A closer look at hard-to-see cells could answer new questions about nerve damage and recovery

Researchers at Texas A&M University have developed a workflow for imaging mitochondria inside terminal Schwann cells without first removing the cells from their muscle environment. Published October 12, 2026, the method reveals differences between healthy and dystrophic tissue, giving scientists a way to study how nerve–muscle connections respond to injury. It is a laboratory tool, not a demonstrated treatment for people with nerve damage.

What the Texas A&M team developed

The method was reported by Texas A&M University and published in the Journal of Visualized Experiments as “Methods for In situ Quantification of Mitochondrial Morphology in Muscle and Terminal Schwann Cells of Mice” (DOI 10.3791/69732). Its central aim is to show the mitochondria inside terminal Schwann cells while those cells remain in their native muscle setting.

That distinction matters because terminal Schwann cells can change rapidly when they are removed from tissue. A preparation that distorts the cell can make an internal structure look different from the way it looked in the body. The workflow labels mitochondria inside the Schwann cells before the fluorescent dye spreads extensively through the surrounding muscle, then prepares the tissue quickly to limit shape changes.

What are terminal Schwann cells?

They sit at the nerve–muscle junction

Terminal Schwann cells are specialized support cells positioned at neuromuscular junctions, where motor nerves communicate with muscle fibers. In healthy tissue they have a distinctive branched, pretzel-like shape that wraps around the junction. Their position allows them to help maintain the connection as well as respond when it is disrupted.

They become active after injury

Following nerve damage, these cells can change shape, move into injured tissue, clear cellular debris, release growth factors and form a pathway that helps regenerating axons find the muscle again. Instead of remaining in their usual stationary configuration, they can become more spherical and mobile. Those changes are useful for repair, but they also make the cells difficult to examine in an undisturbed state.

Why mitochondria are important to the question

Mitochondria supply cells with usable energy and can be arranged as connected networks rather than isolated units. In a terminal Schwann cell, the organization of that network may reflect how the cell is maintaining a neuromuscular junction or responding to stress. To test that idea, scientists need to see both the cell’s shape and its mitochondrial architecture together.

Muscle creates a technical obstacle: it contains many mitochondria of its own. A dye applied broadly can label the muscle as well as the much smaller Schwann cells, making it hard to assign a fluorescent signal to the correct cell. Removing the Schwann cells introduces a second obstacle by allowing their natural structure to change before imaging.

What the new imaging workflow showed

Tissue condition Mitochondrial pattern observed Interpretation supported by the study
Healthy terminal Schwann cells Interconnected mitochondrial networks, including small doughnut-shaped loops near branch points A relatively organized network can be visualized while the cells retain their native structure
Reported dystrophic model More fragmented networks with the loop configurations no longer evident Mitochondrial architecture differs between healthy and dystrophic tissue

The contrast demonstrates that the method can capture a structural difference rather than merely producing a general mitochondrial stain. Aaron Morton, an assistant professor at Texas A&M, described the challenge this way: “If you want to take a look at the things inside of them, as they are in the body, then somehow you have to image them without them knowing anything is disrupted.”

What the findings do—and do not—establish

The result is evidence that mitochondrial morphology can be measured in terminal Schwann cells in place and compared across tissue conditions. It does not establish that a particular mitochondrial pattern causes nerve failure, nor does it show that changing the pattern restores movement or sensation.

  • The report provides no numerical accuracy measure for the imaging workflow.
  • No patients were studied, and no human recovery outcome was reported.
  • The current work did not investigate amyotrophic lateral sclerosis, diabetic neuropathy or spinal muscular atrophy.
  • Those disorders remain possible future applications because earlier studies have implicated terminal Schwann cells in them, not because this method has already been validated in those diseases.

Morton compared the visual difference between conditions to cells that either cooperate or separate: “When everyone’s happy, they hold hands and exchange contents. When they’re mad, they all break apart and keep to themselves.” The analogy describes the observed network pattern; it is not a clinical diagnostic rule.

How this could change nerve-injury studies

A native-state view lets investigators ask whether mitochondrial fragmentation appears early or late after an injury, whether it accompanies the Schwann cell’s shift from a pretzel-like form to a mobile form, and whether recovery reverses the change. It also gives experiments a way to distinguish a Schwann-cell signal from the much larger mitochondrial background in muscle.

Morton’s laboratory plans to use the workflow in nerve-injury experiments and to test whether an experimental biomaterial intended to promote mitochondrial health improves nerve recovery. That work is planned; the biomaterial is not an established treatment, and the imaging method itself has not demonstrated a benefit for patients.

Why the method matters now

Many questions about nerve repair are questions of timing and location: which cells change first, where their energy systems fail, and whether a damaged neuromuscular junction can be rebuilt. A method that preserves the junction and exposes internal mitochondrial organization can make those questions experimentally approachable. As Morton put it, “When a tool is created or when a new method is made, oftentimes it opens the door to new investigations, opening the door to new answers.”

For now, the most defensible conclusion is narrower: terminal Schwann-cell mitochondria look organized and interconnected in healthy tissue but more fragmented in the reported dystrophic model, and the new workflow makes that comparison possible without relying on a fully isolated cell. Establishing what those patterns mean for specific injuries, diseases or therapies will require follow-up experiments.
