by-products across a 96-well plate of visible-light photocatalytic reactions. In many cases, the indicator response correlated with starting-material conversion and product yield, making it useful for prioritizing reactions for further analysis.

How can a colour change reveal whether a photocatalytic reaction worked?

Different photocatalytic reactions can produce very different products, so a single colour test cannot identify every product directly. Poznik and König’s approach instead detects shared stoichiometric by-products, including reactive oxygen species and protons. After irradiating the reactions, the researchers add a chemical indicator and measure its response spectroscopically.

The logic is indirect: when a reaction produces a measurable by-product in a quantity associated with reaction progress, the indicator can provide evidence that the chemistry occurred. The authors found that by-product quantities correlated with substrate conversion and product yield in many cases—not in every reaction. The result is a semi-quantitative signal for screening, rather than a universal yield measurement or product identification.

What did the 2016 study demonstrate?

The researchers parallel-irradiated and evaluated 96 visible-light photocatalytic reactions in a microtiter plate. They applied the method to known photooxidations using riboflavin tetraacetate (RFTA), reproduced published results, and compared colorimetric results with gas-chromatographic analyses.

They also used the screen to identify two photocatalysts for hydroxylating boronic acids and to find new substrates for generating aryl radicals from aryl halides. Their screened reactions included photochemical dehalogenation of aryl-halide-containing drug molecules; selected indicator results were checked by gas chromatography. The 96-reaction capacity is what this paper demonstrated, not a stated maximum for high-throughput screening generally.

What the indicator can—and cannot—tell you

  • It can help compare candidates. Where the by-product response tracks reaction progress, it can help researchers identify promising conditions or substrates for follow-up.
  • It does not directly identify every product. A colour response to a common by-product is not a compositional profile of the reaction mixture.
  • Its relationship to yield is not universal. The paper reports correlation with conversion and yield in many cases, so an indicator response should not be treated as proof of a particular isolated yield.
  • It is not a substitute for confirmation when detail matters. Product identity and more precise composition analysis require suitable analytical methods.

Colorimetric screening compared with chromatography and mass spectrometry

Approach What the readout measures Practical role Trade-off
Indicator-based colorimetric screen A proxy based on selected shared by-products, such as reactive oxygen species or protons Parallel initial screening and prioritization of reaction candidates Semi-quantitative; does not provide the same product or composition detail as direct analytical methods
Gas chromatography, high-performance liquid chromatography, or mass spectrometry More direct and detailed analysis of reaction components, depending on the method Follow-up analysis when composition, product identity, or stronger confirmation is needed Requires appropriate equipment and analytical capacity

The choice depends on the decision at hand. A proxy readout may be adequate to decide which conditions deserve attention next. A claim about product identity or a need for detailed composition calls for more informative analysis. The paper presents colorimetry as a practical initial-screening option, particularly for laboratories that may not have high-throughput analytical instrumentation readily available—not as a way to eliminate chromatography or mass spectrometry.

Why the title is a metaphor

“A colour is worth a thousand spectra” captures the appeal of a compact screening signal: one indicator response can help sift through many reactions. It should not be read literally as a claim that colour replaces spectra in all analytical contexts. In this study, the indicator response was itself measured spectroscopically, and the method traded some analytical detail for parallel screening and accessibility.
